Sequence analysis of mRNA polyadenylation signals of rice genes

The formation of eukaryotic mRNAs involves the cleavage and polyadenylation of pre- mRNAs. To investigate the sequence requirement of putative polyadenylation signals (PASs), poly(A) sites and downstream elements (DUEs) in 3′-end-pro- cessing in rice, we compared expressed sequences tags (ESTs) with poly(A) extremity to full-length cDNA sequences and constructed a database of 12.69 pre- mRNA sequences in ?4.―+4. nt surrounding the poly(A) sites, which were from 9953 genes. The al- ternative poly(A) sites were revealed in approxi- mately 2.% of mRNAs. Nearly 80% of pre-mRNAs showed stringent requirement of the YA (CA or UA) at poly (A) sites for polyadenylation. About 7.9% had the AAUAAA signals on ?4.―?1 nt upstream of the poly(A) sites. Over 60% of mRNAs probably used the one- or two-base variants of AAUAAA hexamers as their PASs in 3′ fragments. The single-base variants of AAUGAA revealed the high frequency in 11.5% of 3′ fragments. The DUEs were detected in 90% of pre- mRNAs, especially more than half of the pre-mRNAs with multi-base variants of AAUAAA had the DUEs surrounding the poly(A) site. The location of DUE is also important for defining the cleavage site. Al- though most of the rice pre-mRNAs did not contain AAUAAA signal, the existence of downstream ele- ments ensured the efficiency of cleavage-polyade-

The coupled atmosphere-ocean general circulation models (CGCM) are nowbecoming the indispensable tools for the climate study. However, most models still suffer fromunrealistic simulations. One problem is the troublesome climate drift. It may come from someunresolved aspects. One possibility is the poor air-sea surface flux description now adopted inCGCM. The commonly used solution for this problem is the so-called flux correction. To deal with it,Hasslemann proposed an alternative solution of prospect. That is to incorporate the ocean surfacewave model into this coupled GCM system so as to establish a coupled atmosphere-wave-ocean *** necessity is clearly seen from the recent field experiment that air-sea momentum flux dependsstrongly on the sea state. So the explicit calculation of the air-sea flux with the aid of the oceansurface wave model is highly desired. Theoretical progress has provided feasibility to such ideaand experiments have been conducted. Recent work also reveals that coupling ocean surface wave modelwith oceanic general circulation model can result in much better reproduction of the upper oceanthermal structure. This further strengthens our belief in the final realization of the coupledatmosphere-wave-ocean modeling system.

Land Use of the Three Gorges Reservoir Area and the Effect on Its Landscape Pattern in the Recent 50 Years

The paper makes detailed analysis on the changing processes of the main types of land use in Three Gorges Reservoir Area through the techniques of RS （Remote Sense） and GIS （Geography Information System）. Based on the theory of landscape ecology, many types of landscape pattern indexes are calculated and made correlation analysis. The results indicate that： in 1995 the ecological environment of Three Gorges Reservoir Area was the most vulnerable in the recent 50 years, but resumed in 2.02. the landscape pattern diversity index, fractal dimension index and fragmentation index has close correlation with the area of land use/cover change （LUCC）. The research also releases that the increase of the cultivated land, grassland and construction land deteriorates the landscape ecological pattern, while the increase of forest land area will help restore the landscape ecological pattern to its original state. The rapid change of grassland is an important index of the change of landscape pattern, because it has the most effect on change of landscape pattern index.

Semi-analytical analysis of the response of the air temperature over the land surface to the global vegetation distribution

Response of the air temperature over the land surface to the global vegetation distribution is investigated, using a three-dimensional governing equation to simulate the steady, large-scale, limited amplitude perturbation of the free, inviscid and adiabatic atmosphere. The adoption of the static equation leads to a temperature governing equation in the terrain following coordinate. With the prescribed temperature as the upper boundary condition and the radiation balance as the lower boundary condition, the semi-analytical solution of the global circulation temperature can be calculated. In this article, only the air temperature (at 2.m height) over the land surface is analyzed, and the result suggests that this model can simulate the air temperature pattern over the land surface reasonably. A better simulation occurs when a simple feedback of the albedo on the temperature is included. Two sensitivity experiments are analyzed through this model. One suggests that the air temperature over the land surface descends obviously when the land surface is covered with ice all over, while another suggests that the air temperature rises a little when the land surface is covered with forest except the ice-covered area. This model appears to be a good tool to study the response of the air temperature to the vegetation distribution. Limitations of the model are also discussed.

Ethylene plays important roles in plant growth, development, and stress responses, and ethylene receptors have been identified and studied extensively in various plant species. Here we report the cloning of four ethylene receptor genes from soybean, i.e. GmETR1, GmERS1, GmETR2.and GmEIN4. Construction of the phylogenic tree showed that GmETR1 and GmERS1 belong to subfamily I whereas GmETR2.and GmEIN4.belong to subfamily II. The four ethylene receptor genes showed different tissue-specific expression patterns in roots, stems, leaves, cotyledons, flowers, pods and seeds of soybean. These genes were differentially regulated by various abiotic stresses and plant hormones. The possible roles of the four genes in soybean plant were also discussed.

