Can DNA fragmentation of neat or swim-up spermatozoa be used to predict pregnancy following ICSI of fertile oocyte donors？

This study compared the potential of assessing sperm DNA fragmentation （SDF） from neat semen and the subsequent swim-up （SU） procedure to predict pregnancy when conducting ICSI of fertile donor oocytes. Infertile females （n=81） were transferred embryos resulting from intracytoplasmic sperm injection （ICSI） of their partner＇s spermatozoa and proven donor oocytes. This model normalized the impact of female factor in putative sperm DNA repair. Semen was blindly assessed for SDF using Halosperm immediately following ejaculation （NS） and after swim-up at the time of ICSI fertilisation. There was a decrease in SDF values of the ejaculated semen sample following the swim-up protocol （P=0.000）. Interestingly, pregnancy could be equally predicted from SDF values derived from either neat or swim-up semen samples. Receiver operator curves and the derived Youden＇s indices determined SDF cutoff values for NS and SU of 24.8% and 17.5%, respectively. Prediction of pregnancy from NS SDF had a sensitivity of 75% and a specificity of 69%, whereas for SU SDF was 78% and 73%, respectively. While increased levels of SDF negatively impact reproductive outcome, we have shown that a reduction in SDF following sperm selection using ICSI with proven donor oocytes is not mandatory for achieving pregnancy. This suggests that a certain level of DNA damage that is not detectable using current technologies could be impacting on the relative success of assisted reproductive technology （ART） procedures. Consequently, we propose a modification of the so called ＇iceberg model＇ as a possible rationale for understanding the role of SDF in reproductive outcome.

Current cell-wall models assume no covalent bonding between cellulose and hemicelluloses such as xyloglu-can or mixed-linkageβ-D-glucan(MLG).However,Equisetum hetero-trans-β-glucanase(HTG)grafts cellu-lose onto xyloglucan oligosaccharides(XGOs)-and,we now show,xyloglucan polysaccharide-in vitro,thus exhibiting CXE(cellulose:xyloglucan endotransglucosylase)*** addition,HTG also catalyzes MLG-to-XGO bonding(MXE activity).In this study,we explored the CXE action of HTG in native plant cell walls and tested whether expansin exposes cellulose to HTG by disrupting hydrogen *** quantify and visu-alize CXE and MXE action,we assayed the sequential release of HTG products from cell walls pre-labeled with substrate *** demonstrated covalent cellulose--xyloglucan bonding in plant cell walls and showed that CXE and MXE action was up to 15%and 60%of total transglucanase action,respectively,and peaked in aging,strengthening tissues:CXE in xylem and cells bordering intercellular canals and MXE in *** bacterial expansin(EXLX1)strongly augmented CXE activity in *** and MXE ac-tion in living Equisetum structural tissues potentially strengthens stems,while expansin might augment the HTG-catalyzed CXE reaction,thereby allowing efficient CXE action in *** methods will enable surveys for comparable reactions throughout the plant ***,engineering similar hetero-polymer formation into angiosperm crop plants may improve certain agronomic traits such as lodging tolerance.

Time Course of Age-dependent Changes in Intraocular Pressure and Retinal Ganglion Cell Death in DBA/2J Mouse

Purpose:To characterizes the progression of glaucoma in DBA/2J mice by measuring intraocular pressure(IOP) and retinal ganglion cells(RGCs) numbers in mice of various ages. Methods:A quantitative assessment of the pathophysiology of the DBA/2J mice was performed and the C57/BL6 mice was used as control. The IOP was measured by the servo-null micropipette system; the regional patterns of the loss of RGCs were determined by cell count of retrogradely-labeled RGCs. Results:The baseline IOP for DBA/2J mice at 7 weeks was (16.6 ± 1.2)mm *** IOP increased extend to 12 months, with the peak of (25.2 ± 1.2)mm Hg at 6 months of age. Retinal ganglion cell numbers did not decrease relative to control until 12 months of age(P=0.006), when the loss was proportionally higher in peripheral regions(P<0.05). Conclusion:The elevation in IOP precedes the loss of RGCs by several months. RGCs cell loss occurs particularly in peripheral regions of the retina. These findings expand our understanding of the changes in DBA/2J mice and provide information for experiments design when they are used as a glaucoma model for future studies of RGCs degeneration in glaucoma.

Nrf2 and Snail-1 in the prevention of experimental liver fibrosis by caffeine

Constructive thinking,rational intelligence and irritable bowel syndrome

AIM： To evaluate rational and experiential intelligence in irritable bowel syndrome （IBS） sufferers. METHODS： We recruited 100 subjects with IBS as per Rome 11 criteria （50 consulters and 50 non-consulters） and 100 healthy controls, matched by age, sex and educational level. Cases and controls completed a clinical questionnaire （including symptom characteristics and medical consultation） and the following tests： rational- intelligence （Wechsler Adult Intelligence Scale, 3rd edition）; experiential-intelligence （Constructive Thinking Inventory）; personality （NEO personality inventory）; psychopathology （MMPI-2）, anxiety （state-trait anxiety inventory） and life events （social readjustment rating scale）. Analysis of variance was used to compare the test results of IBS-sufferers and controls, and a logistic regression model was then constructed and adjusted for age, sex and educational level to evaluate any possible association with IBS. RESULTS： No differences were found between IBS cases and controls in terms of IQ （102.0 ± 10.8 vs 102.8 ± 12.6）, but IBS sufferers scored significantly lower in global constructive thinking （43.7± 9.4 vs 49.6 ± 9.7）. In the logistic regression model, global constructive thinking score was independently linked to suffering from IBS [OR 0.92 （0.87-0.97）], without significant OR for total IQ. CONCLUSION： IBS subjects do not show lower rational intelligence than controls, but lower experiential intelligence is nevertheless associated with IBS.

