Serum response factor play a regulative role in the gene expression in heart failure

Objective To investigate the relationship between transcription factor and the change of protein expression levels in heart failure. Methods Bioinformatic method was used to analyze the data of binding-sites on the 5' flaking regions of four genes whose mRNA level changed in failing heart from three databases about nucleic acid-EMBL, transcriptional regulation factor-TRANSFAC and protein-SWISS-PORT. The expression level of selected transcription factor was determined by immunohischemical method. Results Nine transcription factors were inferred to influence the proteins' levels in occurrence and development of heart failure. Serum response factor (SRF) was selected from the nine factors and assayed. The results showed that there was a higher level of SRF in healthy group than in chronic heart failure (CHF), and the level was associated with the degree of CHF. It was also found that there was a relative higher level of SRF in the acute myocardial infarction (AMI) than that in CHF, but which was lower than the healthy. Conclusion It showed that SRF had a quantitative change in the development of heart failure, and suggested SRF might play an important regulative role in heart failure. The expression changes of proteins related to myocardial function might be regulated by the quantitative change of transcription factor(s).

Expression of the difference between the Cold(Han) and Hot(Re) natures of traditional chinese medicines(Strobal and Rhubarb) based on the cold/hot plate differentiating assay

In this study, objective differences between the Cold (Han) and Hot (Re) nature of traditional chinese medicines, e.g. Strobal and Rhubarb, are determined by using a cold/hot plate differentiation technology. A novel, self-designed cold/hot plate differentiating instrument, with methodological study, was used to investigate the intervention of Strobal and Rhubarb on the temperature tropism of mice. Compared with the ICR and BALB/c mice, it was found that KM mice on the cold/hot plate were more sensitive to the change of temperature, within the tolerant temperature range of 15-40 ℃ . The temperature tropism behavior of mice is influenced by treatment with Rhubarb and Strobal, as is the activity of ATPase in liver tissue. These trends are consistent with the definition of the Cold/Hot nature of chinese medicines based on traditional chinese medicinal theory. This study showed that the differences of the Cold/Hot nature of traditional chinese medicines. might be objectively represented by the temperature tropism of animal by means of cold/hot differentiating assay.

Protective effect of tubuloside B on TNFα-induced apoptosis in neuronal cells

AIM: To investigate the neuroprotective effect of tubuloside B, one of the phenylethanoids isolated from the stems of Cistanche salsa, on tumor necrosis factor-alpha (TNFα)-induced apoptosis in SH-SY5Y neuronal cells. METHODS: Cell viability was analyzed using MTT assay. Apoptotic cells were detected using Hoechst33342 staining, and confirmed by DNA fragmentation and flow cytometric analysis. The activity of caspase-3 was measured with special assay kit. The concentration of free intracellular calcium was determined with the probe Indo-1 by spectrometer. The level of intracellular reactive oxygen species and the potential of mitochondrial membrane were determined by laser scanning confocal microscopy (LSCM) combined with fluorescence probe H2DCFDA or JC-1 respectively. RESULTS: SH-SY5Y cells treated with TNFa 100 μg/L for 36 h showed typical morphological changes of apoptosis. DNA ladder could be observed by agarose gel electrophoresis. Therapeutic effects of berberine in impaired glucose tolerance rats and its influence on insulin secretion

AIM: To explore the anti-diabetic effects of berberine and its influence on insulin secretion. METHODS: Impaired glucose tolerance rats induced by iv injection of streptozotocin 30 mg/kgwere treated with berberine 187.5 and 562.5 mg/kg while fed with high fat laboratory chow. After rats were treated for 4 weeks, oral glucose tolerance was determined, and for 8 weeks, the fasting blood glucose, insulin, lipid series were determined. In insulin secretion experiments, berberine 93.75, 187.5, and 562.5 mg/kg was administered orally to BALB/c mice at a bolus. The murine serum was collected 2 h after the berberine administration for insulin determination. Insulin released from HIT-T15 cells and pancreatic islets incubated with berberine 1-100 μmol/L for 12 h was determined. RESULTS: The levels of fasting blood glucose (7.4±1.5 or 7.3±1.3 vs 9.3±1.3 mmol/L), triglycerides (0.61±0.22 or 0.63± 0.17 vs 1.8±0.7 mmol/L), total cholesterol (1.8±0.3 or 1.9±0.3 vs 2.2±0.2 mmol/L), free fatty acid (456±93 or 460±72 vs 550±113 μmol/L) and apolipoprotein B (0.37±0.02 or 0.42±0.05 vs 0.46±0.04 g/L) were reduced greatly in berberine-treated groups at doses of 187.5 and 562.5 mg·kg-1·d-1, respectively as compared with those in control group (P5 or P5±0.3 or 1.4±0.3vs 1.1±0.1 g/L), apolipoprotein AI (0.80±0.08 or 0.87±0.08 vs 0.71±0.06 g/L) were significantly increased (P5 or P5 cells and murine pancreatic islets in a dose-dependent manner. Meanwhile murine serum insulin level (27.5±2.7 or 29±4 or 29±4 vs 24.3±2.8 pIU/L) was undoubtedly promoted and blood glucose (4.52±0.31 or 4.45±0.29 or 4.30±0.19 vs 4.87±0.21 mmol/L) was reduced after berberine administration at doses of 93.75, 187.5, and 562.5 mg/kg, respectively in the BALB/c mice. Effective use of corticosteroids in treatment of plastic bronchitis with hemoptysis in chinese adults

Aim:To investigate whether corticosteroids are effective in the treatment of plas-tic bronchitis with ***:A retrospective,clinical study *** two patients with only first episodes of plastic bronchitis withhemoptysis were divided into a steroid group(n=18)treated with glucocorticoids,and a non-steroid group(n=14).The supportive therapy was uniformly applied toboth groups,except for *** such as temperature and whiteblood cell counts were ***,the volume of hemoptysis andbronchial casts were evaluated in detail ***:There was no difference inthe demographic data and variables at baseline between both groups(all P>0.05).On days 5,6,7 and 8,the volume of hemoptysis was significantly decreased in thesteroid group compared with the non-steroid group(43±15 mL vs 117±33 mL ond 5,29±12 mL vs 97±23 mL on d 6,18±10mL vs 80±20 mL on d 7,and 13±8 mLvs 66±14 mL on d 8;all P5),and on d 10 after fibreoptic bronchoscopy,thecases with bronchial casts was reduced evidently in the steroid group in compari-son with the non-steroid group(OR=5.69,95% CI=1.76-43.6;P=0.005).There wasno significance in mechanical ventilation and mortality between both ***:Despite some limitations of this study,it has been demonstratedthat,on the basis of common supportive therapy,corticosteroids would be effec-tive and safe for the treatment of plastic bronchitis with hemoptysis.