Pre-irradiation with low-dose ^(12.C^(6+) beam significantly enhances the efficacy of AdCMV-p53 gene therapy in human non-small lung cancer

The combination of ionizing radiation and gene therapy has been investigated. However, there are very few reports about the combination of heavy-ion irradiation and gene therapy. To determine if the pre-exposure to low-dose heavy ion beam enhances the suppression of AdCMV-p53 on non-small lung cancer (NSLC), the cells pre-irradiated or non-irradiated were infected with 2., 4. MOI of AdCMV-p53. Survival fraction and the relative biology effect (RBE) were determined by clonogenic assay. The results showed that the proportions of p53 positive cells in 12.6+ beam induced AdCMV-p53 infected cells were more than 90%, which were signifi-cantly more than those in γ-ray induced AdCMV-p53 infected cells. The pre-exposure to low-dose 12.6+ beam significantly prevented the G0/G1 arrest and activated G2.M checkpoints. The pre-exposure to 12.6+ beam significantly improved cell to apoptosis. RBEs for the 12.6+ + AdCMV-p53 infection groups were 30%-60%, 2.%-130% and 30%-70% more than those for the 12.6+-irradiated only, AdCMV-p53 infected only, and γ-irradiation induced AdCMVp53 infected groups, respectively. The data suggested that the pre-exposure to low-dose 12.6+ beam significantly promotes exogenous p53 expression in NSLC, and the suppression of AdCMV-p53 gene therapy on NSLC.

Geographical Agglomeration of chinese Manufacturing Industries

Based on the plant level data from the First National Census of Basic Units, the Second National Census of Basic Units, and the First National Economic Census of China, this paper ex- amines the geographical agglomeration of chinese manufacturing industries at a more disaggregated level both by industrial classi-fication and spatial unit analysis. Applying the agglomeration index proposed by Maurel and Sedillot, studies show that the most agglomerated industries are resource-intensive, and capital and technology intensive while industries demanding localized inputs or serving localized markets or favored by local governments are fairly dispersed. The more disaggregated industries are more spa- tially agglomerated. At the finer spatial scale, industries are more dispersed. Results indicate that within-county spillovers are stronger than nearby-county spillovers, and within-prefecture spillovers are stronger than nearby prefecture spillovers. Local- ized spillovers are still quite substantial at a range beyond that of counties. Comparing the agglomeration index of chinese manu- facturing industries in 1996, 2.01 and 2.0{2. agglomeration seems to be a general tendency. Results also indicate that some industries have experienced remarkable changes in their levels of agglom- eration in the period 1996–2.0{2.

^(137)Cs Redistribution in Thin Stony Soil of a Carbonate Rock Slope in Southwest China

The fallout radionuclide cesium-137(137 Cs) has been widely employed as a tracer for assessment of soil loss from thick uniform soils;however,few studies have been conducted on thin stony soils on slopes underlain by carbonate rocks which are widely distributed in karst *** derived from 137 Cs measurement of soil samples collected along a carbonate rock slope with thin stony soil where neither soil erosion nor deposition occurred was used to investigate the characteristics of 137 Cs redistribution in a karst area of Southwest *** results indicated that the 137 Cs inventories of the surface soil on the slope studied were much lower than that of the local 137 Cs reference inventory and the 137 Cs activities were much higher than those on slopes with thick uniform *** spatial distribution of 137 Cs inventories was characterized by considerable *** high 137 Cs depletion in the stony soil of the slope studied was mainly because a considerable proportion of the fallout input of 137 Cs could be lost with runoff and the dissolution of carbonate particles in the soil promoted the loss of 137 *** demonstrated that the rates of soil loss could not be estimated from the degree of depletion of the 137 Cs inventory relative to the local reference inventory for the thin stony soil of the rocky slope underlain by carbonate rocks in the study area in the way that has been widely used in areas with thick uniform soils.

A Test on Different Types of the Time Curve of Hardness Ratio of Gamma-Ray Bursts based on the Curvature Effect

We analyzed a sample of 66 gamma-ray bursts （GRBs） and statistically confirmed the prediction on the time curve of the hardness ratio of GRBs made by Qin et al. based on the curvature effect. In their analysis, GRB pulses are divided into three types according to the shape of their raw hardness ratio （RHR） time curves, defined as to include the background counts to the signal counts, so as to make use of counts within small time intervals. Of the three types, very hard sources exhibit a perfect pulse-like profile （type 1）, hard bursts possess a pulse-like profile with a dip in the decay phase （type 2., and soft bursts show no pulse-like profile but have only a dipped profile （type 3）. In terms of the conventional hardness ratio, type 3 sources are indeed generally softer than those of type 1 and type 2. in agreement with the prediction. We found that the minimum value of RHR is sensitive in distinguishing the different types. We propose that GRB pulses can be classified according to the minimum value of RHR and that the different type sources may be connected with different strengths of the shock or/and the magnetic field.