Prevalence of metabolic syndrome, obesity and diabetes type 2 in cryptogenic cirrhosis

AIM: To evaluate the prevalence of metabolic syndrome (MS), obesity and type 2 diabetes mellitus (T2DM) in a group of Mexican Mestizo patients with cryptogenic cirrhosis (CC) and to compare this group with patients with cirrhosis secondary to other causes (disease controls). METHODS: Patients with CC, diagnosed between January, 1990 and April, 2005, were included in a retrospective study. Patients with cirrhosis caused by chronic hepatitis C, alcohol abuse or autoimmune hepatitis (AIH) served as disease controls. RESULTS: A total of 134 patients with CC were analyzed. Disease controls consisted of 81 patients with chronic hepatitis C, 33 with alcohol abuse and 20 with AIH. The median age of patients with CC was 57 years (range, 16-87); 83 (61.9%) patients were female; 53 (39.6%) were Child A, 65 (48.5%) Child B, and 16 (11.9%) were Child C cirrhosis. The prevalence of MS (29.1% vs 6%; P < 0.001), obesity (16.4% vs 8.2%; P = 0.04) and T2DM (40% vs 22.4%; P = 0.013) was higher in CC patients than in disease controls. There were no differences in sex, age or liver function tests between the two groups. CONCLUSION: The prevalence of MS, obesityand T2DM were higher in patients with CC than in patients with cirrhosis secondary to others causes. Our findings support the hypothesis that non-alcoholic steatohepatitis (NASH) plays an under-recognized role in CC.

Pathways of Phosphorus Absorption and Early Signaling between the Mycorrhizal Fungi and Plants

This review highlights the key role that mycorrhizal fungi play in making phosphorus(Pi)more available to plants,including pathways of phosphorus absorption,phosphate transporters and plant-mycorrhizal fungus symbiosis,especially in conditions where the level of inorganic phosphorus(Pi)in the soil is *** fungi colonization involves a series of signaling where the plant root exudates strigolactones,while the mycorrhizal fungi release a mixture of chito-oligosaccharides and liposaccharides,that activate the symbiosis process through gene signaling pathways,and contact between the hyphae and the *** the symbiosis is established,the extraradical mycelium acts as an extension of the roots and increases the absorption of nutrients,particularly phosphorus by the phosphate *** then moves along the hyphae to the plant root/fungus *** transfer of Pi occurs in the apoplectic space;in the case of arbuscular mycorrhizal fungi,Pi is discharged from the arbuscular to the plant’s root symplasm,in the membrane that surrounds the *** is then absorbed through the plant periarbuscular membrane by plant phosphate ***,plants can acquire Pi from soil as a direct absorption *** a result of this review,several genes that codify for high-affinity Pi transporters were *** plants,the main family is Pht1 although it is possible to find others such as Pht2,Pht3,Pho1 and *** in plants,mycorrhizal fungi have genes belonging to the Pht1 *** arbuscular mycorrhizal fungi we found L1PT1,GiPT,MtPT1,MtPT2,MtPT4,HvPT8,ZmPht1,TaPTH1.2,GmosPT and ***1,HcPT2 and BePT have been characterized in ectomycorrhizal *** gene has a different way of expressing *** this review,we present diagrams of the symbiotic relationship between mycorrhizal fungi and the *** knowledge allows us to design solutions to regional problems such as food production in soils with low levels of Pi.

Papillary fibroelastoma is an uncommon tumor. Recognitions of this tumor have been improved thanks to modern imaging advances, which have made more patients to be identified, but at the same time, the management of this disease is still controversial. There aren’t enough studies to basic clinical actions. We present 2 interesting cases involving the semilunar valves. The purpose of presenting these cases is to review this rare disease and the superiority of the three-dimensional echocardiography in their diagnosis.

Introduction, Collection and Characterization of Lotus spp. Germplasm in Chile

The information related with results of a project on germplasm characterization of three Lotus species, important as forage plants, curried out in five environments of the central-southern zone of Chile, was reviewed. An evaluation was made of 12 L. corniculatus cultivars introduced from different parts of the world, as well as germplasm naturalized in Chile, being 11 accessions of L. tenuis and 21 accessions ofL. uliginosus collected from 33°S to 38°S and 38°S to 45°S, respectively. Phenological, morphological, and agronomical characters were evaluated. Nitrogen fixation in field conditions and concentrations of condensed tannins in herbage were also included. It is concluded that there are cultivars of high value in L. corniculatus introduced germplasm which can be recommended for different environments of the central zone of Chile, as well as for local species breeding programmes. Information obtained from L. tenuis and L. uliginosus accessions shows genetic variability within both species. L. tenuis germplasm is adapted to clay, medium-textured, or sandy soils with water restrictions and phosphorous deficiency. Therefore, characterized accessions could be used to breed cultivars for low input production systems in constrained environments. L. uliginosus accessions include genotypes collected in sites with acid soils and variable tannin content in plant tissues that may be promissory genetic materials for breeding programmes with the objective of producing cultivars for animal production systems for the wet and acid soils in the southern zone of the country.
