Constitutive expression of codon optimized Trichoderma reesei TrCel5A in Pichia pastoris using GAP promoter

To address the deficient activity of TrCel5A in naturally secreted cellulase preparation,this study used the GAP promoter to induce constitutive expression of Trichoderma reesei TrCel5A in Pichia pastoris.A recombinant TrCel5A was screened out after gene optimization,synthesis,and *** biochemical and enzymatic properties of the new recombinant were *** a result,optimization of shake-flask fermentation of the recombinant was obtained at 28℃,2%inoculum volume,an initial pH of 6.0,as well as glycerol and Tween-80 additions of 30 g/L and 6 g/L,*** the above-optimized conditions,the recombinant produced 14.8 U/mL of the enzyme activity at 96 h of *** further enhance enzyme production,pilot-scale cultivation was evaluated using 5-L *** high-cell-density fermentation,the recombinant strain increased enzyme activity to 130.4 U/ml and protein content to 2.49 g/*** addition,the kinetic factors,including K_(m) and V_(max) values for TrCel5A,were detected to be 5.1 mg/mL and 265.9μmol/(***),***,TrCel5A was effectively expressed in *** under the GAP promoter,and it demonstrated its potential in commercially relevant enzyme hydrolysis of lignocellulosic biomass.

Ensuring food safety is paramount *** effective detection methods to ensure food safety can be challenging owing to trace hazards,long detection time,and resource-poor sites,in addition to the matrix effects of *** glucose meter(PGM),a classic point-of-care testing device,possesses unique application advantages,demonstrating promise in food ***,many studies have used PGM-based biosensors and signal amplification technologies to achieve sensitive and specific detection of food *** amplification technologies have the potential to greatly improve the analytical performance and integration of PGMs with biosensors,which is crucial for solving the challenges associated with the use of PGMs for food safety *** review introduces the basic detection principle of a PGM-based sensing strategy,which consists of three key factors:target recognition,signal transduction,and signal *** studies of existing PGM-based sensing strategies combined with various signal amplification technologies(nanomaterial-loaded multienzyme labeling,nucleic acid reaction,DNAzyme catalysis,responsive nanomaterial encapsulation,and others)in the field of food safety detection are *** perspectives and potential opportunities and challenges associated with PGMs in the field of food safety are *** the need for complex sample preparation and the lack of standardization in the field,using PGMs in combination with signal amplification technology shows promise as a rapid and cost-effective method for food safety hazard analysis.

This present study includes twelve species that represent the Ficus genus,namely;aspera,carica,tinctoria ***,hirta,hispida,neriifolia,palmata,pumila,racemosa,septica,sur,and sycomorus,belonging to the Moraceae *** species samples were collected from various locations in *** study focused on the anatomical and molecular characteristics of mature foliage *** the identification and classification of taxa are highly dependent on the anatomical features of leaves,the anatomical characteristics were recorded in the form of a comparison between the examined plants in the data *** study aims to contribute to the identification of the studied species based on the anatomical details of the matured *** characterization includes the variations in upper and lower epidermal layers that are covered by a thin or thick cuticle;the number of palisade and spongy layers;crystals;secretory elements;lithocysts;the midrib zone has parenchyma associated with mechanical tissue,vascular system,and investigation of trichomes;on the other hand,in the current study,the phylogenetic analysis was conducted by using the ITS and 5.8 S *** the analysis of all the available data,it could be stated that there is an overall agreement with the anatomical character dendrogram.

