Tyroserleutide tripeptide affects calcium homeostasis of human hepatocarcinoma BEL-74.2 cells

This study aimed to observe the effects of tyroserleutide (tyrosyl-seryl-leucine, YSL) on the growth of human hepatocarcinoma BEL-74.2 that was transplanted into nude mice, and explore its anti-tumor mechanism preliminarily. YSL, at doses of 80 μg·kg?1·d?1, 160 μg·kg?1·d?1 and 320 μg·kg?1·d?1 significantly inhibited the growth of the human hepatocarcinoma BEL-74.2 tumor in nude mice, producing inhibition of 21.66%, 4..34., and 34.78%, respectively. Ultra structure of BEL-74.2 tumor in nude mice showed that YSL could induce tumor cells apoptosis and necrosis, cell organelle mitochondria and endoplasmic reticulum damage, and calcium over-load. By confocal laser scanning microscopy and flow cytometry, we found that 10 μg/mL YSL rapidly induced an increase of the concentration of cytoplasmic free calcium in BEL-74.2 cells in vitro, and maintained high concentrations of cytoplasmic free calcium for 1 h. Then the calcium concentration began to decrease after 2 h, and was lower than that of the control group at 4.h and 24.h (p4.2 cells in vitro, but had no effect on the calcium homeostasis or mitochondrial transmembrane potential of Chang liver hepatocytes. So affecting calcium homeostasis, then inducing apoptosis and necrosis may be a mechanism by which YSL inhibits the tumor growth in animal model.

FKBP23 in ER binds to BiP specifically

FKBP23 was found in mouse endoplasmic reticulum (ER) in 1998. It consists of an N-terminal peptidyl-prolyl cis-trans isomerase (PPlase) domain and a C-terminal domain with Ca2+ binding sites. The fusion protein of mouse FKBP23 and glutathione S-transferase (GST), GST-FKBP23, and the fusion protein of BiP, a member of heat shock protein 70 (Hsp 70) in ER, and GST, GST-BiP, were subcloned in E. coli, expressed and purified. The fusion proteins were restrictively digested by Factor Xa (FaXa) to obtain the free cloned proteins FKBP23 and BiP. With the assay of adsorption of free FKBP23 or BiP with GST-BiP or GST-FKBP23 attached to the Glutathione-Sepharose 4., the adsorbed FKBP23 or BiP could be detected by Immunoblot. It means that FKBP23 binds to BiP. Furthermore, BiP in leukocyte ER-extract can be adsorbed with GST-FKBP23 attached to the glutathione-Sepharose 4.. It shows that FKBP23 binds to natural BiP in ER, too. These experiments show that a PPIase binds to a molecular chaperone of the Hsp70 family.

Second mode unstable disturbance measurement of hypersonic boundary layer on cone by wavelet transform

Experimental investigation of hypersonic boundary layer instability on a cone is performed at Mach number 6 in a hypersonic wind tunnel. Time series signals of instantaneous fluctuating surface-thermal-flux are measured by Pt-thin-film thermocouple temperature sensors mounted at 28 stations on the cone surface in the streamwise direction to investigate the development of the unstable disturbance. Wavelet transform is employed as a mathematical tool to obtain the multi-scale characteristics of fluctuating surfacethermal-flux both in the temporal and spectrum space. The conditional sampling algorithm using wavelet coefficient as an index is put forward to extract the unstable disturbance waveform from the fluctuating surface-thermal-flux *** generic waveform for the second mode unstable disturbance is obtained by a phase-averaging technique. The development of the unstable disturbance in the streamwise direction is assessed both in the temporal and spectrum space. Our study shows that the local unstable disturbance detection method based on wavelet transformation offers an alternative powerful tool in studying the hypersonic unstable mode of laminar-turbulent transition. It is demonstrated that, at hypersonic speeds, the dominant flow instability is the second mode, which governs the course of laminar-turbulent transition of sharp cone boundary layer.

