Objective Colorectal cancer is one of the most common malignancies in the world and a multipathway *** T-lymphocyte antigen-4(CTLA-4) is a potent immunoregulatory molecule that suppresses antitumor response by down-regulating T-cell *** most studied +49A>G polymorphism of CTLA-4 gene has been associated with various kinds of *** aim was to investigate the association between this genetic variant and the risk and progression of colorectal cancer in Chinese. Method We conducted a case-control study of 124 colorectal cancer cases and 407 healthy *** was extracted from blood specimens,and +49A>G polymorphism in the CTLA-4 gene was genotyped by polymerase chain reaction-ligation detection reaction(PCR-LDR).Results In our study group,the frequency of AG or GG or carrying at least one G allele at position +49 was significantly different in colorectal cancer patients and the control group,indicating that the risk of CRC was significantly higher among subjects with the AG or GG genotype or carrying at least one G allele at position +49 than among the subjects with the AA ***,we observed no association between CTLA-4 +49A>G polymorphism and the progression of CRC including tumor stage,tumor grade,status of lymph node and distant ***,the CTLA-4 +49A allele was in non-significantly higher numbers in CRC patients with distant *** Our results suggested that CTLA-4 +49A>G polymorphism was associated with an increased risk of colorectal cancer,but this polymorphism did not play an important role in Chinese patients with CRC.

Adenovirus-expressed preS2 antibody inhibits hepatitis B virus infection and hepatic carcinogenesis

Isolated metastatic melanoma of the pancreas is very ***,there is very limited experience with surgical resection of pancreatic *** potential benefit of metastasectomy can improve the quality of life and survival time of *** present a case of a 39-year-old Chinese male with a solitary pancreatic tumor which was considered a cystic benign lesion for *** and immunohistochemistry showed that the tumor in pancreatic tail was a metastasis from a malignant melanoma of the *** other metastastic foci were found in *** tumor was completely resected with combined distal pancreatectomy and *** patient has survived 25 mo without any signs of local recurrence or other metastatic lesions after operation,indicating that complete surgical resection of a solitary metastatic melanoma of the pancreas can prolong the survival time of patients.

CYP2E1 Pst Ⅰ/Rsa Ⅰ polymorphism and colorectal cancer risk:A meta-analysis

AIM:To clarify the association between CYP2E1 PstⅠ/RsaⅠ polymorphism and susceptibility to colorectal ***:A meta-analysis based on 10 eligible casecontrol studies involving 4979 cases and 6012 controls was carried out to summarize the data on the association between CYP2E1 RsaⅠ/PstⅠ polymorphism and colorectal cancer ***:In comparison of the homozygote c2c2 and c2 carriers(c1c2 + c2c2) and the homozygous wild-type genotype(c1c1),no association was found between CYP2E1 RsaⅠ/PstⅠ polymorphism and colorectal cancer risk [odds ratio(OR)=1.24(95% CI:0.93-1.66) for c2c2;OR = 1.02(95% CI:0.88-1.19) for c2 carriers].In stratif ied analysis,Caucasians with c2c2 homozygote appeared to have an increased risk of colorectal cancer(OR=2.67,95% CI:1.03-6.89,P=0.043),no signif icant associations were found in other ***:c2c2 homozygote of CYP2E1 PstⅠ/RsaⅠ polymorphism may be associated with the increased risk of colorectal cancer in Caucasians,which needs further investigations.

Phage display selection on whole cells yields a small peptide specific for HCV receptor human CD81

The human CD81 (hCD81), the most recently proposed receptor of hepatitis C virus (HCV), can especifically bind to HCV envelope glycoprotein 2 (E2). In this study, hCD81-expressing murine NIH/3T3 cells were used to select hCD81-binding peptides from a phage displayed nonapeptide library (PVIII9aaCys). Eighteen of the 75 clones selected from the library showed specific binding to the hCD/81-expressing NIH/3T3 cells by enzyme linked immunosorbent assay (ELISA) and competitive inhibition test. Twelve out of the 18 clones shared the amino acid motif SPQYWTGPA. Sequence comparison of the motif showed no amino acid homology with the native HCV E2. The motif-containing phages could competitively inhibit the ability of HCV E2 binding to native hCD81-expressing MOLT-4 cells, and induce HCV E2 specific immune response in vivo. These results suggest that the selected motif SPQYWTGPA should be a mimotope of HCV E2 to bind to hCD81 molecules. Our findings cast new light on developing HCV receptor antagonists.

In order to study structure-function details of TGF-β1, the recombinant mature form of rat TGF-β1 was expressed in bacteria. Synthesis of the 112 amino-acid carboxyl-terminal part of TGF-β1 (amino acid 279-390) was controlled by an inducible gene expression system based on bacteriophage T7 RNA polymerase. This system allowed an active and selective synthesis of recombinant TGF-β1. The molecular weight of expressed TGF-β1 monomer determined on SDS-polyacrylamide gel under reducing conditions was about 13 kD. Serial detergent washes combined with a single gel-filtration purification step were sufficient to purify the expression product to homogeneity. Amino-terminal sequencing revealed that the N-terminal of the recombinant protein was identical to the published data. In Western blot analysis the recombinant polypeptide showed excellent antigenicity against polyclonal TGF-β1 antibody. The mature recombinant rat TGF-β1 expressed in this study provides a useful tool for future detailed structural and functional studies.

Relation between upregulation of CD40 system and complex stenosis morphology in patients with acute coronary syndrome

AIM: To investigate whether upregulation of CD40-CD40 ligand system is related to matrix metalloproteinases level and stability of coronary atherosclerotic plaque in patients with acute coronary syndrome (ACS). METHODS: Sixteen normal controls and 56 patients including 24 with stable angina (SA), 20 with unstable angina (UA), and 12 with acute myocardial infarction (AMI) were investigated. The expression of CD40 and CD40L on platelet was analyzed by flow cytometry. Serum soluble CD40L (sCD40L), MMP-9 and MMP-3 level was determined by ELISA. All coronary stenosis with ≥30 % diameter reduction were assessed by angiographic coronary stenosis morphology. RESULTS: Patients with ACS showed a significant increase of CD40 (75±12 MIF) and CD40L (13±4 MIF) coexpression on platelets compared with control and SA group (P6 μg/L, P60, P<0.01). CONCLUSION: Patients with ACS show increased coexpression of CD40 system, especially expression of CD40L, which may create a proinflammatory and prothrombotic milieu for aggravating the development of atherosclerosis and instability of atherosclerotic plaques, and may be a valuable marker for predicting the severity of ACS.
