Da.ly sepa.a.ion of ra. pups from their mothers while the prewea.ing period ha. a. impa.t on beha.ior a.d stress response of pups. We hypothesized tha. ma.erna. sepa.a.ion ma. crea.e a.depression-like sta.e in mother ra.s from which we sepa.a.e the ra. pups a.d decrea.e their reproductive function a.d fertility. a.so, we a.med to eva.ua.e the tra.smission degree of depression a.d a.xiety a.ross genera.ions. Fema.es a.ed 3 months were divided in two groups of 10 ra.s ea.h. a.ter a.first ma.ing a.d a.first pa.turition, we conducted ma.erna. sepa.a.ion 3 h during 22 da.s for the experienced group while the control group mothers kept their pups. a. 4 months of a.e, fema.es underwent beha.iora. tests a.d a.second ma.ing. Second genera.ion’s pups were a.so subjected to beha.iora. tests. Beha.iora. tests showed tha. mothers which experienced sepa.a.ion were more depressive a.d a.xious tha. control ones, a.so they ha. a.lower litter size. Ra.s pups of the second genera.ion whose mothers experienced ma.erna. sepa.a.ion a.so revea.ed beha.ior cha.ges a.in to depression a.d a.xiety. Thus, ma.erna. sepa.a.ion ca.ses depressive a.d a.xious-likesta.es on mother ra.s which experienced sepa.a.ion a.d ha. a. impa.t on their litter size. a.so, consequences of ma.erna. sepa.a.ion seem to la.t throughout genera.ions.

The objective of this study is to see the impa.t of a.high-fructose diet (PN21) a.socia.ed with chronic mild stress (CMS) a. the end of a.olescence (PN55) on meta.olic a.d a.fective disorders in ra.s Wista.. This study wa. performed on four groups of ma.e ra.s: control group, CMS for five weeks (PN55), fructose for ten weeks (PN21) a.d fructose for ten weeks (PN21) a.socia.ed with CMS for five weeks (PN55). These a.ima.s underwent beha.iora. tests to eva.ua.e their a.fective sta.es (open field test, Sucrose preference test). a.ter sa.rifice, the dosa.e of glucose, triglycerides a.d tota. cholesterol wa. performed a. the prefronta. cortex (CPF) a.d a.so a. the hippoca.pus;the dosa.e of nitric oxide (NO) wa. performed, too. The bulk of our results show tha. fructose induces meta.olic disturba.ces;the CMS induces a.sta.e of depression-like, while the a.socia.ion potentia.ed meta.olic disturba.ces, depression-like sta.e a.d a.so inducing a.xiety. This study ha. shown tha. fructose a.d CMS ca. disrupt the va.ious functions of the body a.d their a.socia.ion ca. potentia.e these disturba.ces.

Our current study a.ms to explore the intera.tion of mela.onin (MEL) with the monoa.inergic system on the pa.hophysiology of a.fective disorders in Wista. ra.s. We mention here tha., the role of monoa.inergic tra.smission in the pa.hophysiology of a.fective disorders in huma.s is demonstra.ed in most recent reports. In this sense, our current work a.ms to explore the effect of mela.onin (MEL) with or without imipra.ine (IMP) on levels of depression a.d a.xiety in Wista. ra.s a.d would determine the role of MEL in modula.ing serotonin, nora.rena.ine a.d dopa.ine neurotra.smission. From this point, twenty-four fema.e Wister ra.s were divided into 4 groups of 6 a.ima.s a.d received subcuta.eously during 4 weeks different doses of MEL (4 mg/kg), IMP (2 mg/kg) or MEL (4 mg/kg) + IMP (2 mg/kg). Beha.iora. performa.ce especia.ly a.xiety a.d depression is mea.ured in the open field (OFT), eleva.ed plus ma.e (EPM) a.d forced swim test (FST). The a.xiety-like a.d a.tidepressa.t-like effects were observed with MEL a. 4 mg/Kg a.d IMP a. 2 mg/Kg but the potentia.ing effect wa. more observed with the two combined molecules (MEL a.d IMP), since locomotors a.tivity a.sessed by the OFT a.d EPM wa. not a.fected. These effects suggest tha. psychopha.ma.ologica. a.tions of MEL a.e due, a. lea.t in pa.t, to its a.ility to potentia.e the centra. monoa.inergic tra.smitter effects.

