Based on particle approach and tidal flow model this article studies the behavior of the oil slick on the water surface in the Huangpu River, a tidal waterway in shanghai. In order to track the oil slick motion, a two-dimensional oil trajectory model is used. The dynamical properties of spilled oil characterized by advection, oil spreading and turbulent diffusion are considered in the model. The simulation results consistent with the flume experimental data show that the model is applicable. Both simulation and experiment illustrate that the tidal flow has a great influence on the oil slick motion. The calculated results can be used as a reference for the response to oil spill accidents in rivers.

Gal α(1, 3) Gal (gal epitope) is a carbohydrate epitope and synthesized in large amount by α(1, 3) galactosyltransferase [α(1, 3) GT] enzyme on the cells of lower mammalian animals such as pigs and mice. Human has no gal epitope due to the inactivation of α(1, 3) GT gene but produces a large amount of antibodies (anti-Gal) which recognize Gal α(1, 3) Gal structures specifically. In this study, a replication deficient recombinant adenoviral vector Ad5sGT containing pig α(1, 3) GT cDNA was constructed and characterized. Adenoviral vector-mediated transfer of pig α(1, 3) GT gene into human tumor cells such as malignant melanoma A375, stomach cancer SGC-7901, and lung cancer SPC-A-1 was reported for the first time. Results showed that Gal epitope did not increase the sensitivity of human tumor cells to human complement-mediated lysis, although human complement activation and the binding of human IgG and IgM natural antibodies to human tumor cells were enhanced significantly after Ad5sGT transduction. Appearance of gal epitope on the human tumor cells changed the expression of cell surface carbohydrates reacting with Ulex europaeus I (UEA I) lectins, Vicia villosa agglutinin (VVA), Arachis hypogaea agglutinin (PNA), and Glycine max agglutinin (SBA) to different degrees. In addition, no effect of gal epitope on the growth in vitro of human tumor cells was observed in MTT assay.

Asip is a mammalian homologue of polarity protein Par-3 of Caenorhabditis elegans and Bazooka of Drosophila melanogaster. Asip/Par-3/Bazooka are PDZ-motif containing proteins that localize asymmetrically to the cell periphery and play a pivotal role in cell polarity and asymmetric cell division. In the present study, we have cloned human asip cDNA and its splicing variants by 5’-RACE and RT-PCR using candidate human EST clones which have a high homology to rat asip cDNA. The full-length cDNA of human asip encodes a 1,353 aa protein exhibiting 88% similarity to the rat one. Human asip is a single copy gene consisting of at least 26 exons and localizing in human chromosome 10, band p11.2, with some extraordinarily long introns. All exon/intron boundary nucleotides conform to the "gt-ag" rule. Three main transcripts were detected by Northern blot analysis, and at least five variants, from alternative splicing and polyadenylation, have been identified by RT-PCR and liver cDNA library screening. Exon 17b deleted asip mRNAs expressed ubiquitously in normal human tissues, including liver, on RT-PCR analysis. However, they were absent from most human liver cancer cell lines examined. More interestingly, the expression of exon 1 7b deleted variants was down regulated in 52.6% (10/19) clinic specimens of human hepatocellular carcinomas (HCCs), compared with the surrounding nontumorous liver tissues from the same patients. The presence of various splicing transcripts, the variation of their distribution among different tissues and cells, and their differential expressions in human HCCs suggest that human Asip isoforms may function in different context.

The hmr19 gene was cloned from Streptomyces hygroscopicus subsp. yingchengensis strain 10- 22, a bacterium strain producing agricultural antibiotics. Sequence similarity comparison indicates that hmr19 gene may encode a predicted protein with 14 putative transmembrane α-helical spanners, belonging to the drug:H+ antiporter-2 family of the major facilitator superfamily. The expression of hmr19 in the mycelium of strain 10-22 was detected by Western blotting analysis. Gene replacement technology was employed to construct an hmr19 disruption mutant. The growth inhibition test against different antibiotics indicated that the mutant strain was 5–20 fold more susceptible to tetracycline, vancomycin and mitomycin C than the parental wild type strain. The mutant took up tetracycline much faster and accumulated more antibiotics than the wild type strain 10-22. While with the addition of an energy uncoupler, carbonyl cyanide m- chlorophenylhydrazone, the characteristics of the accumulation of [3H]tetracycline in these two strains were almost the same. It was thus concluded that hmr19 encoded a multidrug resistance efflux protein.

Microstructure and Electromechanical Properties in PMnN-PZT Ceramics Sintered at Different Temperatures

The microstructure and piezoelectric properties of Pb[(Zr0.52Ti0.48)0.95(Mn1/3Nb2/3)0.05]03 (PMnN-PZT) ceramics have been investigated at different sintering temperatures from 1070℃ to 1280℃. The experimental results suggest that grain sizes and electromechanical properties of the ceramics show strong sintering temperature dependences. Double polarization to electric field (P-E) loops of the PMnN-PZT ceramics are assumed to be the result of the pinning effect of the defect dipoles. The results obtained through X-ray photoelectron spectrum suggest that sintering temperature affects valence of the Mn ion and quantity of oxygen vacancies. Hence, a proper explanation is proposed to illustrate the fluctuation of mechanical quality factor (Qm) due to the sintering temperature.

Human group IB phospholipase A2 (IB-PLA2) and its zymogen (proIB-PLA2) were purified from E. coli. Refolding was carried out by diluting the denatured forms of both IB-PLA2 and proIB-PLA2 with renaturation buffer in which the disulfide bonds were completely reduced. The refolding yield of proIB-PLA2 was increased by about 50% over that of the mature enzyme. The refolding of IB-PLA2 usually produced aggregates under normal conditions, as determined by light scattering. In addition, the unfolding experiments showed that the mature enzyme was more stable than the proenzyme toward denaturants in the presence of DTT. Results suggested that the N-terminal sequence rather than its conformation of human proIB-PLA2 played an important role in the refolding process.

The effective reflection area of a cube corner retroreflector is defined. It is testified for a cube corner retroreflector (CCR) that the ray reflected from a CCR is not parallel with the ray incident on the CCR undersurface. The effective reflection area of CCR is calculated when the ray incident on the CCR undersurface vertically, and the effective reflection area of a CCR is two thirds of the CCR undersurface.

A new method of realizing coherent split of atomic wave packet has been presented in this paper. Due to using Doppler effect and stimulated Raman transition, it Can be applied directly in thermal atomic beam and only needs a single-frequency standing wave laser field. Theoretical analyses show that of both atoms and light field meet some conditions, atomic wave packet will be split coherently after interaction. Finally,the experiment of the atomic beam splitter by using this method is described. The results are in good agreement with the theoretical predication.
