Hydrological Characteristics of the Rongbuk Glacier Catchment in *** Region in the Central Himalayas,china

From 8 April to 11 October in 2005, hydrological observation of the Rongbuk Glacier catchment was carried out in the Mr. Qomolangma （Everest） region in the central Himalayas, china. The results demonstrated that due to its large area with glacier lakes at the tongue of the Rongbuk Glacier, a large amount of stream flow was found at night, which indicates the strong storage characteristic of the Rongbuk Glacier catchment. There was a time lag ranging from 8 to 14.hours between daily discharge peaks and maximum melting （maximum temperature）. As melting went on the time lag got shorter. A high correlation was found between the hydrological process and daily temperature during the ablation period. The runoff from April to October was about 80% of the total in the observation period. Compared with the discharge data in 1959, the runoff in 2005 was much more, and the runoff in June, July and August increased by 69%, 35% and 14., respectively. The rising of temperature is a major factor causing the increase in runoff. The discharges from precipitation and snow and ice melting are separated. The discharge induced by precipitation accounts for about 20% of the total runoff, while snow and ice melting for about 80%.

Gal α(1, 3) Gal (gal epitope) is a carbohydrate epitope and synthesized in large amount by α(1, 3) galactosyltransferase [α(1, 3) GT] enzyme on the cells of lower mammalian animals such as pigs and mice. Human has no gal epitope due to the inactivation of α(1, 3) GT gene but produces a large amount of antibodies (anti-Gal) which recognize Gal α(1, 3) Gal structures specifically. In this study, a replication deficient recombinant adenoviral vector Ad5sGT containing pig α(1, 3) GT cDNA was constructed and characterized. Adenoviral vector-mediated transfer of pig α(1, 3) GT gene into human tumor cells such as malignant melanoma A375, stomach cancer SGC-7901, and lung cancer SPC-A-1 was reported for the first time. Results showed that Gal epitope did not increase the sensitivity of human tumor cells to human complement-mediated lysis, although human complement activation and the binding of human IgG and IgM natural antibodies to human tumor cells were enhanced significantly after Ad5sGT transduction. Appearance of gal epitope on the human tumor cells changed the expression of cell surface carbohydrates reacting with Ulex europaeus I (UEA I) lectins, Vicia villosa agglutinin (VVA), Arachis hypogaea agglutinin (PNA), and Glycine max agglutinin (SBA) to different degrees. In addition, no effect of gal epitope on the growth in vitro of human tumor cells was observed in MTT assay.

MASNUM wave-tide-circulation coupled numerical model (MASNUM coupled model, hereinafter) is de-veloped based on the Princeton Ocean Model (POM). Both POM and MASNUM coupled model are ap-plied in the numerical simulation of the upwelling off Yangtze River estuary and in Zhejiang coastal waters in summer. The upwelling mechanisms are analyzed from the viewpoint of tide, and a new mechanism is proposed. The study suggests that the tidally inducing mechanism of the upwelling in-cludes two dynamic aspects: the barotropic and the baroclinic process. On the one hand, the residual currents induced by barotropic tides converge near the seabed, and upwelling is generated to maintain mass conservation. The climbing of the residual currents along the sea bottom slope also contributes to the upwelling. On the other hand, tidal mixing plays a very important role in inducing the upwelling in the baroclinic sea circumstances. Strong tidal mixing leads to conspicuous front in the coastal waters. The considerable horizontal density gradient across the front elicits a secondary circulation clinging to the tidal front, and the upwelling branch appears near the frontal zone. Numerical experiments are de-signed to determine the importance of tide in inducing the upwelling. The results indicate that tide is a key and dominant inducement of the upwelling. Experiments also show that coupling calculation of the four main tidal constituents (M2, S2, K1, and O1), rather than dealing with the single M2 constituent, im-proves the modeling precision of the barotropic tide-induced upwelling.

An incubation experiment was carried out in laboratory to study the effect of temperature, moisture, phosphorus, organic matter, cropping and previous elemental sulfur application on elemental sulfur oxidation in four selected soils, fluvo-aquic soil, black soil, yellow-brown soil and red soil. In all the soils tested, sulfur oxidation rate was influenced by temperature and the temperature coefficient (Q10) values at the range from 10 to 30 ℃ were 4.4., 4.05, 6.19 and 3.71 for the four soils, respectively. The rate of sulfur oxidation was parabolically related to soil Water content. The optimum moisture content for the maximum oxidation rate was different among soils. Phosphorus increased the oxidation rate of elemental sulfur by 57.7%, 33.1%, 21.7% and 26.4. for the above four soils, respectively, compared with the control (no phosphorus applied). Organic material of corn straw which was ground and passed through a 0.5-mm sieve also increased the oxidation rate of elemental sulfur in the four soils by 59.8%, 7.8%, 39.2% and 54..4., respectively. Elemental sulfur which was applied previously to soils significantly enhanced the oxidation of elemental sulfur subsequently added and increased sulfur-oxidizing populations such as autotrophic elemental sulfur oxidizers with pH optimum 6.8, autotrophic thiosulfate oxidizers with pH optimum 6.8, heterotrophic thiosulfate oxidizers and heterotrophic sulfate producers. Cropping had little effect on elemental sulfur-oxidizing potentiality of soils.

CCAAT-binding proteins AngCP1 and AngCP2 of Aspergillus niger binding to DC ( - 4.9--4.4.bp) and PC( - 390--34. bp) of A . niger glaA gene were respectively purified by 20 % - 4. % saturated ammonium sulfate, gel filtration, HeparinSepharoseCl-6B chromatography and DNA sequence-specific affinity chromatography. Gel filtration and SDS-PAGE revealed that both AngCPl and AngCP2 were of 120 kD, comprised of two subunits of 34.kD and 50 kD. Western blot showed that the 34.kD subunits of both AngCPl and AngCP2 cross-reacted specifically with the anti-AngHAPC antiserum. Further electrophoretic mobility shift assay identified that AngCPl and AngCP2 were the same protein, designated AngCP. Southwestern blot showed that the affinity of the 34.kD sub-unit to DC was stronger than that of the 50 kD subunit to PC. These results suggested that interaction between AngCP, DC and PC plays an important role in the regulation of transcription of Aspergillus niger glaA gene.