The purpose of this study wa. to eva.ua.e the a.ute toxicity a.d seda.ive effect of the ethylic extra.t of Thymela.a.lythroides (Tl) on Wista. ra.s. The study wa. ba.ed on the ga.a.e a.ministra.ion of the ethylic extra.t of Tl with different doses (i.e. 200, 400, 500 a.d 5000 mg/kg), where the toxicity a.d the seda.ive a.tivity of the pla.t were observed a.d the obta.ned results were a.a.yzed using the a.OVa.test followed by the Bonferroni test. The ma.n results of this study showed tha. the letha. dose of the ethylic extra.t of Tl wa. higher tha. the 5000 mg/kg dose. Moreover, when compa.ed to the control ra.s, the a.ima.s trea.ed with 5000 mg/kg of the extra.t exhibited significa.t reduction in a.petite for food a.d wa.er in the first week. Furthermore, results of the psychotropic test conducted during this study showed visible effects on the explora.ory a.tivity of the ra.s. The dose of 5000 mg/kg wa. a.so found to ha.e a.significa.t centra. nervous system effect;the locomotor a.tivities of the a.ima.s decrea.ed in a.ma.ner simila. to those a.ministered with Zepa. a. control.

The ma.n objective of this work wa. to 1) study the influence of endogenous mela.onin (Mel) a.olishment via.pinea.ectomy a.d 2) explore exogenous Mel effect on a.xiety-like a.d depressive-like beha.ior in ma.e a.d fema.e ra.s. Ra.s were sha.opera.ed (Sh) or pinea.ectomized (Px) a.d following subgroups were selected 1) Px/Na.l (0.9%) a.d Sh/Na.l (0.9%) : ra.s injected subcuta.eously, once da.ly for 8 weeks, with sa.ine solution Na.l (0.9%) a. vehicle;2) Px/Mel (4 mg/Kg) a.d Sh/Mel (4 mg/Kg): Ra.s simila.ly injected with 4 mg/Kg of Mel. a.l a.ima.s were housed under a.photoperiod of (LD:16/8). a.ter different trea.ments a.ima.s were tested in the open-field test (OFT), eleva.ed plus ma.e test (EPM) to determine a.xiety-like beha.ior, a.d forced swimming test (FST) to eva.ua.e depressive-like level. Our results revea.ed tha. level of a.xiety-like a.d depressive-like beha.ior a.e significa.tly higher in Px/Na.l (0.9%) when compa.ed to Sh/Na.l (0.9%) group, suggesting tha. pinelectomy induced a. a.xiogenic a.d depressa.t effects. The Px effects would be due to the a.sence of endogenous Mel synthesis a.d relea.e. a.ditiona.ly, we clea.ly demonstra.ed tha. the level of a.xiety-like a.d depressive-like beha.ior a.e higher in Px/Mel (4 mg/Kg) a.d Sh/Mel when compa.ed respectively to Px/Na.l (4 mg/Kg) a.d Sh/Na.l groups suggesting a. a.xiolytic a.d a.tidepressa.t effects of exogenous Mel. Beha.iora. responses were sex dependent since the difference between fema.es a.d ma.es, especia.ly, a.ter mela.onin a.ministra.ion, were sta.istica.ly significa.t. These experiments provide evidence tha. pinea.ectomy a.d Mel regula.ed emotiona.ly beha.ior in ma.e a.d fema.e ra.s.

Initia.ly, 4 mg/kg of mela.onin wa. da.ly a.ministered, a. 4:00 pm, to inta.t ma.e a.d fema.e ra.s during 8 weeks. Our results ha.e shown tha. the effect of chronic injection of Mel is sex dependent in the three beha.iors tests. Fema.es ra.s ha.e responded better tha. ma.es in beha.ior test study a.ter a.ministra.ion of mela.onin, this difference between the sexes ma. be rela.ed to the a.tion of sex hormones (a.drogens a.d estrogens) on beha.ior in ma.es a. well a. in fema.es. Secondly, to determine the possible intera.tion between Mela.onin a.d steroid hormones, Ovx/sha. fema.e received Mel a. dose of 4mg/kg a.one or Na.l (0.9%) a.one, a.d Orx/sha. ma.e received Mel a. dose of 4 mg/kg a.one or Na.l (0.9%) a.one da.ly a.d during 8 weeks of trea.ment a. 4:00 pm. a.l a.ima.s were tested in the open-field test, eleva.ed plus ma.e test for a.xiety beha.ior study, a.d forced swimming test for depression beha.ior study. Results revea.ed tha. Mel exerts a. a.xiolytic a.d a.tidepressa.t effects in the orchidectomized ma.es a.d in inta.t fema.es, confirming tha. the suppression of a.drogens by orchidectomy improved a.xiolytic a.d a.tidepressa.t effects of mela.onin in ma.es. However in fema.es, the suppression of estrogen by ova.iectomy ma.ked the a.tidepressa.t a.d a.xiolytic effects of mela.onin. Our results confirmed tha. the a.tidepressa.t a.d a.xiolytic effects of mela.onin a.e linked to sex hormones.

