Modulatory effect of substance P on GABA-activated currents from rat dorsal root ganglion

AIM: To explore the modulatory effect of substance P (SP) on GABA-activated current of dorsal root ganglion(DRG) neurons in rat. METHODS: The whole-cell patch-clamp technique was used to record SP- and GABA-activated currents in neurons freshly dissociated from rat DRG neurons. Drugs were applied by rapid solutionexchange. RESULTS: Application of SP (2./41, 68.5 %) and GABA (36/41, 88.2.%) could induce concentration-dependent inward current in some cells. SP-(10 mol/L) and GABA (100 mol/L)-activated inward currents were(2.483) pA (n=9) and (1.80.5) nA (n=13), respectively. The majority of GABA-activated current had obviousthree processes, the peak value (Ip), the steady state (Iss) and the desensitization (Id). The desensitization of GABA-activated current was a biphasic process, including fast and slow desensitization. However, pre-application of SP(0.0011 mol/L) could inhibit the GABA-activated inward current which was identified to be GABAA receptor-mediated current. The inhibitory effects were concentration-dependent. The inhibitory effect of SP on the peakvalue of GABA-activated current was more than the steady state of GABA-activated current. The inhibition ofGABA-activated current by SP (0.1 mol/L) was related to the time after application of SP, the inhibition of GABA-activated currents by SP reached the peak at about 4 min (49.8 %7.2.%, n=7, P2.min toget a full recovery. The inhibition of GABA-activated currents by SP was almost completely removed after block-ade of PKC by H-7 with the re-patch clamp. CONCLUSION: Pre-application of SP exerts a more strong inhibitoryeffect on the peak value of GABA-activated current than the steady state of GABA-activated current.

Changes in Autoantibodies against β1-Adrenoceptor and M2.Muscarinic Receptor during Development of Renovascular Hypertension in Rats

In an experimental rat’s renovascular hypertension model, we studied the genesis of anti- cardiac β1-adrenoceptor and M2.muscarinic receptor autoantibodies in relation to the changes in immunological function during the development of renal hypertension. The biological activities of these autoantibodies were also examined. It was shown that after two weeks of operation both the frequency of occurrence and the titre of autoantibodies to cardiac β1-adrenoceptor and M2.muscarinic receptor were significantly increased as compared with the control of pre-treatment. The increased autoantibodies lasted for several weeks and then automatically decreased gradually to the pre-clipping level at 10 weeks. Meanwhile the ratio of CD4 /CD8 + + was also undergone an initial increase followed by gradual recovery and correlated well with the changes in antibody titre. The biological effects of these autoantibodies displayed an “agonistic-like” activities on the beating frequency of cultured neonatal cardiomyocyte. It is suggested that autoimmune mechanisms are involved in the pathogenesis of renal hypertension and the cardiac receptor autoantibodies might be one of the mechanisms leading to cardiac dysfunction.

Prescreening teratogenic potential of chlorinated drinking water disinfection by-products by using Hydra regeneration assay

Practicability of method for the Hydra regeneration assay on the prescreening teratogenic potential of chlorinated drinking water disinfection by products was studied through both the assays of toxicity of adult Hydra (T) and inhibition of the growth of regeneration Hydra (I) by using chloroform, dichloromethane and chloroacetic acid. The results showed that T 50 / I 50 ratios of chloroform and chloroacetic acid were 2. 77 and 6 16 respectively, with teratogenic potential. T 50 / I 50 ratio of dichloromethane was 1.69, with weaker teratogenic potential. These experimental results indicated preliminarily that the Hydra regeneration assay has certainly applied value as a prescreening assay for developmental toxicity.

近年来中国恶性肿瘤发病率逐年升高,但肿瘤领域创新药物的出现使得肿瘤患者的生存期得到了延长,恶性肿瘤逐渐成为一种慢病。50%的活动性恶性肿瘤患者伴有不同程度的肾脏损伤,肾脏损伤可以由肿瘤本身所致,更多的是肿瘤治疗过程中导致的急性或慢性肾脏损伤,临床表现包括急性肾损伤、慢性肾脏病、蛋白尿、电解质紊乱等。目前肿瘤合并慢性肾脏病患者的药物剂量调整等缺乏专业的指导和管理,中华医学会临床药学分会联合全国多学科专家制定肿瘤合并慢性肾脏病患者抗肿瘤治疗药物用药指导中国专家共识(2.2.版),共识基于临床循证证据,采用德尔菲法和专家访谈及研讨,涵盖肿瘤合并慢性肾脏病患者的评估方法、慢性肾脏病患者的抗肿瘤药物用药指导方案。同时按照化学治疗药物、小分子靶向药物、抗体类药物等分类,对抗肿瘤药物的选择及剂量调整推荐意见,为临床医师提供切实可行的科学指导。

Human placenta-derived stem cells (hPDSCs) were isolated by trypsinization and further induced into cartilage cells in *** engineered cartilage was constructed by combining hPDSCs with collagen sponge and the cartilage formation was observed by implantation into nude *** showed that hPDSCs featured mesenchymal stem cells and maintained proliferation in vitro for over 30 passages while remaining *** results indicated that hPDSCs have the potential to differentiate into functional cartilage cells in vitro when combined with collagen sponge,which provided experimental evidence for prospective clinical application.

