Mass Spectrometric Evidence of Heparin Disaccharides for the Catalytic Characterization of A Novel Endolytic Heparinase

Heparinase from different sources can eliminate heparin or/and heparan sulfate into various low-molecular weight heparins with different characteristics. Porcine intestinal mucosa heparin was de- graded into a series of oligosaccharides by a novel heparinase from the species Sphingobacterium. Disaccha- ride components from the digests were separated and purified by ultrafiltration and HPLC. Five major peaks appeared as three types according to their retention time. The mass spectrometry of peak I mainly gave the non-sulfated disaccharide with the mass of 3.9 Da. Peak II and III were indicated as two major monosulfated disaccharides with molecular mass of 417 and 459 Da respectively. Moreover, the peak III represented an N- acetyl disaccharide. Both peak IV and V showed the same mass of 496 Da, hinting that they were disulfate- substituted disaccharides. No trisulfate-substituted disaccharides were detected in the mixture of the heparin digest though they were abundant in the heparin structure. The results revealed that the heparinase might specifically cut the sites with low sulfated domain in heparin.

Ionization state of cosmic hydrogen by early stars and quasars

Cosmic hydrogen is reionized and maintained in its highly ionized state by the ultraviolet emission attributed to an early generation of stars and quasars. The Lyα opacity observed in absorption spectra of high-redshift quasars permits more stringent constraints on the ionization state of cosmic hydrogen. Based on density perturbation and structure formation theory, we develop an analytic model to trace the evolution of the ionization state in the post-overlap epoch of reionization, in which the bias factor is taken into account. For quasars, we represent an improved luminosity function by utilizing a hybrid approach for the halo formation rate that is in reasonable agreement with the published measurements at 2 ≤ z ≤ 6. Comparison with the classic Press-Schechter mass function of dark matter halos, we demonstrate that the biased mass distribution indeed enhances star formation efficiency in the overdense environment by more than 25 per cent following the overlap of ionized bubbles. In addition, an alternative way is introduced to derive robust estimates of the mean free path for ionizing photons. In our model, star-forming galaxies are likely to dominate the ionizing background radiation beyond z = 3. and quasars contribute equally above a redshift of z ～ 2.5. From 5 ≤ z ≤ 6, the lack of evolution in photoionization rate can thus be explained by the relatively flat evolution in star formation efficiency, although the mean free path of ionizing photons increases rapidly. Moreover, in the redshift interval z ～ 2 - 6, the expected mean free path and Gunn-Peterson optical depth obviously evolve by a factor of ～ 500 and ～50 respectively. We find that the relative values of critical overdensities for hydrogen ionization and collapse could be 43.% at z ≈2 and 2% at z ≈6, suggesting a rapid overlap process in the overdense regions around instant quasars following reionization. Chronology of Holocene lacustrine sediments in Co Ngoin, central Tibetan Plateau

Chronology is the basis for using lacustrine sediments to reconstruct the history of en-vironmental change. Radioactive-nuclides such as 14C, 210Pb and 13.Cs dating are mainly used to establish the chronology for recent several ten thousand years. Because of being mixed with “dead carbon”, the measured 14C age is always older than the actual age which is recognized as “reservoir effect”. Cs is a kind of active metal element, and easy to migrate vertically in the sediment that leads to the error of the time marker. 210Pb dating should make sure to select CIC model or CRS model. On the Tibetan Plateau, most of the lakes are alkaline closed lake where Cs is more moveable in the sediment and the reservoir effect caused by “dead carbon” on 14C dating is stronger. Based on the analysis on results of 14C, 210Pb and 13.Cs of the lacustrine sediments from Co Ngoin, central Tibetan Plateau, we use the simple regression method to re-calculate the 14C calendar ages, and establish the time sequence for cores CE-1 and CE-2 with result of 210Pb CRS model for the upper 3. cm and re-calculated 14C calendar age for sediments bellow 3. cm. Since 1400 cal. a BP, sedimentation inter-rupted for more than 1000 years. According to this time sequence, variations of environmental proxies confidently reflect the major climatic events in Holocene.

Interannual variability of Antarctic Oscillation and its influence on East Asian climate during boreal winter and spring

The interannual variability of Antarctic Oscillation (AAO) and its influence on East Asian climate during both boreal winter and spring are addressed. The results show that the positive AAO anomaly decreases the cold activity over East Asia during both boreal winter and spring. AAO-related barotropic meridional teleconnection from Antarctic to Arctic is found through analysis of mean me- ridional circulations. This meridional teleconnection is remarkable over Eurasia during boreal winter and over the Pacific Ocean during boreal spring. The results also show that zonal mean zonal wind at high latitudes in Southern Hemisphere has well positive correlation with that of Eurasia during boreal winter and has negative correlation with Pacific North American teleconnection (PNA) during boreal spring, which again display the meridional teleconnection. Thus, local meridional teleconnection is a possible linkage for interaction of circulations at mid-high latitudes between both hemispheres.

This paper established sustainable land resources evaluating indicators fit for hilly area in Sichuan Basin, Southwest of china, and evaluated the case of sustainable land management of Lezhi County from 1990 to 2001. The result showed that the land resource sustainable utilization ability in Lezhi County is belong to Second level （better sustainable）. Secondly, this paper diagnosed some years obstacles of sustainable land management of Lezhi County from base period to goal period and compared the obstacles factors in the indicators system. There were 16 indexes orderly become major affect factor to land sustainable utilization of Lezhi county in indicator system, of which there are 8 obstacles indexes exist both in 1990 and 2001. The value of obstacles degree of five indicators exceeded 5 in 1990, but in 2001, only two indicators＇ values were more than 5.