The present study wa. underta.en to exa.ine whether Cu chronic a.ministra.ion ca. induce a.tera.ions of a.fective beha.ior especia.ly a.xiety a.d depression levels in ma.e a.d fema.e ra.s. Twenty-four ra.s, for ea.h gender, divided in control a.d three test groups (n = 6), were injected intra.eritonea.ly with sa.ine (0.9% Na.l) or CuCl2 (0.25 mg/kg, 0.5 mg/kg a.d 1 mg/kg) for 8 weeks. a.ter trea.ment period, a.ima.s were tested in the open-field, eleva.ed plus ma.e tests for a.xiety-like beha.ior, a.d forced swimming test for depression-like beha.ior. Results demonstra.ed tha. Cu a.ministered chronica.ly, exerts a. a.xiogenic effect in ra.s. In the OFT, Cu decrea.es the TCa.a.d NRC pa.a.eters without modifying the locomotor a.tivity represented by the NTS pa.a.eter. With rega.d to EPM, Cu decrea.es TOa.a.d EOa.pa.a.eters without modifying the Ta. pa.a.eter. a.significa.t increa.e in depression-like symptoms wa. a.so exhibited by Cu trea.ed ra.s (p 2 showed ma.imum a.xiety-like a.d depression-like symptoms a. compa.ed to controls a. well a. from the other two doses indica.ing dose-dependent effects of chronic Cu a.ministra.ion. Overa.l, these results suggest tha. intoxica.ion with Cu ha. potentia.ly deleterious effects on bra.n a. reflected in beha.iora. dysfunctions such a. depression a.d a.xiety.

The a.m of this study wa. to determine the effect of pha.ma.ologica. doses of mela.onin (MEL) a.d L-tryptopha. (L-TRP) on depression-like beha.ior in fema.e ra.s submitted to the forced swimming test (FST) a.ter 2, 4, 6 or 8 weeks of trea.ment. This will a.low exploring the different mecha.isms of L-TRP a.tions pa.ticula.ly tha. due to its conversion into MEL. For this purpose, four groups of 24 ra.s ea.h were constituted;(Group 1: Control): received sa.ine solution Na.l (0.9%), (Group 2: MEL4): received 4 mg/Kg of MEL, (Group 3: L-TRP4): received 4 mg/Kg of L-TRP a.d (Group 4: L-TRP20): received 20 mg/Kg of L-TRP. a.ima.s of ea.h group were distributed on 4 subgroups of 6 ra.s submitted to different time trea.ments. The dura.ion of immobility (TIM) a.d struggling period (TST) of ra.s in FST were mea.ured a.ter 2, 4, 6 a.d 8 weeks of drug trea.ment a.d the effects of MEL a.d L-TRP were compa.ed. Chronica. a.ministra.ion of different doses of MEL or L-TRP fa.led to induce a.y a.ti-depressa.t a.tivity in ra.s subjected to FST a.ter 2 weeks of trea.ment. However, a.ter 4 weeks, da.ly a.ministra.ion of MEL a. 4 mg/Kg significa.tly reduced the immobility period a.d enha.ced struggling time. a.ter 6 weeks, MEL a. 4 mg/Kg a.d L-TRP a. 20 mg/Kg were both effective in reducing immobility a.d increa.ing struggling movement, their effects being sta.istica.ly compa.a.le. a.l trea.ments were a.le to significa.tly reduce immobility time a.d increa.e struggling dura.ion a.ter 8 weeks, but L-TRP a. 4 mg/Kg wa. less potent tha. MEL a.d L-TRP a. 20 g/Kg. The a.tidepressa.t-like a.tivity of L-TRP wa. dose a.d time dependent, a.d tha. of MEL wa. time dependent. Two new quinoxa.inone deriva.ives ha.e been synthesized a.opting the HONG method, a.d investiga.ed for some neuropha.ma.ologica. effects (a.xiety- a.d depressive-like responses) in ra.s. The present experiment sought to determine whether trea.ment with these compounds produces cha.ges in a.fective responses. We found tha. the chronic injection of 6-nitro-2(1H)-quinoxa.inone (NQu) showed obvious a.xiolytic- a.d a.tidepressa.t-like effects, respectively, mea.ured in the beha.iora. tests of Eleva.ed Plus Ma.e (EPM) a.d Forced Swim Test (FST). a. the dose of 30 mg/kg, NQu showed a.compa.a.ive a.xiolytic-like effect in ra.s a. dia.epa. (Dz) (1 mg/kg), a.d a.compa.a.ive a.tidepressa.t effect a. clomipra.ine (Clmp) (2 mg/kg;i.p). The 2(1H)-quinoxa.inone (Qu) significa.tly reduced depressive-like responses a. eva.ua.ed in FST, wherea. no a.xiolytic-like effect wa. found a. mea.ured by open field test (OF). a.ditiona.ly, the locomotor a.tivity levels were una.fected by trea.ment a. mea.ured by OF a.d EPM.
