Background & Aims:Overexpression of estrogen receptors (ESRs) is implicated in the development of hepatocellular carcinoma (HCC) in both animal models and humans. We examined whether the ESR1 polymorphisms were related to HCC risk among chronic hepatitis B virus (HBV) carriers. Methods: Six ESR1 polymorphisms, which are (TA)n repeat in the promoter, T29C at codon 10 in exon 1, Pvu Ⅱ and Xba Ⅰ site in intron 1, C136474G at codon 325 in exon 4 and A252966G in intron 5, were genotyped in 248 HCC patients and 239 controls. The associations with the susceptibility to HCC were estimated by logistic regression. Allele-specific transcription difference of ESR1 mRNA was performed by real-time quantitative PCR. Results: We observed a statistically significantly increased susceptibility to HCC associated with the homozygous alleles with a high number of TA repeats (assigned as H/H genotype, odds ratio [OR] = 2.66,95% confidence interval [Cl] = 1.44-4.94, P= 0.0018), T29C C/C genotype (OR = 2.31, 95% Cl=1.25-4.26, P= 0.0076) and Pvu Ⅱ C/C genotype (OR = 2.19, 95% Cl = 1.27-3.78, P = 0.0048), compared with the homozygous alleles with a low number of TA repeats (assigned as L/L genotype), T29C T/T and Pvu Ⅱ T/T genotype, respectively. In accordance, the relative mRNA levels of the at-risk C allele of T29C were consistently higher than those of the T allele in heterozygous cells. Conclusions: Our findings suggest that the genetic polymorphism in ESR1 may play a role in mediating susceptibility to HCC in chinese HBV carriers.

已知微型倒转重复转座因子 (MITEs)广泛分布于真核生物基因附近的非编码区。为探讨MITEs在基因组一级结构中的分布规律 ,通过对 82kb的水稻BAC克隆l1 3 3 2全序列分析 ,精确鉴定了 1 0个蛋白质编码基因的位置和转录方向以及 7个MITEs的位置 ,毫无例外的观察到 ,MITEs只要存在 ,总是位于最近的基因的上游 ,不管该基因与之相距多远、转录方向如何。另外还发现 ,1个MITE被 2个基因共享。由于MITEs易于鉴定 ,因而这种位置模式可以用作探测其下游基因存在的工具。

＜正＞Analysis of single nucleotide polymorphisms（SNPs） emerges as an important tool in evolutionary biology and *** we report the in silico discovery and analysis of SNPs across the whole genome of two protoplast monokaryons（TPMs） of basidiomycete fungi Lentinula edodes via the high-throughput sequencing *** our knowledge,this is a novel study in which different protoplast monokaryons are utilized for SNPs detection and *** sequence information including 210M and 250M nucleotide bases was generated from TPMs yielding approximately 80%of whole genome *** used surpassed 380K and 410K reads of TPMs for the de novo assembly of two contig sets with an overall estimated depth of 5.67-fold and 6.05-fold coverage,*** of the contigs between TPMs identified 108,235 putative SNPs owning a high probability of at least 10-fold *** SNPs reveal TPMs having similar intra-strain SNP frequencies,and the ratio of transition to transversion among these SNPs being similar to the ratio reported in other *** verify the confidence of detected SNPs,25,044 SNPs were mapped onto 43,912 positions that uniformly distributed over the sequenced chromosomes of Coprinopsis *** result shows high sequence conservation among closely related fungal *** addition,the gene structure of TPMs was constructed for SNPs classification according to their distribution in genome ***,this study indicates the utility of using next generation sequencing technologies to identify large numbers of reliable ***,the method combination of SNP detection and novel algorithm allows an efficient approach for the development of high-density genotyping platforms in fungal species.

Obesity is a medical condition with excess body fat accumulation to the extent which leads to serious health consequences. Abdominal obesity, also known as central obesity, refers to the presence of excess fat in the abdominal area. Obesity, especially abdominal obesity, contributes to many metabolic disorders including metabolic syndrome （MetS）, type 2 diabetes （T2DM） and cardiovascular diseases （CVD）. The incidence of obesity has increased dramaticaUy in recent yeats worldwide. In china, more than one-third of adults are overweight or obese and 10%-20% of all adults are affected by MetS. The pathogenesis underlying the abdominal obesity remains unclear. The ultimate health outcome of obesity and its related metabolic disorders have prompted physicians to take aggressive treatments （lifestyle changes, pharmacological interventions and surgical therapies） before a serious consequence becomes clinically apparent. In this review, we discuss the prevalence, pathogenesis and clinic features of obesity in china.

