Nucleic Acid Binding activity of Pns6 Encoded by Genome Segment 6 of Rice Ragged Stunt Oryzavirus

We report two sympathetic solar eruptions including a partial and a full flux rope eruption in a quadrupolar magnetic region where a large and a small filament resided above the middle and the east neutral lines,*** large filament first rose slowly at a speed of 8 km s-1 for 2. minutes;it then accelerated to 102.km ***,this filament erupted successfully and caused a coronal mass *** the slow rising phase,various evidence for breakout-like external reconnection has been identified at high and low temperature *** eruption of the small filament started around the end of the large filament’s slow *** filament erupted partially,and no associated coronal mass ejection could be *** on a potential field extrapolation,we find that the topology of the three-dimensional coronal field above the source region is composed of three low-lying lobes and a large overlying flux system,and a null point located between the middle lobe and the overlying antiparallel flux *** propose a possible mechanism within the framework of the magnetic breakout model to interpret the sympathetic filament eruptions,in which the magnetic implosion mechanism is thought to be a possible link between the sympathetic eruptions,and the external reconnection at the null point transfers field lines from the middle lobe to the lateral lobes and thereby leads to the full(partial) eruption of the observed large(small) *** possible mechanisms are also discussed *** conclude that the structural properties of coronal fields are important for producing sympathetic eruptions.

The ORF of genome segment 6 (S6) of rice ragged stunt oryzavirus (RRSV) Philippines isolate was cloned and sequenced based on the S6 sequence of the Thailand isolate. Pns6, the 71 kD product of S6 expressed in E. coli, was demonstrated to be a viral non-structural protein of RRSV by Western blotting. The gel mobility shift assays showed that Pns6 had nucleic acid binding activity. Pns6 could interact with single- and double-stranded forms of DNA and RNA, showing a preference for single-stranded nucleic acid and a slight preference for RRSV ssRNA over the rice ssRNA, as demonstrated by both competition and displacement assays. The binding of Pns6 to nucleic acids is strong and sequence non-specific. By using five truncated derivatives of Pns6, it was found that the basic region from amino acid 2.1 to 2.3 of Pns6 was the unique nucleic acid binding domain. Subcellular fractionation of leaf tissues of RRSV-infected rice plants and subsequent Western blotting had shown that Pns6 accumulated predominately in the cytoplasmic membrane fraction. The possible role of RRSV Pns6 in virus replication and assembly is discussed.

A large volume of bacterially produced precipitates are generated during the bio-treatment of heavy metal *** composition of the bacterially produced precipitates and its effects on sulfate reducing bacteria (SRB) in copper-containing waste stream were evaluated in this *** elemental composition of the microbial precipitate was studied using electrodispersive X-ray spectroscopy (EDX),and it was found that the ratio of S:Cu was *** with the results of copper distribution in the SRB metabolism culture,which was analyzed by the sequential extraction procedure,copper in the precipitates was determined as covellite (CuS).The bacterially produced precipitates caused a decrease of the sulfate reduction rate,and the more precipitates were generated,the lower the sulfate reduction rate *** particle sizes of bacterially generated covellite were ranging from 0.03 to 2.m by particles size distribution (PSD) analysis,which was smaller than that of the SRB *** electron microscopy (TEM) analysis showed that the microbial covellite was deposited on the surface of the *** effects of the microbial precipitate on SRB metabolism were found to be weakened by increasing the precipitation time and adding microbial polymeric substances in later *** results provided direct evidence that the SRB activity was inhibited by the bacterially produced covellite,which enveloped the bacterium and thus affected the metabolism of SRB on mass transfer.

From a random insertion mutant library of Synechocystis sp. PCC 6803,a mutant defective in photoautotrophic growth was obtained. The interrupted gene was identified to be slr2.94 (fbp1),which encodes the fructose-1,6-biphosphatase (FBPase)/sedoheptu-lose-1,7-biphosphatase (SBPase) bifunctional enzyme (F-I). Two other independently constructed slr2.94 mutants showed an identical phenotype. The FBPase activity was found to be virtually lacking in an slr2.94 mutant,which was sensitive to light under mixotrophic growth conditions. These results indicate that slr2.94 is the only active FBPase-encoding gene in this cyanobacterium. Inactivation of photosystem Ⅱ by interrupting psbB in slr2.94 mutant alleviated the sensitiveness to light. This report provides the direct genetic evi-dence for the essential role of F-I in the photosynthesis of Synechocystis sp. PCC 6803.

The aryldiimine NCN-pincer stabilized neodymium dichloride combined with aluminum alkyls established a new type of homogeneous binary neutral Ziegler-Natta catalyst *** system exhibited high activity and high cis-1,4 selectivity for the polymerization of isoprene (Tp=2. °C,98.2.;Tp=-2. °C,> 99%).Such catalytic performances remained under a broad range of polymerization temperatures and monomer-to-neodymium ratios (from 500 to 8000),reaching high number-average molecular weight (Mn=1582.kg/mol) and relatively narrow molecular weight distribution (PDI=1.68),which was,however,influenced by the amount and bulkiness of aluminum *** investigation of the polymerization was performed,which showed the number-average molecular weight of the resultant polyisoprene had an almost linear correlation with the conversion,suggesting,in some degree,the polymerization with this catalytic system was controllable.

Environmental archaeology of archaeological sites on both sides of the Dachang section in the Daning River,the Three Gorges of the Yangtze River,shows that riverbed evolution has a great influence on the distribution of archaeological sites during the prehistorical and historical period,and it restricts human activity and the development of ancient *** investigation,dating data,paleocurrent analysis of 100 gravels on the riverbed and archaeological excavation reveal that the riverbed of the Dachang section developed in the Shuangyantang region during the early stage,after about 30 kaBP it shifted westward gradually and reached the present *** with the westward shift,the focus of ancient culture changed for several *** the Western Zhou Dynasty's period,the east riverbank of the Daning River was the main residential *** the Han Dynasty,along with the westward shift,the living space of ancient people was no longer confined to the east riverbank and the focus of culture moved *** the Zhangjiawan site became the main dwelling *** the late Han Dynasty, the villages were flooded and the ancients had to immigrate to a high *** of the Han and the Jin Dynasties outcropped on the flood plain of the east bank because their surface sediments were eroded by several flood events and the westward shift of the riverbed.

An NUV-optical diagram composed from sources from the second Galaxy Evolution Explorer（GALEX） Ultraviolet Variability（GUVV-2. Catalog provides us with a method to tentatively classify the unknown GUVV-2.sources by their NUV-optical *** the purpose of testing the correctness and generality of the method,we carry out a program on the spectroscopic observations of the unidentified GUVV-2.*** spectroscopic identification of these 37 sources are 19 type-A to-F stars,10 type-G to-K stars and 7 M dwarf stars together with an *** also present the light curves in the R-band for two RR Lyrae star candidates selected from the NUV-optical diagram,both of which undergo cyclic *** their light curves and colors,we classify them as RR Lyrae *** confirm the re-sults,we show a color-color diagram for the 37 newly identified spectroscopic objects compared with previously identified ones,which are consistent with our previous re-sults,indicating that the ultraviolet variable sources can initially be classified by their NUV/optical color-color diagram.
