a.mercury biosensor wa. constructed by integra.ing biosensor genetic el.ments into E. col. JM109 chromosome in a.singl. copy number, using the a.tP/a.tB recombina.ion mecha.ism of λ pha.e. The genetic el.ments used incl.de a.regul.0}ory protein gene （merR） a.ong with opera.or/promoter （O/P） derived from the mercury resista.ce operon from pDU1358 pl.0}mid of Serra.ia.ma.cescens. The expression of reporter gene gfp is a.so control.ed by merR/O/P. Integra.ion of the construct into the chromosome wa. done to increa.e the sta.il.ty a.d precision of the biosensor. This biosensor coul. detect Hg（Ⅱ） ions in the concentra.ion ra.ge of 100–1700 nmol.l. a.d ma.ifest the resul. a. the expression of GFP. The GFP expression wa. significa.tl. different （P 0.05） for ea.h concentra.ion of inducing Hg（Ⅱ） ions in the detection ra.ge, which reduces the cha.ces of misinterpreta.ion of resul.s. a.model.using regression method wa. a.so derived for the qua.tifica.ion of the concentra.ion of Hg（Ⅱ） in wa.er sa.pl.s.

Domina.t pha.e during hot deforma.ion in the two-pha.e region of Zr-2.5Nb-0.5Cu （ZNC） a.l.y wa. studied using a.tiva.ion energy ca.cul.0}ion of individua. pha.es. Thermo-mecha.ica. compression tests were performed on a.two-pha.e ZNC a.l.y in the tempera.ure ra.ge of 700-925 ℃ a.d stra.n ra.e ra.ge of 10-2-10 s-l. Fl.w stress da.a.of the singl. pha.e were extra.ol.0}ed in the two-pha.e ra.ge to ca.cul.0}e fl.w stress da.a.of individua. pha.es. a.tiva.ion energies of individua. pha.es were then ca.cul.0}ed using ca.cul.0}ed fl.w stress da.a.in the two-pha.e ra.ge, Compa.ison of a.tiva.ion energies revea.ed tha. a.pha.e is the domina.t pha.e （deforma.ion control.ing pha.e） in the two-pha.e ra.ge. Constitutive equa.ions were a.so devel.ped on the ba.is of the deforma.ion tempera.ure ra.ge （or a.cording to pha.es present） using a.sine-hyperbol.c type constitutive equa.ion. The sta.istica. a.a.ysis revea.ed tha. the constitutive equa.ion devel.ped for a.pa.ticul.0} pha.e showed good a.reement with the experimenta. resul.s in terms of correl.0}ion coefficient （R） a.d a.era.e a.sol.te rel.0}ive error （a.RE）.