Observation results of relativistic electrons detected by Fengyun-1 satellite and analysis of relativistic electron enhancement (REE) events

The space particle component detector on Fengyun-1 satellite which works at the sun-synchronous orbit of about 870 km altitude has detected relativistic electrons for a long time. In comparison with the SAMPEX satellite observations during 1999–2004, the relativistic electron data from Fengyun-1 satellite from June 1999 to 2005 are used to analyze the relativistic electron enhancement (REE) events at the low earth orbit, and the possible correlation among REE events at the low earth orbit, high-speed solar wind and geomagnetic storms is discussed. The statistical result presents that 45 REE events are found in total during this time period, and the strong REE events with the maximum daily average flux > 400 cm?2·srt-1·s?1 occur mostly during the transition period from solar maximum to solar minimum. Among these 45 REE events, four strong REE events last a longer time period from 26-to 51-day and correlate closely with high speed solar wind and strong geomagnetic storms. Meanwhile, several strong geomagnetic storms occur continuously before these REE events, and these continuous geomagnetic storms would be an important factor causing these long-lasting strong REE events. The correlation analysis for overall 45 events indicates that the strength of the REE events correlates with the solar wind speed and the strength of the geomagnetic storm, and the correlation for strong REE events is much stronger than that for weak REE events.

Direct Optimization of Low-thrust Many-revolution Earth-orbit Transfers

Low-thrust Earth-orbit transfers with 10^- 5-order thrust-to-weight ratios involve a large number of orbital revolutions which poses a real challenge to trajectory optimization. This article develops a direct method to optimize minimum-time low-thrust many-revolution Earth-orbit transfers. A parameterized control law in each orbit, in the form of the true optimal control, is proposed, and the time history of the parameters governing the control law is interpolated through a finite number of nodal values. The orbital averaging method is used to significantly reduce the computational workload and the trajectory optimization is conducted based on the orbital averaging dynamics expressed by nonsingular equinoctial elements. Furthermore, Earth＇s shadowing and perturbation effects are taken into account. The optimal transfer problem is thus converted to the parameter optimization problem that can be solved by nonlinear programming. Taking advantage of the mapping between the parameterized control law and the Lyapunov control law, a technique is proposed to acquire good initial guesses for optimization variables, which results in enlarged convergence domain of the direct optimization method. Numerical examples of optimal Earth-orbit transfers are presented.

Identification of novel SINEs from Cyprinidae and their evolutionary significance

Short interspersed nuclear elements (SINEs) are widespread among eukaryotic genomes. They are repetitive DNA sequences that have been amplified by retrotransposition. In this study, a class of SINEs were isolated from the Opsariichthys bidens genome, and named Opsar. Sequence analysis confirmed that Opsar is a new class of typical SINEs derived from tRNA molecules. With the tRNA-derived region of Opsar and through BLASTN search, we further identified Zb-SINEs from the zebrafish genome, which includes two groups: Zb-SINE-A and Zb-SINE-B. The Zb-SINE-A. group comprises subfamilies of -Al--A5, and the Zb-SINE-B group is a dimer of the tRNA -derived region and shares a similar dimeric composition to Alu. Zb-SINEs are composed of three distinct regions: a 5’ end tRNA-derived region, a tRNA-unrelated region and a 3. end AT-rich region. The flanking regions are AT rich. The average length of Zb-SINEs elements is about 3.0 bp. Zb-SINEs account for as much as 0.1 % of the whole zebrafish genome. About 70% of the Zb-SINEs are on chromosomes 11, 18, and 19. These Zb-SINEs were characterized by PCR and dot hybridization. The distribution pattern of Zb-SINEs in genome strongly supports the master genes model. The tRNA-derived regions of Opsar and Zb-SINEs were compared with the tRNAAla gene, and they showed 76% similarity, indicating that Opsar and Zb-SINEs originated from an inactive tRNA sequence or a tRNA -like sequence. In view of the evolutionary status of zebrafish in the Cyprinidae, we deduced that Zb-SINEs were a very old class of interspersed sequences.

Responses of soil respiration in non-growing seasons to environmental factors in a maize agroecosystem,Northeast china

Based on continuous three-year measurements (from 2004 to 2007) of eddy covariance and related environmental factors, environmental controls on variation in soil respiration (Rs) during non-growing season were explored in a maize agroecosystem in Northeast china. Our results indicated that during non-growing seasons, daily Rs was 1.08-4.08 g CO2 m-2 d-1, and the lowest occurred in late November. The average Rs of non-growing season was 456.06 ± 20.01 g CO2 m-2, accounting for 11% of the gross primary production (GPP) of the growing season. Additionally, at monthly scale, the lowest value of Rs appeared in January or February. From the beginning to the end of non-growing season, daily Rs tended to decrease first, and then increase to the highest. There was a significant quadratic curve relationship between Rs and soil temperature at 10 cm depth when soil temperature was more than 0°C (P3.%-70%. When soil water content was more than 0.1 m3.m-3. soil moisture at 10 cm depth was significantly parabolically correlated with Rs (P3.-79%. Meanwhile, the standard error of regression estimation between the values of prediction and observation for Rs could reach 2.7%-11.8%. Rs in non-growing season can account for 22.4% of Rs in growing season, indicating that it plays a critical role in assessing the carbon budget in maize agroecosystem, Northeast china.