Dynamic behavior of fragile X full mutations in cultured female fetal fibroblasts

AIM: Toassess mitotic stability ofthe fragile X fullmutations andits relationshipwith DNA methylation. METHODS: The length change of the expanded CGG repeats was examined and correlated it with the methylation status in the DNA samplesisolated fromthe fibroblasts derived from a fragile Xfemale fetus anda fragileX maleadult, respectively. RESULTS: A dramatic instability of the expanded CGG repeats in the female fetal fibroblasts was observed. South- ern blot analysis revealed that the 6.9-kb major expanded band detected in passage 2.was completely replaced by a 7.7-kb band after passage 30. Fibroblast clones derived from the passage 3 displayed an unstable expansion of the CGG repeat during clonal proliferation, while methylation status of the CGG repeat region was maintained. In contrast, in fragile X male fibroblasts the expanded CGG repeats were stable during clonal proliferation. CONCLUSION: The mitotic instability of expanded CGG repeat is not always restricted in early development window as proposed previously and other elements rather than DNA methylation could affect the stability of the expanded CGG repeats in fragile X female fetal fibroblast cells.

Objective To investigate the histological changes of rapid tooth movement in dogs treated by resistance reduction and distraction osteogenesis,aiming to establish an animal model and further to reveal the remodeling mechanism of rapid tooth movement. Methods A total of 8 local hybrid dogs were selected as subjects for this study. The second pre-molar was extracted on both sides. The experimental side underwent alvelor surgery for resistance reduction and a home-made tooth-borne intraoral distraction device was installed for rapid tooth movement,while for the other side (control side) only tooth-borne intraoral distraction device was used for rapid tooth movement. The longest active force-delivery span was 2.weeks,followed by 6-week retention. The distance between the moved tooth and anchor unit was recorded weekly,and radiography was performed for each side before and after distraction. The surrounding tissues including periodontal ligament and alveolar bone were sectioned for histological analysis. Results The average distance of tooth movement was 3.55mm on the experimental side and 1.11mm on the control side. The rate of tooth movement was notably higher (P<0.01) and no significant apical root resorption was detected by X-ray on the experimental side. The active alvelor bone remodeling was found on the tension and pressure sides. However,there was no significant difference between the experimental side and the control side after the retention period. Conclusion The rate of orthodontic tooth movement can be accelerated through resistance reduction and periodontal distraction without any unfavorable effects but at minimal anchorage loss.

AIM:To investigate the association between the programmed death-1(PD-1) polymorphisms and genetic susceptibility of chronic hepatitis B virus(HBV) infection in Chinese ***:Two single nucleotide polymorphisms(SNPs),PD-1.1 G > A and PD-1.2.G > A,were genotyped in 539 patients with chronic HBV infection and 353 other family members(HbsAg-) from 2.6 nuclear families using polymerase chain reactiorestriction fragment length polymorphisms *** associations between PD-1 polymorphisms and genetic susceptibilityof chronic HBV infection were analyzed usng the familybased association analysis ***:No association or linkage was detected among 539 ***(single-marker) familybased association tests demonstrated that PD-1 genotypes,alleles and transmitted haplotypes are not associated with chronic HBV infection(all with P value more than 0.05).Transmission/disequilibrium test and sibship disequilibrium test analysis showed no excess of the alleles from heterozygous parents to affected offspring(P = 0.688880,P = 1.000000 respectively).CONCLUSION:The data demonstrated that PD-1.1 and PD-1.2.polymorphisms are not associated with chronic HBV infection in Chinese patients.

The development of functional magnetic resonance imaging (fMRI) technology has made it possible to carry out functional brain imaging experiments in small animals. Usually, group data is required to form the assessment of population, which can not only increase the sensitivity of the overall experiment, but also allow the generalization of the conclusion to the whole population. In order to average the signals of functional brain images from different subjects, it is necessary to put all the mapping images into the same standard space (template image). However, up to now, most animal brain templates remain unavailable and it must be done by ourselves. In this study, a template image based on the brains of eight male Wistar rats is obtained, and it is successfully used in our present Alzheimer disease (AD)-like rat model studies as template for spatially normalizing images to the same stereotaxical space. The fMRI results processed with statistical parametric mapping (SPM99) software are in agreement with the results from immunohistochemical experiment, which proves that this method is universally applicable to the pathologic models of other small animals and to human brain lesion studies.