Recent studies on neuron-restrictive silencer factor(NRSF) / neuronal repressor element-1 silencing transcription factor(REST) have revealed that NRSF participates in the pathological processes of nervous system diseases,such as global ischemia,epilepsy and Huntington *** l-methyl-4-phenyI-pyridinium ion(MPP) treatment triggers the mRNA and protein expression of NRSF in human dopaminergic SH-SY5Y cells,and alteration of NRSF expression exacerbates MPP-induced cell ***,we generated a neuron-specific conditional knockout mice of NRSF(cKO) by crossing NRSF floxed mice with the neuron-specific enolase(NSE)-Cre transgenic *** or wild type mice were injected with l-methyl-4-phenyl-l,2,3,6-tetrahydropyridine(MPTP) at a dose of 15mg/kg,four times within two days,and behavioral assays were performed respectively on 1,3,7,14,21 and 28 days after the last MPTP *** were sacrificed one day after the last behavioral *** levels of DA,its metabolite 3,4-dihydroxiphenylacetic acid (DOPAC),and 5-hydroxytryptamine(5-HT) or non-relevant 5-hydroxyindoleacetic acid(5-HIAA) in striatum were analyzed by ***,the protein level of tyrosine hydroxylase(TH) in striatum was detected by Western blotting *** results showed that in the experiment of swim test,the force swimming time in both cKO and wild type mice was obviously decreased after MPTP administration,but the rate of decline in the cKO mice was higher than that in the wild type *** intoxication reduced the levels of DA,DOPAC and 5-HT in cKO and wild type mice significantly. The ratio of decrease in the cKO mice was more dramatic than that in the wild type *** the same situation happened at the level of TH in *** results suggested that neuron-specific knockout of NRSF mouse is more vulnerable to the dopaminergic neurotoxin MPTP.

Identification of Intergeneric Hybrid Plants Between Oryza sativa and *** via GISH and RAPD

To transfer desirable resistance traits from O. minuta to O. sativa, intergeneric hybrid plants between O. sativa (AA, 2n=2X=24) and O. minuta (BBCC, 2n=4X=48) were produced by embryo rescue after sexual cross. Morphological observation and chromosome counts indicated their hybrid status (ABC, 2n=3X=36). Genomic in situ hybridization (GISH) was further applied to confirm the parentage of the chromosomes of F 1 hybrids. Chromosomes of O. minuta and O. sativa were distinguishable in the hybrids in different fluorescence colors. GISH indicated that A and BC chromosomes were not randomly assembled in a cell. RAPD profiles unequivocally revealed their hybrids with double parent patterns. The results of blast tests showed that the hybrids had obtained disease resistance from O. minuta, and had a level of susceptibility between the parents.

The GJB2 gene(connexin 26) has been shown to be responsible for DFNB1 and DFNA3. We screened the GJB2 gene in 488 patients with prelingual deafness(Group 1), 124 with postlingual deafness(Group 2), and 117 normal hearing subjects(Group 3). We found that, in Group 1, 65 patients(13.32%) were homozygotes or compound heterozygotes and 51 patients (10.45%) carried a single pathogenic mutation. The 235delC mutation was the most frequent mutation, accounting for 73.22% of the known pathogenic alleles in Group 1. No homozygotes or compound heterozygotes were detected in Group 2 or Group 3. Some postlingual deaf patients (2.42%) and normal hearing subjects(4.27%) were 235delC carriers. Our preliminary data indicate that 235delC, the most frequent mutation identified in this study, is a major cause for prelingual deafness.

Background Currently available vaccines against leptospirosis are low efficacy, have an unacceptable side-effect profile, do not induce long-term protection, in addition, provide no cross-protection against the different serovars of pathogenic leptospira. There is a need to screening vaccine candidate genes encoding surface-exposed proteins in the genome of Leptospira interrogans. The reverse vaccinology, which has been successful in the identification of vaccines candidates, can be used to identifying the potential vaccine candidates against leptospirosis. Although several papers tried to identify the surface-exposed proteins of leptospire, but vaccine candidate genes encoding the surface-exposed proteins that can not only provide cross-protection against the different serovars of pathogenic leptospires but also have relatively high transcription level are still awaited. Results Bioinformatics, comparative genome analysis as well as transcriptional analysis were used for the identification of novel vaccine candidates in the genome of Leptospira interrogans strain #56601. There were 686 genes (14.5%, 686/4727) were predicted encoding surface-exposed proteins by P-CLASSIFIER, currently the most accurate predicting system for subcellular localization in Gram-negative bacteria analysis combined with signal peptides prediction, a-helix transmembrane topology prediction, integral β-barrel outer membrane protein and putative lipoproteins prediction. The DNA microarray of L. interrogans strain #56601 has been constructed for comparative genome hybridization (CGH) and transcriptome analysis at 37癈 in vitro. 307 differential genes were identified in ten pathogenic serogroups by CGH. The numbers of high transcriptional level (cy3 signal ≥342 and cy5 signal ≥363.5 respectively) are 1427 genes.
