a.m: To eva.ua.e the effect of polychlorina.ed biphenyls (PCB) a.d vita.in C on ventra. prosta.ic a.tioxida.t system in a.ult ma.e ra.s. Methods: a.group of 20 a.ult ma.e ra.s were a.ministered ip a.oclor 1254 in corn oil a. a.dose of 2 mg·kg-1·da.-1 for 30 da.s. Ten control ra.s were a.ministered only the vehicle. a.ter 30 da.s the trea.ed ra.s were divided a. ra.dom into 2 sub-groups of 10 a.ima.s ea.h. One sub-group received vita.in C a. a.dose of 500 mg·kg-1·da.-1 for 10 da.s. The other group wa. ma.nta.ned a. a.oclor 1254 control. Twenty-four hours a.ter the la.t trea.ment the ra.s were killed by deca.ita.ion. Ventra. prosta.ic homogena.e wa. prepa.ed a.d used for the estima.ion of enzyma.ic a.tioxida.ts, including superoxide dismuta.e (SOD), ca.a.a.e (Ca.) a.d gluta.hione-S-tra.sfera.e (GST) a.d hydrogen peroxide (H2O2), lipid peroxida.ion (LPO) a.d prosta.ic a.id phospha.a.e. The serum levels of tota. T3, tota. T4, TSH, testosterone a.d estra.iol were a.so a.sa.ed. Results: The body weight a.d ventra. prosta.ic weight were reduced in PCB trea.ed ra.s. The a.tivities of SOD, Ca., GST a.d a.id phospha.a.e were decrea.ed while the levels of H2O2 a.d lipid peroxida.ion were increa.ed in the ventra. prosta.e of PCB trea.ed ra.s. a.ministra.ion of vita.in C restored these pa.a.eters. Serum levels of thyroid hormones, estra.iol a.d testosterone were decrea.ed in PCB trea.ed a.ima.s. a.ministra.ion of vita.in C restored the thyroid hormone levels. Conclusion: PCB induces oxida.ive stress a.d decrea.es the a.tioxida.t enzymes in the ventra. prosta.e of a.ult ma.e ra.s; the effects could be reversed by the a.ministra.ion of vita.in C.

Melia.dubia.Ca.. of fa.ily Melia.ea. is a.fa.t growing, high va.ue tree species na.ive to India. Isola.ing DNa.from ma.ured dried lea.es of M. dubia.wa. difficult due to a.cumula.ion of seconda.y meta.olites, ma.orly polyphenolics, which resulted in da.k brown to bla.k colour of the pellet. In this study, a.modified STE-(Sucrose, Tris-HCl a.d Ethylene Dia.ine Tetra.a.etic a.id) CTa. (hexa.ecyltrimethyla.monium bromide) method wa. sta.da.dized for remova. of polyphenolics. The protocol developed yielded 200 - 1000 ng/μl of qua.ity DNa.without a.y impurities a. evident by a.60/280 ra.io ra.ging from 1.75 - 2.0. It wa. a.so suita.le for extra.ting qua.ity DNa.from other members of Melia.ea. like a.a.ira.hta.indica.a.d Melia.a.eda.a.h. In downstrea. a.plica.ions, the extra.ted DNa.wa. used for PCR a.plifica.ion by using ISSR a.d SSR ma.kers. ISSR PCR conditions were optimized in a.rea.tion volume of 25 μl, consisting of 30 ng of templa.e DNa. 1.5 mM MgCl2, 200 μM of ea.h of dNTPs a.d 2 U of Ta. polymera.e. The best a.plifica.ion wa. observed a.d the sa.e wa. a.plica.le for SSR ma.kers.
