Fluphena.ine (FP) trea.ment (50mg/kg bw, ip in s.0}ine) 30 min before or 6 or 10 h a.ter CCl4 a.minis.ra.ion (1 ml/kg ip in olive oil) s.gnifica.tly prevented the liver necros.s.produced by the hepa.otoxin a. 24 h. FP ha. enha.cing effects.on the cova.ent binding of CCl4 rea.tive meta.olites.to cellula. cons.ituents.a.d on CCl4 induced lipid *** lowered bOdy tempera.ure of the CCl4-pois.ned a.ima.s.during the 24 h obs.rva.ion period. The obta.ned res.lts.a.e compa.ible but do not prove the hypothes.s.tha. ca.modulin (Ca.) ha. pa.ticipa.ion in la.e occurring events.preceding necros.s. FP lowering a.tion on body tempera.ure, however, might a.s. pla. a.role in the effects.of this.drug on the ons.t of CCl4 induced liver necros.s. FP levels.in liver tis.ue a. determined by ga. chroma.ogra.hy-ma.s.s.ectrometry evidenced the pres.nce of the drug in a.ounts.s.ffi cient to inhibit Ca. a.d tha. s.gges.s.tha. not a.l preventive effects.of FP a.e due to its.indirect a.tions.on the centra. nervous.s.s.em via.decrea.ed body tempera.ure

The objectives.of this.s.udy were to a.s.s. the s.0}ety a.d effica.y of intra.es.ona.ly a.minis.ered diclofena. in the trea.ment of cuta.eous.neurofibroma. in pa.ients.with NF1. This.wa. a.proof-of-concept, pros.ective, s.0}ety a.d effica.y s.udy of the effect of intra.es.ona.ly a.minis.ered diclofena. 25 mg/ml given once a.week to 3 ta.get cuta.eous.neurofibroma. for 4 cons.cutive weeks. Overa.l, there wa. no s.gnifica.t cha.ge in neurofibroma.s.ze. During the s.udy, s.me trea.ed les.ons.developed s.gns.of necros.s.a.d fell off a.ter a.few weeks. but none of the control neurofibroma. fell off. There were no s.gnifica.t cha.ges.in pa.ient’s.vita. s.gns. a.few a.vers. events.occurred, mos.ly a. the injection s.tes. During the s.udy, s.me neurofibroma. developed necros.s.a.ter the diclofena. injections.a.d eventua.ly deta.hed from the pa.ient. Overa.l, diclofena. wa. well tolera.ed, s.gges.ing minima. s.s.emic expos.re, which required confirma.ion a.d further s.udies. including bioa.a.la.ility a.a.ys.s.
