Characterization of a novel developmentally retarded mutant (drm1) associated with the autonomous flowering pathway in Arabidopsis

A developmentally retarded mutant (drm1) was identified from ethyl methanesulfonate (EMS)-mutagenized M2.seedsin Columbia (Col-0) genetic background. The drm1 flowers 109 d after sowing, with a whole life cycle of about 160 *** also shows a pleiotropic phenotype, e.g., slow germination and lower germination rate, lower growth rate, curlingleaves and abnormal floral organs. The drm1 mutation was a single recessive nuclear mutation, which was mapped tothe bottom of chromosome 5 and located within a region of 2.-30 kb around MXK3.1. There have been no mutantswith similar phenotypes reported in the literature, suggesting that DRM1 is a novel flowering promoting locus. Thefindings that the drm1 flowered lately under all photoperiod conditions and its late flowering phenotype was significantlyrestored by vernalization treatment suggest that the drm1 is a typical late flowering mutant and most likely associatedwith the autonomous flowering pathway. The conclusion was further confirmed by the revelation that the transcriptlevel of FLC was constantly upregulated in the drm1 at all the developmental phases examined, except for a very earlystage. Moreover, the transcript levels of two other important repressors, EMF and TFL1, were also upregulated in thedrm1, implying that the two repressors, along with FLC, seems to act in parallel pathways in the drm1 to regulateflowering as well as other aspects of floral development in a negatively additive way. This helps to explain why the drm1exhibits a much more severe late-flowering phenotype than most late-flowering mutants reported. It also implies that theDRM1 might act upstream of these repressors.

Subcutaneous injection of bee venom causes long-term neural activation and hypersensitization in the dorsal horn of the spinal cord,which contributes to the development and maintenance of various pain-related *** unique behavioral 'phenotypes' of nociception and hypersensitivity identified in the rodent bee venom test are believed to reflect a complex pathological state of inflammatory pain and might be appropriate to the study of phenotype-based mechanisms of pain and *** this review,the spinal processing of the bee venom-induced different 'phenotypes' of pain and hyperalgesia will be *** accumulative electrophysiological,pharmacological,and behavioral data strongly suggest that different 'phenotypes' of pain and hyperalgesia are mediated by different spinal signaling *** the phenotype-based mechanisms of pain might be useful in development of novel therapeutic drugs against complex clinic pathological pain.

Dear Editor,Histone recognition by reader modules constitutes a major mechanism for epigenetic regulation （Jenuwein and Allis 2.01）. BAHD1 （bromo adjacent homology domain containing protein 1） is a vertebrate-specific nuclear protein （Fig. S1） involved in gene silencing by promoting heterochromatin formation. BAHD1 is characteristic with an N-terminal proline-rich region, a nuclear localization signal motif, and a C-terminal bromo adjacent homology （BAH） domain （Fig. 1A）. Previous study revealed that BAHD1 could act as a scaffold protein and tether diverse heterochromatinassociated factors including HP1, MBD1, SETDB1, HDAC5, and several transcriptional factors to trigger facultative heterochromatin formation （Bierne, Tham et al. 2.09）. Consistent with a ＂repressive＂ role, BAHD1 binds to CpG-rich P3 promoter region of IGF2.（insulin-like growth factor II） then represses IGF2.and IGF2.antisense transcription via the recruitment of MBD1 and HDAC5 （Bierne, Tham et al. 2.09）.

Solvation effects on brain uptakes of isomers of ^(99m)Tc brain imaging agents

Analysis of electrostatic hydration free energies of the isomers of the 99mTc-BAT and 99mTc-DADT complexes is carried out using the computer simulation technique. The results show that not only a correlation exists between the logarithm of the brain uptake and the electrostatic hydration free energy for the isomers of 99mTc-brain radiopharmaceu-ticals, but also a linear relationship exists between the logarithm of the ratio of the brain uptake of the syn isomer to that of the anti one and the difference between the electrostatic hydration free energy of the syn-isomer and that of the anti one. Furthermore, the investigation on the important factors influencing the brain uptakes of 99mTc-radiophar-maceuticals and the reasons of the different biodistribution of the isomers of the 99mTc-complexes is explored at the molecular level. The results may provide a reference for the rational drug design of brain imaging agents.

