Recent studies on neuron-restrictive silencer factor(NRSF) / neuronal repressor element-1 silencing transcription factor(ReST) have revealed that NRSF participates in the pathological processes of nervous system diseases,such as global ischemia,epilepsy and Huntington *** l-methyl-4-phenyI-pyridinium ion(MPP) treatment triggers the mRNA and protein expression of NRSF in human dopaminergic SH-SY5Y cells,and alteration of NRSF expression exacerbates MPP-induced cell ***,we generated a neuron-specific conditional knockout mice of NRSF(cKO) by crossing NRSF floxed mice with the neuron-specific enolase(NSe)-Cre transgenic *** or wild type mice were injected with l-methyl-4-phenyl-l,2,3,6-tetrahydropyridine(MPTP) at a dose of 15mg/kg,four times within two days,and behavioral assays were performed respectively on 1,3,7,14,21 and 28 days after the last MPTP *** were sacrificed one day after the last behavioral *** levels of DA,its metabolite 3,4-dihydroxiphenylacetic acid (DOPAC),and 5-hydroxytryptamine(5-HT) or non-relevant 5-hydroxyindoleacetic acid(5-HIAA) in striatum were analyzed by ***,the protein level of tyrosine hydroxylase(TH) in striatum was detected by Western blotting *** results showed that in the experiment of swim test,the force swimming time in both cKO and wild type mice was obviously decreased after MPTP administration,but the rate of decline in the cKO mice was higher than that in the wild type *** intoxication reduced the levels of DA,DOPAC and 5-HT in cKO and wild type mice significantly. The ratio of decrease in the cKO mice was more dramatic than that in the wild type *** the same situation happened at the level of TH in *** results suggested that neuron-specific knockout of NRSF mouse is more vulnerable to the dopaminergic neurotoxin MPTP.

Variational iteration solving method for el Nio phenomenon atmospheric physics of nonlinear model

A class of el Nino atmospheric physics oscillation model is considered. The el Nino atmospheric physics oscillation is an abnormal phenomenon involved in the tropical Pacific ocean-atmosphere interactions. The conceptual oscillator model should consider the variations of both the eastern and western Pacific anomaly patterns. An el Nino atmospheric physics model is proposed using a method for the variational iteration theory. Using the variational iteration method, the approximate expansions of the solution of corresponding problem are constructed. That is, firstly, introducing a set of functional and accounting their variationals, the Lagrange multiplicators are counted, and then the variational iteration is defined, finally, the approximate solution is obtained. From approximate expansions of the solution, the zonal sea surface temperature anomaly in the equatorial eastern Pacific and the thermocline depth anomaly of the sea-air oscillation for el Nino atmospheric physics model can be analyzed. el Nino is a very complicated natural phenomenon. Hence basic models need to be reduced for the sea-air oscillator and are solved. The variational iteration is a simple and valid approximate method.

The completely embryonic stem(eS)cell-derived mice(eS mice)produced by tetrapioid embryo complementation provide us with a rapid and powerful approach for functional genome ***,inbred eS cell lines often fail to generate eS *** genome of mouse eS cells is extremely unstable during in vitro culture and passage, and the expression of the imprinted genes is most likely to be *** the eS mice retain or repair the abnormalities of the donor eS cells has still to be *** we report that the inbred eS mice were efficiently produced with the inbred eS cell line(SCR012).The eS fetuses grew more slowly before day 17.5 after mating,but had an excessive growth from day 17.5 to *** imprinted genes we examined(H19,Igf2,Igf2r,Peg1,Peg3)were expressed abnormally in eS *** remarkable,the expression of H19 was dramatically repressed in the eS fetuses through the embryo developmental stage,and this repression was associated with abnormal biailelic methylation of the 1119 upstream *** altered methylation pattern of H19 was further demonstrated to have arisen in the donor eS" cells and persisted on in vivo differentiation to the fetal *** results indicate that the eS fetuses did retain the epigenetic alterations in imprinted genes from the donor eS cells.

Smad4 is the central intracellular mediator of transforming growth factor-β(TGF-β) signals. To study the role of Smad4 in skeletal development, we introduced a conditional mutation of the gene in chondrocytes using Cre-loxP system. We showed that Smad4 was expressed strongly in prehypertrophic and hypertrophic chondrocytes. The abrogation of Smad4 in chondrocytes resulted in dwarfism with a severely disorganized growth plate characterized by expanded resting zone of chondrocytes, reduced chondrocyte proliferation, accelerated hypertrophic differentiation and ectopic bone collars in perichondrium. Meanwhile, Smad4 mutant mice exhibited decreased expression of molecules in Indian hedgehog/parathyroid hormone-related protein (Ihh/PTHrP) signaling. The cultured mutant metatarsal bones failed to response to both bone morphogenetic protein-2 (BMP-2) and TGF-β1, while the hypertrophic differentiation was largely inhibited by Sonic hedgehog (Shh). These indicated that Ihh/PTHrP inhibited the hypertrophic differentiation of chondrocytes independent of the Smad4 mediated BMP and TGF-P signals. All these data provided the first genetic evidence demonstrating that Smad4 mediated TGF-P signals inhibit the chondrocyte hypertrophic differentiation, and are required for maintaining the normal organization of chondrocytes in the growth plate.

The acid-sensing ion channel(ASIC) has a prominent role in tissue pH-sensing and neuronal signalling.
