Spectrophotometric Assay for the Quantification of Plasma Ethanol Levels in Mice through Chromium-Ethanol Oxidation-Reduction Reaction

The quantification of blood/plasma ethanol concentration (BEC/PEC) is of great importance in experiments involving basic research, clinical studies, and bioethanol production. Traditional methods commonly used to measure BEC can be expensive and require high-cost equipment and qualified labor. The aim of this study was to develop a low-cost method that can be performed with simple infrastructure commonly available in research laboratories. For this, we developed a protocol to quantify PEC in mice, using the method of reduction of potassium dichromate by ethanol. However, this oxidation-reduction (redox) reaction is not specific to ethanol. Thus, the PEC was measured following a sequence of chemical reactions to eliminate the reductive interfering substances presented in the samples. Firstly, we evaluated the sensitivity of the dichromate reactive to ethanol and to different reducing substances found in the plasma, in order to determine which the main interfering substances are. Next, once the main interfering substances were determined in the dichromate reduction, plasma was assayed for PEC. First, mice received intraperitoneally (i.p.) saline (basal reading, 0% ethanol) or ethanol injections (0.5, 1, 2, 3, and 4 g/kg) and had their plasma collected. After plasma deproteinization and plasma glucose oxidation, it was mixed with the dichromate/acetic acid reactive, and then the products of the redox reaction were determined by the spectrophotometric method. Then, we determined the PEC with the same plasma samples using a commercial ethanol assay kit as a positive control. We found an excellent correlation between the administered ethanol doses and PECs in both the methods analyzed. The values of PEC found in the dichromate reaction method were similar to those obtained in the literature with the same ethanol doses, and to the commercial enzyme activity assay. Therefore, despite the need for a background reading, this method can be successfully applied to determine PEC

Sprouts seasonal elongation of two olive cultivars in a high-density orchard

The new high-density cropping systems (>1200 trees ha-1) represent a very interesting proposal for olive orchard profitability. It is crucial to know the morphology and the dynamics of sprout elongation of a cultivar in order to fully assess its suitability for a high-density olive orchard. For this reason we planned a research on two cultivars, Coratina and Arbequina, in a high-density orchard. The apical sprouts elongation of Arbequina early stopped at fruit set without a further step, while Coratina showed a little growth flux after pit hardening. Similar trends showed the lateral proleptic sprouts. Only the sylleptic sprouts of both cultivars had a second period of activity. In all cases, the sprouts elongation finished at the end of summer, when oil accumulation started. Coratina showed higher apical shoot growth and internodes mean length than Arbequina. On the contrary, Coratina showed lower lateral proleptic shoot growth and nodes number than Arbequina, but the same internodes mean length. No significant differences were observed between cultivars for growth, nodes number and internodes mean length of sylleptic shoots. The differences observed between the two cultivars could be explained considering their different vigour. The introduction of this innovative cropping system is allowed to register a considerable reduction of production costs. The result is a considerable increase in the economic performance of the olive grove and a consequent reduction in the unit cost for kg of oil. These data are very useful for varietal choice and field management in high-density orchards and then for new olive breeding programs.

Building construction using cement bricks on the one hand,and mud bricks on the other hand,presents problems of resistance and durability in the face of natural weathering in our country(Republic of Guinea).These difficulties could be linked to non-compliance with standards and the failure to master certain physico-chemical,mechanical and technical parameters when constructing a *** order to remedy this problem,a series of studies were carried out on samples of mud bricks used in the construction of century-old straw hut buildings in the urban commune of Kouroussa(Republic of Guinea).To this end,the compressive strength of the brick samples was assessed either:0.10 MPa,and sensory techniques of certain characteristics were carried out to evaluate the cohesion of the elements in the brick samples,such as:coward boule technique,hand washing test,withdrawal of a soil(pellet or ring test)and of the jar *** physico-chemical analyses methods were carried out using XRF(X-Ray Fluorescence Spectrometry),granulometry and Atterberg limits in accordance with NF P *** results showed a high insoluble residue content(49.9%)and the sample had a clay-like appearance with a low PI(Plasticity Index)=8%.This study could be an alternative guide for the authorities in charge of housing and construction in the Kouroussa prefecture(Republic of Guinea).because,the majority of houses in Upper Guinea are built with mud raw bricks or fired bricks.

Effect of Wholemeal Durum Wheat Varieties on Bread Quality

In this work the effect of six varieties of durum wheat semolina on the bread physico-chemical and sensorial properties was addressed. In particular, whole grains of durum wheat (Anco Marzio, Claudio, Core, Iride, Saragolla and Cappelli) were finely milled by using an ancient stone milling system. Texture?analysis was carried out on both dough and bread samples to evaluate their firmness. Furthermore, tomographic analysis was performed on the bread samples in order to provide a more detailed view of their texture. The Glucose Equivalent, the chemical and the sensory analyses of the bread were also determined. Results highlighted that the lowest Glucose Equivalent valuewas achieved in bread produced with the Anco Marzio cultivar, which appeared instead the worst in terms of texture. Among the investigated samples, bread from the Cappelli variety showed good structural characteristic, a moderate Glucose Equivalent compared to the reference sample (CTRL) and the highest sensory quality.
