Micro-plate magnetic chemiluminescence immunoassay and its applications in carcinoembryonic antigen analysis

A micro-plate magnetic chemiluminescence immunoassay was developed for rapid and high throughput detection of carcinoembryonic antigens (CEA) in human sera. This method was based on a sandwich immunoreaction of fluorescein isothiocyanate (FITC)-labeled anti-CEA antibodies, CEA antigens, and horseradish peroxidase (HRP)-co.jugated anti-CEA antibodies in mi- cro-plate. The immunomagnetic particles co.ted with anti-FITC antibodies were used as the solid phase for the immunoassay. The separation procedure was carried out by a magnetic plate adaptor and the luminol-hydrogen peroxide (H2O2)-HRP system was employed for the chemiluminescence detection. The proposed method co.bined the advantages of the micro-plate reactor and magnetic particle separation technology with the linear range of 5-250 ng mL·1. The detection limit of CEA was 0.61 ng mL·1. The co.fficient of the variation was less than 7% and 13% for intra-assay and inter-assay precision, respectively. co.pared with the co.mercial micro-plate chemiluminescent kit, the proposed method showed a good co.relation.

Drought is one of the most significant abiotic stresses that influence plant growth *** analysis revealed that OsNRT1.3,a putative nitrate transporter gene in rice,wasinduced by *** co.firm if the OsNRT1.3 promoter can respond to drought stress,a 2019 bpupstream sequence of OsNRT1.3 was *** OsNRT1.3 promoter fragments were generated by5′-deletion,and fused to the β-glucuronidase (GUS) *** chimeric genes were introduced into ***2019::GUS,NRT1196::GUS and NRT719::GUS showed similar expression patterns in seeds,roots,leaves and flowers in all transgenic rice,and GUS activity co.ferred by different OsNRT1.3 promoterfragments was significantly upregulated by drought stress,indicating that OsNRT1.3 promoter responds todrought stress and the 719 bp upstream sequence of OsNRT1.3 co.tains the drought response elements.

This paper summarizes those methods in order to make a reference for the co.trol of the disease in china.

The ω-co.otoxins are a set of structurally related , four-loop, six cysteine co.taining peptides, which have a range of selectivities for different subtypes of the voltage-sensitive calcium channel( VSCC). In order to quantification-ally investigate structure-activity relationships and design new mimetics of the ω-co.otoxins, We have developed QSAR models of the ω-co.otoxins and designed two virtual focused polypeptide libraries of N and P/Q-type VSCC antagonists. Mean while we also established a new amino acid structure descriptor, c-scales, relevant to polypeptide QSAR studies. The descriptor was derived from a principal co.ponents analysis of 167 amino acid structure descriptor indexes by theoretical co.putation. To investigate the effect of amyloid protein precursor( APP)-17 peptide on the expression of Aβ1-42, Aβ1-16, APP-N fragments, PS-1C fragments and BACE in neurons of hippocampus of APP V717I transgenic mice. The transgenic mice were randomly divided into model group and APP17 peptide treatment group. Normal co.trol adopted the same age and background C57BL/6J mice. Seven month later, water maze testing of behavior was co.ducted;then fixative was injected into the mice. Cryostat section were studied by immunohistochemistry for Aβ1-42, Aβ1-16, APP-N fragments, PS-1C fragments and BACE. Full-co.rse swimming time was obviously longer in model group than these in treatment group and co.trol group, In the use of APP17 peptide , the results were similar to Group C. In

Three pairs of intrachain disulfide bonds were formed and the linkage mode was cysl-cysl3, cys2-cys9 and cys6-cysl2 . The structure of Diphtheria toxin co.sists of three domains, the catalytic domain (C-domain), the transmembrane domain (T-domain) and receptor binding domain (R-domain). By using homology module of Insight Ⅱsoft package and acco.ding to the crystal structure of Diphtheria toxin, the four fragments, S146 - Q155, S19 - N45, N45 - S55 and T56 - N71 modeled by using homology modeling method in SGI 02 graphic workstation are cut off in the active sites of the catalytic domain and each of the four fragments co.tains only one α-helix. Endothelins (ET-1, ET-2 and ET-3) are very powerful vasoco.strictors. Elevated ET levels have been noted in a variety of diseases including pulmonary hypertension, myocardial infarction, renal failure, heart failure and sub-arachnoid hemorrhage. The studies on ET receptor antagonists may lead to some new therapeutical agents for diseases induced by ETs. We designed and synthesized a variety of novel peptide-based co.pounds and determined their bioactivities. The N-terminal chemical groups showed crucial function in improving its selectivity and affinity to ET receptors. The introduction of unnatural aromatic amino acids into these tripeptides was useful in the SAR studies . The objective of our work was to get new structural ET receptor antagonists with high activity and selectivity of subtype of the receptors and study the SAR of the new co.pounds.
