Rapid accumulation of bacterial genomic data offered an unprecedented opportunity to understand bacterial biology from a holistic view of *** can thus closely look at the way in which a pathogen is evolved,and these data has been applied to molecular epidemiology and microbial forensics,and screening of novel diagnostic,vaccine and drug *** newly developed high-throughput low-cost sequencing technologies,such as 454,Solexa and SOLiD,will promote the acquisition and application of genomic data in new research areas that we dared not imagine previously,such as the metagenomics of human gastric-intestinal tract,for better and comprehensive understanding of human health and disease.

DNA polymerase delta is encoded by the POLD1 gene,the transcription of which is strictly cell ***,the means by which POLD1 transcription is regulated by the cell cycle mechanism is currently *** discovered a novel element in the POLD1 promoter known as a CDE(cell cycle-dependent element)/CHR(cell cycle gene homology region) element.A series of luciferase reporter constructs containing various POLD1 promoter mutations were used to investigate the role of the CDE/CHR element in POLD1 *** the CDE/CHR element was mutated,the promoter activity was up-regulated,and the cell-cycle related factors E2F1 and p21 stopped regulating the ***,cell cycle-dependent changes in the promoter activity required the integrative CDE/CHR *** mobility shift assay(EMSA) revealed the presence of at least three types of DNA/protein complexes binding to the CDE/CHR *** findings provide strong evidence that the CDE/CHR-like sequence is an active functional element in the POLD1 promoter,which is important for the cell cycle regulation of the POLD1 gene.

Secreted Protein HP1286 of Helicobacter pylori Strain 26695 Induces Apoptosis of AGS Cells

Objective Secreted proteins of Helicobacter pylori （H. pylori） interact with gastric epithelium cells and may contribute to cell damage. Considering the fact that HPO17S and hypothetical conserved protein HP1286 are included in the group and that HP0175 is a well-known apoptosis-induced factor, the present study aims to clarify whether HP1286 plays a role in bacterial pathogenicity or even functions as an apoptosis-induced factor in human stomach. Methods Two genes encoding HP1286 and HPO175 were cloned into pET32a vector and expressed as recombinant proteins in Escherichia coil （E. coil） BL21. Signal peptide sequences were not included. The recombinant proteins were purified with His SpinTrap and desalted by using HiTrap Desalting. Immunoreactivity of the proteins was determined by Western blot. Human gastric epithelial cell AGS was challenged with these endotoxin-free proteins; and apoptosis of cells was assayed with the Cell Death ELISA kit. Results Recombinant proteins and their respective products whose N-terminal his-tag were removed with thrombin were recognized by serum from the patient infected with H. pylori. Apoptotic AGS cells challenged by HP1286 of H. pylori 26695 were four times more than untreated cells. In addition, apoptosis-induced ability of HP1286 of SS1 was not as strong as that of H. pylori 26695 strain. Conclusion HP1286 of H. pylori 26695 induces apoptosis of AGS cells in a time-dependent manner, however the apoptosis-induced ability of HP1286 may differ due to variations between different strains.

Plague,caused by the gram-negative bacterium Yersinia pestis,is a serious and rapidly progressing illness in humans that can be fatal if not treated *** Qinghai-Tibet Plateau is the largest area of natural Himalayan marmot(Marmota himalayana) plague foci in china and covers more than 630000 *** County in Gansu Province is a part of this natural focus of plague and was chosen as a study *** study used an ecological niche modeling(ENM) approach to predict the potential distribution of the Himalayan *** and Disaster Monitor Satellite(HJ-1) data was used to investigate environment factors that affect plague host animal *** animal point data from active surveillance was combined with environmental variables from the HJ-1 satellite and other databases,and the models of the potential distribution of Himalayan marmot were produced with the Genetic Algorithm for Rule-Set Production(GARP).The probability of marmot presence was divided into 0-5%,5%-20%,20%-40%,40%-80%,and 80%-100% *** with 80%-100% probability exhibited the greatest potential for the presence of Himalayan *** to the predicted potential distribution of Himalayan marmot in the study area,active surveillance of plague hosts and plague control and prevention could be more efficient.