Patient privacy and data protection have been crucial concerns in Ehealthcare systems for many *** modern-day applications,patient data usually holds clinical imagery,records,and other medical ***,the Internet of Medical Things(IoMT),equipped with cloud computing,has come out to be a beneficial paradigm in the healthcare ***,the openness of networks and systems leads to security threats and illegal ***,reliable,fast,and robust security methods need to be developed to ensure the safe exchange of healthcare data generated from various image sensing and other IoMT-driven devices in the IoMT *** paper presents an image protection scheme for healthcare applications to protect patients’medical image data exchanged in IoMT *** proposed security scheme depends on an enhanced 2D discrete chaotic map and allows dynamic substitution based on an optimized highly-nonlinear S-box and diffusion to gain an excellent security *** optimized S-box has an excellent nonlinearity score of *** new image protection scheme is efficient enough to exhibit correlation values less than 0.0022,entropy values higher than 7.999,and NPCR values around 99.6%.To reveal the efficacy of the scheme,several comparison studies are *** comparison studies reveal that the novel protection scheme is robust,efficient,and capable of securing healthcare imagery in IoMT systems.

Anisotropy of Magnetic Susceptibility (AMS) of Bostonite Rocks, Um-Gir and el-Atshan-II Uranium Prospect Areas, Central Eastern Desert, Egypt

Um-Gir and el-Atshan-II areas represent important structurally-controlled radioactive anomalies and have become promising areas for U exploration. The bostonite rocks represent the most promising rock unit in the two studied areas. They form the most promising rocks due to their high radioactivity and, hence uranium mineralization. Anisotropy of magnetic susceptibility (AMS) measurements was carried out for Um-Gir and el-Atshan-II bostonite rocks. The AMS results show that the mean magnitude of magnetic susceptibilities is low, of the order of 10-5 S.I. units. Consequently, the bostonite rocks of the two areas are considered as non-magnetic, reflecting the presence of hematite as the main ferromagnetic mineral. These results are in agreement with the measurements of initial remnant magnetization (NRM) and the results of isothermal remnant magnetization (IRM) for the two areas. Besides, the results of magnetic anisotropy degree (PJ), refer to and concluded that the magnetic fabric of bostonite of two the areas has secondary fabric. AMS results clearly show the differences between the bostonite dyke of Um-Gir and Bostonite sill of el Atshan-II. The lineation (P1) > foliation (P3), and the negative values of the ellipsoid shape (T) of Um-Gir bostonite reflect clearly magnetic lineation (prolate ellipsoid). However, in el Atshan-II, (P3 > P1), the values of T are positive, which indicate the predominance of magnetic foliation (oblate ellipsoid) over lineation. Furthermore, the foliation planes of the bostonite dyke of Um-Gir possess a shallow to moderate inclination and a nearly N-S direction, while, the foliation planes of the bostonite sill of el-Atshan-II show a shallow to moderate inclination and a NNE direction. Role of zinc as an antioxidant and anti-inflammatory to relieve cadmium oxidative stress induced testicular damage in rats

Objective:To investigate the role of zinc in reducing the deleterious effects of cadmium on male ***:Rats were injected subcutaneously with CdCl_2 and ZnCl_2 at dose level of2.2 mg/kg(1/40 of LD50 of cadmium per day).Results:The rats treated with cadmium exhibited a significant increase in levels of testicular malondialdehyde,tumor necrosis factor-alpha,nitrogen oxide and inducible nitrogen oxide synthase immunostaining reaction,as well as an elevation of blood hydroperoxide and follicle stimulating *** addition,a significant decrease in testicular ascorbic acid,zinc,reduced glutathione,catalase,superoxide dismutase,sex organ weight,plasma testosterone and luteinizing hormone were observed in the cadmium *** motility and count were decreased with cadmium treatment,while sperm abnormalities elevated *** treatment was found to mitigate the toxic effects of cadmium on oxidative stress,spermatogenesis,sex hormones,and inflammatory *** injected with cadmium showed intense histopathological *** manifested protective role and markedly reduced tissues damage induced by ***:The protective effect of zinc can be attributed to its antioxidant and antiinflammatory properties.