Preparation and Characterization of Chitosan/Carboxymethylated Konjac Glucomannan Blend Films

In order to improve the mechanical and water swelling properties of the chitosan (CS) film, a series of transparent films were prepared by blending 2.(weight) chitosan acetic acid solution with 1.5%(weight) carboxymethylated konjac glucomannan (CMKGM) aqueous solution according to predetermined ratio and drying at 30°C. The morphological structure, miscibility, thermal stability, mechanical properies, and swelling capacity of the blend films were studied by infrared (IR), X-ray diffraction (XRD), differential thermal analysis (DTA), scanning electron micrograph (SEM), and measurements of the mechanical properties and swelling properties. The results demonstrated that there was strong interaction and good miscibility between CS and CMKGM resulted from intermolecular hydrogen bonding and electrostatic force. The mechanical properties in dry state and wet state, thermostability, and water swelling properties of the blend films were obviously improved. The best values of the tensile strength in the dry and wet state achieved 89 MPa and 4. MPa, respectively, when the CMKGM content was 30%(weight). The CS/ CMKGM blend films provided promising biomedical applications.

AIM: To study the association between APOE polymorphisms and cerebral infarction through a case-control study among the Chinese Han population. METHODS: First-ever cerebral infarction patients (n=2.6) whose ages ranged from 4. to 60 years old were recruited from Department of Neurology, Zhongshan Hospital, Shanghai, and Zhejiang Chinese Traditional Medicine Hospital, Zhejiang, China. Unrelated healthy controls (n=2.1) were selected from the general population in the same area with similar age and sex distribution. APOE was amplified by one-stage PCR using the forward primer: 5'-GGC ACG GCT GTC CAA GGA GCT-3' and reverse primer: 5'-GAT GGC GCTGAG GCC GCG CT-3'. The PCR product was digested directly with 5 U of CfoI and separated by a 2. % polyacryla- mide (acrylamide: bis-acrylamide=2.:1) nondenaturing gel. RESULTS: Both cerebral infarction patient and control groups were in Hardy-Weinberg equilibrium. The allele frequency of APOE*2. APOE*3, and APOE*4.was 4.6 %, 81.9 %, and 13.5 % respectively in the patients with cerebral infarction; 5.7 %, 87.3 %, and 7.0 % respectively in the healthy control group. Compared with APOE3/3 subjects, APOE4.4.carriers had a 2.1-fold risk of cerebral infarction (odds ratio 2.1, 95 % confidence limits 1.3 to 3.4.. The allele frequency of APOE*4.in the cerebral infarction patient group was significantly higher than that in the control group (13.5 % vs 7.0 %; P=0.002.. CONCLUSION: APOE 4.is a risk factor for cerebral infarction among the Chinese Han population.

BACKGROUND:The prognostic role of diabetes mellitus (DM) coexisting with hepatocellular carcinoma (HCC) remains *** clarify its impact on survival in HCC patients after curative treatments,a meta-analysis was *** SOURCES:Eligible studies were identified through multiple search strategies in the databases PubMed (MEDLINE),EMBASE,the Cochrane Library and ACP Journal Club between January 1950 and March *** studies fulfilled the inclusion criteria,and data were aggregated comparing overall survival and recurrence-free survival in HCC patients according to DM ***:The pooled hazard ratios (HRs) estimate for overall survival was 1.34.(95% CI,1.18-1.51;P4. (95% CI,1.00-2.18;P2.66;P=0.016),while HBV-related cases were not significantly different (HR=1.2.;95% CI,0.69-2.4.;P=0.182..Meanwhile,the coexistence of DM impaired overall survival in HCC patients with a small tumor burden (HR=1.63;95% CI,1.2.-2.12.P<0.0001).CONCLUSION:HCC patients with coexisting DM have a shorter survival time and a higher risk for tumor recurrence after curative treatments,while the precise value should be defined in more clinical trials with consistent methodology,especially prospective studies.