One of the physiological functions of cellular prion protein(PrP C )is believed to work as a cellular resistance to oxidative stress,in which the octarepeats region within PrP plays an important ***,the detailed mechanism is less *** this study,the expressing plasmids of wild-type PrP (PrP-PG5)and various PrP mutants containing 0(PrP-PG0),9(PrP-PG9)and 12(PrP-PG12)octarepeats were generated and PrP proteins were expressed both in *** and in mammalian *** aggregation and formation of carbonyl groups were clearly seen in the recombinant PrPs expressed from *** after treatment of *** and trypan blue staining assays revealed that the cells expressing the mutated PrPs within octarepeats are less viable than the cells expressing wild-type *** significant high levels of intracellular free radicals and low levels of glutathione peroxidase were observed in the cells transfected with plasmids containing deleted or inserted *** more productions of carbonyl groups were detected in the cells expressing PrPs with deleted and inserted octarepeats after exposing to ***,cells expressing wild-type PrP showed stronger resistant activity to the challenge of H2O2 at certain extent than the mutated PrPs and mock. These data provided the evidences that the octarepeats number within PrP is critical for maintaining its activity of *** of its protective function against oxidative stress may be one of the possible pathways for the mutated PrPs to involve in the pathogenesis of familial Creutzfeldt-Jacob diseases.

Interpretation Criteria for Standardized Western Blot for the Predominant Species of Borrelia Burgdorferi Sensu Lato in china

Epstein-Barr virus (EBV) infection has been causally associated with occurrence of many malignant neoplasms. EBV-encoded small RNAs (EBERs) have been detected from about 10% of gastric carcinoma tissue cells, suggesting that EBV infection is associated with the development of gastric carcinoma. The present study pooled the data from the papers concerning EBV-related gastric cancers and performed a meta-analysis of 22 research papers. Among these papers, a total of 5475 cases with gastric cancer were enrolled, of whom 411 cases were found EBV-positive, with the EBV-positive rate being 7.5%. Among the EBV-positive gastric cancer cases, the detection rate was 11.1% in males and 3.0% in females. Compared with EBV-negative gastric cancer, EBV-positive gastric cancer had less lymph node metastasis. Based on the histological typing, of the EBV-positive gastric cancers, the diffuse type was 8.1%, and intestinal type was 8.0%. The examined specimen types included stored paraffin blocks and fresh surgically removed specimens, their EBV positive rates were 7.9% and 6.5% respectively. In terms of geographical distribution, the detection rate of EBV-positive gastric cancer was 9.4% in America, 6.1% in Asia and 9.1% in Europe. Meta-analysis showed that EBV infection occurred only in gastric cancer tissue cells and was significantly associated with the patients’ gender, lymph node metastases, and the location where tumor tissue generated and geographical distribution (P0.05). These results suggested that EBV-positive gastric cancer has distinct clinicopathological features.

Neisseria meningitidis is a major cause of bacterial meningitis and septicemia worldwide. In china, serogroup A strains were responsible for over 95% of the cases, while serogroup B strains were mainly the cause of localized outbreaks and sporadic cases. Before 2003, serogroup C strains were only re-covered from a few sporadic cases. However, a sudden increase in the number of cases due to sero-group C strains occurred during 2003—2005 in Anhui Province, china. Many cases were found in other provinces at the same time. Multilocus sequence typing (MLST) results indicated that the unique se-quence type 4821 clone meningococci, a new hyper-virulent lineage, was responsible for the serogroup C meningitis outbreaks. We have completed the project of sequencing the whole genome of the Chi-nese N. meningitidis serogroup C representative isolate 053442. We fabricated a whole-genome mi-croarray of N. meningitidis isolate 053442 and analyzed the genome composition differences among 81 serogroup C isolates which were isolated from 14 provinces of china during 1966—2005. The com-parative genomic hybridization (CGH) result shows that the genome compositions of nearly all sero-group C isolates are similar to that of 053442. The products of many absent open reading frames (ORFs) are conserved hypothetical proteins. The results will provide a valuable resource from which one can analyze the genome composition and genetic background of serogroup C meningococci in china.

In china, many JE virus strains of genotype Ⅰ are isolated recent years while strains of genotype Ⅲ are still circulating. In order to understand the immunogenicity of vaccines to these new isolates of genotype Ⅰ, we perform this research. In vaccination-challenge protection test of mice, JE live attenuated vaccine （LAV）（≥234 pfu） and inactivated purified vaccine （IPV）（dilution 1:5） can protect effectively to all the tested strains including genotype Ⅰ strains by i.p. challenge. When challenged by i.c. routes, most of the virus strains can be protected （≥60%） by the LAV, and ≤40% by the IPV. This results indicated that JE vaccines used in china can still protect the prevalent strains in china and the LAV appeared stronger protection than that produced by IPV.
