The new numerical algorithms in SUPER/CESE and their applications in explosion mechanics are studied. The researched al-gorithms and models include an improved CE/SE (space-time Conservation Element and Solution Element) method,a local hybrid particle level set method,three chemical reaction models and a two-fluid model. Problems of shock wave reflection over wedges,explosive welding,cellular structure of gaseous detonations and two-phase detonations in the gas-droplet system are simulated by using the above-mentioned algorithms and models. The numerical results reveal that the adopted algorithms have many advantages such as high numerical accuracy,wide application field and good compatibility. The numerical algo-rithms presented in this paper may be applied to the numerical research of explosion mechanics.

To isolate the over-expressed genes in human renal cell carcinoma (RCC) and analyze its molecular basis of carcinogenesis, we used the mRNA from human RCC tissues as tester and that from the matched normal kidney tissues as driver to construct the suppression subtractive hybridization library. 3.9 of the subtracted clones were arrayed onto a nylon mem-brane and the over-expressed genes were then screened by hybridizing the filter with radioac-tively labeled cDNA from RCC and matched normal kidney tissues. 67 clones over-expressed in RCC by a factor of 6 or more were sequenced and its identities were analyzed in GenBank da-tabase. 4 clones were previously unknown fragments and 2 clones represent KIAA genes. The rest clones were the known genes and some of them were RCC-related, including vascular en-dothelial growth factor, vimentin and tissue factor. Most of the known genes were the RCC-related genes previously unknown, including zinc ribbon domain-containing 1 protein (ZNRD1), pituitary tumor transforming gene1 (PTTG1). Northern blot and semi-quantitative RT-PCR confirmed that the mRNA levels of the 3.novel fragments and 1 KIAA and 3.known genes were significantly higher in RCC than in the matched normal kidney tissues. Immunohis-tochemical and Western blot analysis for PTTG1 and ZNRD1 revealed increased protein level in RCC. The over-expressed genes in RCC are the potential molecular targets for diagnosis and therapy and it is very important to understand the molecular mechanism of RCC through the profile of over-expressed genes.

Ectopic ATP synthase in endothelial cells:a novel cardiovascular therapeutic target

Emerging evidence has revealed potential implications of peroxisome proliferator-activated receptor-δ(PPAR-δ) in lipid homeostasis,insulin resistance and cardiovascular ***,its pathophysiological roles and the underlying mechanisms remain to be *** functional genomic approach including gain- and loss-of-function studies,we have examined the effects of PPAR-8 on the expression of genes involved in the lipid biosynthesis in hepatocytes and the in vascular endothelial activation,major pathophysiological processes associated with metabolic *** results have demonstrated that,in hepatocytes,PPAR-δagonist as well as overexpression of PPAR-δinduced the gene expression of Insig-1,a regulatory endoplasmic reticulum protein braking SREBP activation. PPAR-δinhibited the proteolytic processing of the SREBP-1 into the mature active form,thereby suppressing the expression of the lipogenic genes such as fatty acid synthase,stearyl CoA desaturase-I and glycerol-3.phosphate *** effect is achieved by the induction of Insig-1,a direct target gene for PPAR-δFurthermore,overexpression of PPAR-δinduced Insig-1 expression and suppressed SREBP-1 activation,hence,limiting hepatic steatosis in obese diabetic *** vascular endothelial ceils (ECs),PPAR-δligands significantly inhibited TNF-α-induced expression of vascular cell adhesion molecule-1 and E-selectin as well as the ensuing endothelial-leukocyte ***-δligand upregulated the expression of antioxidant genes and decreased reactive oxygen species production in ECs. In hyperlink analysis,most researchers focus on interpreting hyperlink counts,they seldom concern themselves enough about the significance of spatial distribution of *** this paper,we argue that not only can the inlink count to a tourist attraction’s official website reflect its attractiveness,but also the spatial distribution of its inlinks can reflect the distribution of its tourist source *** mountains in Anhui Province,china,are chosen for the case ***,the correlation between the spatial distributions of an attraction’s tourist source markets and its official website’s inlinks is ***,the tourism indexes such as the spatial concentration index(SCI)and the attraction radius(AR),which are calculated by inlinks,are compared to the real indexes based on tourist ***,the detailed discussion and analysis are presented and an improved model,which is used to calculate the distribution of inlinks,is proposed.

Aim:The aim of the present study was to determine the effect of 5-aminoimidazole-4-carboxamide-ribonucleoside （AICAR） on proliferation,cell cycle,and apoptosisin the human epithelial cervical cancer cell line CaSki ***:Cell countand 3.（4,5-dimethylthiazol-2-yl）-2,5-diphenyltetrazolium bromide assay were usedto determine cell proliferation and *** 3.258 staining was con-ducted to distinguish the apoptotic *** cycle and Annexin-V/propidiumiodide staining were analyzed by fluorescence-activated cell sorting （FACS）.AWestern blot assay was used to evaluate the expression of AKT （also known asprotein kinase B）,mammalian target of rapamycin （mTOR）,p53.and extracellularsignal-regulated kinase （ERK）.Results:AICAR （500 μmol/L） significantly inhibi-ted the proliferation of CaSki cells treated for 24,48,and 72 h as determined by *** cells at the G1and G2phases were dramatically decreased while cells atthe S phase were increased in response to AICAR treatment for 24,48,and 72 *** MTT assay showed less viable cells and Hoechst fluorescent staining showedmore apoptotic cells upon AICAR *** results of the Annexin-Vstaining demonstrated a time-dependent increase of apoptosis in cells treatedwith AICAR for 24,3.,and 48 ***,AICAR activated caspase-3.in atime-dependent *** was also found that AICAR inhibited the phosphory-lation of AKT and mTOR,which are important kinases regulating cell growth *** stimulation obviously increased the expression of the tumorsuppressor p53.and the phosphorylation of ***:AICAR inhibitedproliferation and induced S phase arrest and promoted apoptosis in CaSki cells,which might be mediated by the downregulation of the AKT/mTOR pathway andthe upregulation of the p53.ERK pathway.

AIM: Atherosclerosis (AS) is one of the major threatens of the health and life of human being. Accumulating data have supported the idea that the pathogenesis of AS includes a set of inflammatory and immune responses. The migration, proliferation and apoptosis of smooth muscle cells (SMC) also play an important role. Calcitonin gene-related peptide (CGRP) is a neuropeptide with potent vasodilatory effect. Aim: To investigate the changes in plasma level of the chemokine RANTES (regulated upon activation, normal T cells expressed and secreted) and the responsiveness of lipopolysaccharide (LPS)-induced RANTES secretion from monocytes in patients with hyperhomocysteinemia (HHcy). Methods: The plasma levels of homocysteine (Hcy), folate, and RANTES were measured in 3. control patients with normal Hcy levels and 40 patients with HHcy and the mRNA synthesis of RANTES in isolated human monocytes was determined by RNase protection assays. Results: The plasma level of RANTES was elevated in HHcy patients compared with controls (median 5.3.vs 3.5 ng/mL, P<0.01). LPS-induced RANTES production from monocytes of HHcy patients was also increased significantly. In addition, Hcy directly increased the mRNA level of RANTES in isolated normal human monocytes in a time- and dose-dependent manner. Conclusion: Upregulated RANTES from monocytes in HHcy patients may be involved in the atherogenesis of HHcy-induced atherosclerosis.
