Transcriptional regulation of the neuronal nitric oxide synthase gene(nNOS)is particularlycomplex as 12 distinct transcripts derived from different first exons are expressed in a tissue- and cell-specific *** exon 1f mRNA is relatively highly expressed in nervous system and relies upon exon1f-specific promoter *** conventional and real-time reverse transcription-polymerase chain reaction,we found exon 1f mRNA was the major transcript of the nNOS gene in human neuroblastoma *** analyzed a 1090 bp fragment of 1f promoter by TRANSFAC-TESS and Match softwares andluciferase assay,and found an important positive transcriptional regulation region that contained a putativenuclear factor(NF)-κB binding ***,using electrophoresis mobility shift and chromatinimmunoprecipitation assays,we identified this site to be the NF-κB responsive element,a crucial positiveregulator in the activation of the nNOS 1f *** together,our study identified an NF-κB responsiveelement within nNOS 1f promoter and showed that it plays an important role in the transactivation of nNOS1f mRNA,the major transcript of nNOS in SK-N-SH cells.

Aim: A case-report on adenoma of the posterior urethra. Methods: In 131 cases of adenoma of the posteriorurethra, aged 17-79 (mean: 36.4) years, a detailed medical history was taken and urinalysis, urethroscopy, and pro-static specific antigen (PSA) immunohistochemical staining were performed. They were then treated with transurethralresection (TUR) or transurethral electric coagulation (TUEC). Results: Hemospennia occurred in 51% of the cas-es, hemamria in 38%, blood overflow from the urethral orifice in 6%, and dysuria in 5%. The position of the tumorwas at or around the vemmontanum. The appearance of the tumor was similar to those of a papilla, a villus, a dactylor polyp, or simply an engorgement. The tumor contained glandular alveoli and adeno-epithelial cells. PSA immuno-histochemistry was positive in the cytoplasm and nucleus of the adeno-epithelial cell. One hundred and tweenty-ninecases were cured after TUR or TUEC, while 2 patients recurred and were operated again. Conclusion: Adenoma ofthe posterior urethra is a common cause of hemospennia and hematuria in young men. Urethroscopic examination andbiopsy are the principal diagnostic measures. TUR or TUEC are believed to be the treatment of choice with a short-termrecurrence rate of around 1.5%. (Asian J Androl 2000; 3: 67-70)

BACKGROUND:A growing body of evidence suggests that many tumors are initiated by both epigenetic abnormalities and gene mutations,which promote tumor progression. Epigenetic abnormalities include changes in DNA methylation and in the modification of *** study aimed to assess the status of methylation in the CpG island(CGI)of the tumor necrosis factor receptor superfamily member 10c(TNFRSF10C) with combined bisulfite restriction analysis(COBRA)and to evaluate its role in the progression of pancreatic cancer(PC). METHODS:The methylation status of four PC cell lines was assessed using COBRA and/or bisulfite genomic sequencing (BGS).Changes in methylation and TNFRSF10C expression in PC cell lines before and after treatment with 5-aza-2’-deoxycytidine(5-aza-dC)and/or trichostatin A(TSA)were assessed by BGS and real-time *** in the four cell lines was tested by flow cytometry(FCM)and TUNEL assay. RESULTS:The methylation status of the TNFRSF10C promoter was assessed in PC cells(BxPC-3:68.84±8.71%;CFPAC-1:0; PANC-1:96.77±4.57%;SW1990:54.97±7.33%)with the COBRA assay,which was confirmed by the results of *** treatment with 5-aza-dC and/or TSA,apoptosis was induced in PC cells to different degrees,and the levels of TNFRSF10C transcriptional expression in the PC cell lines(except CFPAC-1) increased markedly after 5-aza-dC treatment. CONCLUSIONS:A high frequency of CGI methylation in the TNFRSF10C promoter results in inactivation of the gene and enhancement of tumor growth in most PC cell lines(except CFPAC-1).Inactivation of TNFRSF10C by CGI hypermethylation can play an important role in PC progression and be potentially useful as a diagnostic marker and a new therapeutic approach for PC.

AIM:To create new diabodies with improved binding activity to antigen of the variable light-variable heavy (VH-VL)oriented single-chain Fv dimers genes(scFv). METHODS:The linker between VH and VL genes was shortened to 3-5 amino acid residues and cloned into the vector *** recombinant plasmids were transformed into TG1 cells and *** positive transformed cells were infected by M13K07 helper phage to form human recombinant phage *** products were identified by SDS-PAGE,Western blotting,size exclusion gel chromatography(SEC),ELISA and immunohistochemistry. RESULTS:Three scFv(scFv-3,scFv-4,scFv-5)were constructed successfully with binding ability to hepato-cellular carcinoma 3.5-6 fold greater than their parental *** single-chain Fv dimer(scFv-5,termed BDM3) with the best binding ability was successfully expressed in Yeast pichlia,as shown by SDS-PAGE and Western *** results suggested the molecular weight of the expressed products was about 61 *** products showed significantly stronger binding to hepatocellular carcinoma cells than scFv,still having 50% binding activity even after 16 h incubation as 37℃.The purified dimers were bound specifically to the tumor antigen of HCC. CONCLUSION:we have generated scFv dimers by shortening a series of linkers to 3-5 amino acid residues in VH-linker-VL orientation,resulting in highly stable and affinity-improved dimeric *** will become an attractive targeting moiety in immunotherapeutic and diagnostic applications for HCC.

AIM: To investigate the number of intestinal immunoglobulin A (IgA+) plasma cells and expression of intestinal IgA in mice with acute liver necrosis. METHODS: A model of acute liver necrosis was established by intraperitoneal injection of galactosamine (GalN) and lipopolysaccharide (LPS). Sixty mice were randomly divided into one of 4 equal groups: normal control, acute liver necrosis, LPS, or GalN. Hematoxylin and eosin staining, immunohistochemistry, and an enzyme-linked immunosorbent assay were employed to assess liver and intestinal injury, count intestinal IgA+ plasma cells, and measure the expression level of IgA and interferon γ (IFN-γ) in the small intestinal mucosa of mice. RESULTS: Injured intestinal mucosa was observed in the acute liver necrosis group but not in the normal, LPS or GalN groups. Compared with the normal group,intestinal IgA+ plasma cells were slightly decreased in the LPS and GalN groups [429 ± 20 per high power f ield (HPF), 406 ± 18/HPF, respectively], whereas they were markedly decreased in the acute liver necrosis group (282 ± 17/HPF vs 495 ± 26/HPF in normal group, P 0.05). CONCLUSION: Intestinal IgA+ plasma cells and IgA expression levels indicating that mucosal immune barrier dysfunction, does exist in acute liver necrosis.

Objective To construct lentivirus vectors carrying alphastatin gene,test its secretion expression in human umbilical vein endothelia cells(HUVECs)and observe its effects on growth,migration and tube formation of *** We constructed recombinant lentivirus vectors of NT4-alphastatin fusion gene containing neurotrophin-4 signal peptide,pro-region sequences and alphastatin,then transfected the recombinant lentivirus vectors into HUVECs to obtain secretory protein alphastatin and test its anti-angiogenic activities in *** Our data showed that recombinant self-inactivating lentivirus vectors of NT4-alphastatin were successfully constructed,and stable NT4-alphastatin transduced HUVECs were capable of sustainably secreting alphastatin which significantly suppressed HUVECs migration and differentiation but not VEGF-induced *** This report represents the first time on the use of lentivirus-based vectors to deliver alphastatin,the endogenous angiogenesis inhibitor,and reveals the potential utility of anti-angiogenic gene therapy with lentivirus vectors for treating cancer.
