o.JECTIVE： Ligusticum po.ted is a.tra.itio.a. Na.ive a.erica. herb. The ro.ts o. L. po.ted a.e tra.itio.a.ly used in the trea.ment o. ma.y disea.es, ho.ever, its cyto.o.icity, a.tio.ida.ive a.d immune- mo.ula.o.y effects need to.be investiga.ed. In this study, we eva.ua.ed the effects o. the ro.t extra.t a. different do.es o. huma. periphera. blo.d lympho.ytes （PBLs）. METHo.S： The lympho.ytes were incuba.ed with different co.centra.io.s o. the ro.t extra.ts （0, 50, 100, 200, a.d 400 μg/mL） a.d ha.vested every 6 h fo. 2 d （P〈0.05）. The pro.ective effect o. the herb a.a.nst o.ida.ive da.a.e wa. determined by inducing o.ida.ive stress with the a.ministra.io. o. 50 μmo./L o. hydro.en pero.ide （H202）. RESULTS： Trea.ments with L. po.ted a. 200 a.d 400 pg/mL increa.ed the via.ility o. PBLs. The deleterio.s effect o. H2o. wa. a.elio.a.ed by 400μg/mL L. po.ted trea.ment. a.ditio. o. 400 μg/mL L. po.ted reduced lipid pero.ida.io. in stressed PBLs by 94% （P〈0.05）. Trea.ment with 400 μg/mL o. L. po.ted resulted in a.26.4% increa.e o. reduced gluta.hio.e levels. a.tivities o. supero.ide dismuta.e a.d ca.a.a.e increa.ed by 17.5% a.d 55.2% respectively, when stressed PBLs were trea.ed with 400 μg/mL L. po.ted fo. 2 d （P〈0.05）. Trea.ment with 400 μg/mL L. po.ted increa.ed interfero.-γa.d interleukin-2 expressio.s in H2o.-cha.lenged PBLs （P〈0.05）, ho.ever, the ro.t extra.t did no. ca.se a.significa.t difference in interleukin-10 levels co.pa.ed to.the co.tro. （P〉0.05）.
