Bile a.ids a.e imp.ica.ed a. etiologic a.ents in ca.cer of the ga.trointestina. (GI) tra.t, including ca.cer of the esop.a.us, stoma.h, sma.l intestine, liver, bilia.y tra.t, p.0}crea. a.d colon/rectum. Deleterious effects of bile a.id exp.sure, likely rela.ed to ca.cinogenesis, include: induction of rea.tive oxygen a.d rea.tive nitrogen sp.cies; induction of DNa.da.a.e; stimula.ion of muta.ion; induction of a.op.osis in the short term, a.d selection for a.op.osis resista.ce in the long term. These deleterious effects ha.e, so fa., been rep.rted most consistently in rela.ion to esop.a.ea. a.d colorecta. ca.cer, but a.so to some extent in rela.ion to ca.cer of other orga.s. In a.dition, evidence is reviewed for a. a.socia.ion of increa.ed bile a.id exp.sure with ca.cer risk in huma. p.p.la.ions, in sp.cific huma. genetic conditions, a.d in a.ima. exp.riments. a.model for the role of bile a.ids in GI ca.cinogenesis is p.esented from a.Da.winia. p.rsp.ctive tha. offers a. exp.a.a.ion for how the observed effects of bile a.ids on cells contribute to ca.cer develop.ent.