Radionuclides measurements and mineralogical studies on beach sands, East Rosetta Estuary, Egypt

Radiometric measurements were carried out for the beach sands from East Rosetta estuary to determine the activity concentrations of 238 U, 226 Ra, 232 Th, and 40 K, using a Hyper Pure Germanium spectrometer, to estimate the dose rates and radiation hazard indices. The average specific activities are 778.20 Bq/kg for 238 U; 646.89 Bq/kg for 226 Ra; 621.92 and 627.85 Bq/kg for the 222 Rn daughters 214 Pb and 214 Bi respectively. The average specific activity of 232 Th is 1510.25 Bq/kg, while the calculated specific activity for 40 K has an average of 8.41 Bq/kg. The average specific activity of 235 U is 38.61 Bq/kg. The average absorbed dose rate is 1211.36 nGy/h, 20 times higher than the estimated average global primordial radiation of 60 nGy/h and 6 times higher than that of the world range (10-200 nGy/h). The radium equivalent (Ra eq ) values are from 6 to 9 times the recommended value. The internal and external hazard indices (H int , H ex ) indicate that their values are from 6 to 11 times the permissible values of these indices. These higher values may be due to the presence of economic heavy minerals containing radionuclides as zircon and monazite as well as some trace minerals, thorite and uranothorite. The mineralogical study indicates the beach sands contain heavy minerals, zircon, monazite, rutile, ilmenite, leucoxene, magnetite and garnet. The average abundance of zircon is 0.175 wt% ranging from 0.125 wt% to 0.239 wt%, while it is 0.004wt% ranging from 0.001 wt% to 0.007 wt% for monazite. The average abundance is 0.087 wt% for rutile; 2.029 wt% for ilmenite; 1.084 wt% for magnetite; 0.384 wt% for leucoxene and 0.295 wt% for garnet.

Comparison of bacterial community structures in two systems of a sewage treatment plant using PCR-DGGE analysis

The combination of PCR amplification of 16S rRNA genes with denaturing gradient gel electrophoresis （DGGE） analysis was used to reveal the compositions and dynamics of bacterial communities in a sewage treatment plant with two systems, i.e., an anoxic- anaerobic-aerobic system （inverted A2O） and an anaerobic-anoxic-aerobic one （conventional A2O） over a period from February to July 2009, during which both systems experienced serious sludge bulking problems. The DGGE patterns showed that there were many common bands in both systems, suggesting the high similarity of bacterial communities of the two systems. Meanwhile, the moving window correlation analysis showed that the two systems experienced different microbial community structure changes during the period, which might be related with the different situations of the occurrence and disappearance of sludge bulking, as being reflected by sludge volume index （SVI） values. Major bands of DGGE patterns of sludge samples were further sequenced. Phylogenetic affiliation indicated that the majority of the sequences obtained were affiliated with Actinobacteria, Firmicutes, Bacteroidetes/Chlorobi group and α- and β-Proteobacteria. Two sequences showed high similarities to typical filamentous bacteria Microthrix parvicella and Nostocoida limicola I, indicating that these bacterial species have been involved in the sludge bulking problems.

Today,due to the pandemic of COVID-19 the entire world is facing a serious health *** to the World Health Organization(WHO),people in public places should wear a face mask to control the rapid transmission of *** governmental bodies of different countries imposed that wearing a face mask is compulsory in public ***,it is very difficult to manually monitor people in overcrowded *** research focuses on providing a solution to enforce one of the important preventative measures of COVID-19 in public places,by presenting an automated system that automatically localizes masked and unmasked human faces within an image or video of an area which assist in this outbreak of *** paper demonstrates a transfer learning approach with the Faster-RCNN model to detect faces that are masked or *** proposed framework is built by fine-tuning the state-of-the-art deep learning model,Faster-RCNN,and has been validated on a publicly available dataset named Face Mask Dataset(FMD)and achieving the highest average precision(AP)of 81%and highest average Recall(AR)of 84%.This shows the strong robustness and capabilities of the Faster-RCNN model to detect individuals with masked and un-masked ***,this work applies to real-time and can be implemented in any public service area.
