Neuroprotective effect of ONO-1078, a leukotriene receptor antagonist, on transient global cerebral ischemia in rats

AIM: To determine whether ONO-1078 {pranlukast, 4-oxo-8-[p-(4-phenylbutyloxy)benzoyl-amono]-2-(tetrazol-5-yl)-4H-1-benzopyran hemihydrate}, a potent leukotriene receptor antagonist, possesses a neuroprotective effect on global cerebral ischemia in rats, and to explore its possible mechanism of action. METHODS: Transient global cerebral ischemia was induced by four-vessel occlusion for 10 min and followed by 72-h reperfusion. ONO-1078 (0.03-0.3 mg/kg) and edaravone (MCI-186, 3-methyl-1-phenyl-2-pyrazolin-5-one, a neuroprotective agent) 10 mg/kg were ip injected 30 min before ischemia and 1 h after reperfusion, and once a day afterward. Neurological outcome was evaluated before ischemia and 24, 48, 72 h after reperfusion. Neuron density, the expressions of N-methyl-Daspartate (NMDA) receptor subunit proteins (NR1, NR2A, NA2B) and vascular cell adhesion molecule 1(VCAM-1) in the cerebral cortex and hippocampus were measured at 72 h after reperfusion. Aim:To determine whether the cysteinyl leukotriene receptor 1（CysLT1receptor）modulates brain cryoinjury and whether the CysLT1receptor antagonist pranlukastexerts a time-dependent protective effect on cryoinjury in mice.Methods:Braincryoinjury was induced by applying a liquid nitrogen-cooled metal probe to thesurface of the skull for 30 s.Brain lesion,neuron density,and endogenous IgGexudation were observed 24 h after cryoinjury.Transcription and the expressionof the CysLT1receptor were detected by RT-PCR and immunoblotting,and thelocalization of the receptor protein by double immunofluorescence.Results:ThemRNA and protein expressions of the CysLT1receptor were upregulated in thebrain 6-24 h after cryoinjury,and the CysLT1receptor protein was primarily local-ized in the neurons,not in the astrocytes or microglia.Pre-injury treatments withmulti-doses and a single dose of pranlukast （0.1 mg/kg） attenuated cryoinjury;postinjury single dose （0.1 mg/kg） at 30 min （not 1 h） after cryoinjury was alsoeffective.Conclusion:The CysLT1receptor modulates cryoinjury in mice at leastpartly,and postinjury treatment with its antagonist pranlukast exerts the protec-tive effect with a therapeutic window of 30 min.

We investigate the effect of photons and phonons on Frenkel exciton spectrum. The Hamilto nian for a coupled system of the Frenkel excitons. the photons and the phonons is derived. The phonon-exciton interaction makes the energy band of the Frenkel excitons narrow. The photons and the Frenkel excitons make up Frenkel exciton polaritons which are quasi-particles. The Frenkel exciton polariton spectrum consists of two branches. It is found that the photon-exciton interaction makes the ground energy of the Frenkel excitons increase.

Let {W(t): t≥0} be a standard Wiener process and S be the Strassen set of functions. We investigate the exact rates of convergence to zero (as T→∞) of the variables sup_(0≤≤T-a_T inf_(f∈S sup_(0≤r≤1 |Y_t, T(x)-f(x)] and inf_(0≤t≤T-a_T sup_(0≤x≤1|Y_(t.T)(x)-f(x)| for any given f∈S, where Y_(t.T)(x)=(W(t+xa_T)-W(t))(2a_T(logTa_T^(-1)+log logT))^(-1/2). We establish a relation between how small the increments are and the functional limit results of Csrg-Revesz increments for a Wiener process. Similar results for partial sums of i.i.d, random variables are also given.

Multimedia content analysis and management are a promising and challenging theme.In this paper we develop a novel approach to image representation,which we call group sparse representation (GSR),for image classification and video retrieval.The basic idea is to represent a test image as a weighted combination of all the training images.In particular,we introduce two sets of weight coefficients,one for each training image and the other for each class.Moreover,we formulate our concern as a group nonnegative garrote model.The resulting representations are sparse,and they are appropriate for discriminant analysis.Experiments on Caltech101 and PASCAL VOC2008 image dataset and TRECVID2005 video corpus testify that our proposed approach is efficient and effective.