AIM:To investigate whether serum thymosinβ4.can provide diagnostic or prognostic information in liver failure patients caused by chronic hepatitis B virus(HBV) infection. METHODS:Serum thymosinβ4.levels were measured in 30 patients with acute-on-chronic liver failure(ACLF), 31 patients with chronic liver failure(CLF),30 patients with compensated liver cirrhosis(CR)and 32 patients with chronic hepatitis B and 30 healthy *** thymosinβ4.levels were measured by enzyme-linked immunosorbent assay and Child-Pugh and model for end-stage liver disease(MELD)scores were calculated for each patient on ***:Compared with healthy controls,serum thymosinβ4.levels in ACLF,CLF,CR and chronic hepatitis B patients were significantly lower,6.504. (4.7879-10.5314.μg/mL vs 0.4.32(0.2759-0.8768) μg/mL,0.6981(0.5209-1.2008)μg/mL,1.8053 (0.8110-2.3397)μg/mL,3.7803(1.8570-6.4.22)μg/mL, respectively(P4.in liver failure(ACLF or CLF)patients were markedly lower than that in CR(P4.levels and albumin, choline esterase,and platelet(P4.levels of non-survivors were significantly lower than that of survivors(P=0.007). The reliability of the improved e^N method for the transition prediction of boundary layers on a flat plate

The transition criterion in the improved eN method is that transition would occur whenever the velocity amplitude of disturbance reaches 1%-2% of the free stream velocity,while in the conventional eN method,the N factor is an empirical *** this paper the reliability of this key assumption in the improved eN method is checked by results of transition prediction by using the Parabolized Stability Equations(PSE).Transition locations of an incompressible boundary layer and a hypersonic boundary layer at Mach number 6 on a flat plate are predicted by both the improved eN method and the PSE *** from both methods agree fairly well with each other,implying that the transition criterion proposed in the improved eN method is reliable.

Etiology and management of hemmorrhage in spontaneous liver rupture:a report of 70 cases

AIM: To analyze the causes and management of hemorrhage in spontaneous liver ***: Seventy cases of spontaneous liver rupture were retrospectively analyzed for causes of hemorrhage and therapeutic effects of surgical ***: It was demonstrated that the causes of spontaneous liver rupture were primary liver cancer in 60 cases (85.7 %), cirrhosis in 3 cases (4.3 %), liver angioma in 2 cases (2.9 %), liver adenoma in 4.cases (5.7 %),and secondary liver cancer in 1 case (1.4.%). Hemostasis was achieved with surgical approaches in 68 cases (97.1 %) and non-surgical approaches in 2 cases (2.9 %). Surgical interventions induded suture, ligation of hepatic artery, hepatic artery chemoembolization and partial hepatic ***: The results suggest that surgical intervention is still the main therapeutic method and the best procedure that should be selected according to causes of disease and patient's condition and history.

Integrated modeling of nitrogen oxides formation in diesel engines

To account for the effects of both chemistry and flow turbulence, the present study proposes an integrated NO sub-model that combines the extended Zel’dovich mechanism and engine CFD computations to simulate the NO histories in a diesel engine. NOx sub-model parameters and pollutant formation mechanisms can be more easily investigated by solving the NOx sub-model. The new NO formation model incorporating the effects of both chemical kinetics and turbulent mixing was applied to simulate a diesel engine with a quiescent combustion chamber, and one with a re-entrant combustion chamber; the premise of the model being the reaction rate is mainly determined by a kinetic timescale and a turbulent timescale. The results indicate that the predicted NO formulation from the new model agrees well with the measured data. As the utilization of fossil fuels continues to increase, the control of NOx emissions is a worldwide concern; and it is imperative to understand fully the NOx reaction processes in combustion systems. This technology has the potential to enhance the application of various combustion techniques used to reduce NOx emissions from practical combustion systems.
