Plasmacytoid dendritic cells (pdCs) represent a unique and crucial immune cell population capable of producing large amounts of type I interferons (IFNs) in response to viral *** function of pdCs as the professional type I IFN-producing cells is linked to their selective expression of Toll-like receptor 7 (TLR7) and TLR9,which sense viral nucleic acids within the endosomal *** I IFNs produced by pdCs not only directly inhibit viral replication but also play an essential role in linking the innate and adaptive immune *** aberrant activation of pdCs by self nucleic acids through TLR signaling and the ongoing production of type I IFNs do occur in some autoimmune ***,pdC may serve as an attractive target for therapeutic manipulations of the immune system to treat viral infectious diseases and autoimmune diseases.

The origin of cytoskeleton and the origin of relevant intracellular transportation system are big problems for understanding the emergence of eukaryotic cells. The present article summarized relevant information of evidences and molecular traces on the origin of actin, tubulin, the chaperonin system for folding them, myosins, kinesins, axonemal dyneins and cytoplasmic dyneins. On this basis the authors proposed a series of works, which should be done in the future, and indicated the ways for reaching the targets. These targets are mainly: 1) the reconstruction of evolutionary path from mreB protein of archaeal ancestor of eukaryotic cells to typical actin; 2) the finding of the mreB or mreB-related proteins in crenarchaea and using them to examine J. A. Lake’s hypothesis on the origin of eukaryote from 'eocytes' (crenarchaea); 3) the examinations of the existence and distribution of cytoskeleton made of mreB-related protein within coccoid archaea, especially in amoeboid archaeon Thermoplasm acidophilum;

maturation of dendritic cells (dC) is critical for efficient antigen presentation and initiation of an immune response. Interferon-γ(IFN-γ) is an important Thl cytokine. In this study, we investigated the role of IFN-γin dC maturation using either IFN-γreceptor deficient- or IFN-γoverexpression-models. We showed that immature dC generated in vitro from bone marrow progenitor cells produced low level of IFN-γ. After LPS stimulation, dC produced more IFN-γ, and IFN-γproductions were at comparable levels among C57BL/6 mice-derived dC (C57BL/6 dC), wild-type 129 mice-derived dC (129 dC) and IFN-y receptor deficient 129 mice-derived dC (IFN-γR-/-dC) . We found that IFN-γR-/-dC exhibited decreased expression of Cd54, Cd86, reduced capacity to secrete IL-1βand IL-12p70, and impaired capacity to stimulate alloreactive T cells and to drive Thl differentiation. Transfection of IFN-y gene into dC promoted dC to express higher Cd40, Cd54, Cd80, Cd86, CCR7 and I-Ab, secrete more IL-1βand IL-12p70, and more potently activate both Cd4 and CdS T cells. These data suggest that IFN-γsignaling pathway is important for the maturation of dC in an autocrine fashion.

Results:The initial results of the real-time in vivo measurements with mOSFTEs during serial tomotherapy treatment of head and neck cancer patients are *** mOSFET sensitivity did not change with *** photon field sizes less than 20×20 cm,the mOFET sensitivity varied with field size within 1.0%.The detector angular response was isotropic within 2%over full 360°,and the observed sensitivity variation with depth due to changes in the radiation energy spectrum waa negligible in high energy photon *** mOSFET measurements agreed with the ionization chamber results in ImRT phantom plan verification at all points to within±5%.The overall deviation of in vivo measurements for head and neck cancer patients undergoing serial tomotherapy treatment was on average 3.33%ranging from -2.20%to 7.89%,compared with doses predicted by a commercial treatment planning system (TPS).Less than 5%deviation from the planned dose was measured in 92%of all cases. Conclusion:The developed real-time in vivo mOSFET dosimetry system has proven to be a useful tool both in evaluating the actual dose within individual patients and in avoiding misadministration in the delivery of serial ImRT treatment.

＜正＞more than 170 microsurgical head and neck reconstructions are performed each year at the m. d. anderson cancer center. The most common defects requiring major reconstructions are tongue （24%） and mandible （24%）. Other defects include pharyngoesophagus （17%）, orbitomaxilla and anterior cranial base （15%）, auriculotemporal and lateral cranial base （8%）, lip and other soft tissue （6%）, scalp （5%）, and trachea （1 % ）. The most common flap used for head and neck reconstruction in the last five years is the anterolateral thigh （ALT） flap （42%）, followed by the radial forearm flap （19%）, fibular flap （17%）, rectus abdominis muscle or myocutaneous （RAm） flap （14% ）, latissimus dorsi flap （3%）, and the jejunal flap （2%）. Notable changes include the rapidly increasing number of ALT flaps and a decrease of the RAm and jejunal flaps. The basic goal of head and neck reconstruction is to provide reliable coverage to allow primarily healing and timely delivery of adjuvant therapy as well as to avoid life - threatening complications. Therefore, failure of reconstruction is less tolerable in head and neck cancer patients. Other important goals in head and neck reconstruction include preservation or restoration of speech and swallowing functions, maximizing cosmetic results, and minimizing donor site morbidities. Complicated reconstruction requires careful planning, advanced microsurgical skills, meticulous perioperative care, and prompt management of complications. Expert team effort is essential to achieve these goals. Only these complex reconstructions will be discussed in this chapter.

雷公藤是一种良好的抗风湿、抗免疫中草药,临床和实验研究表明其具有多种药理学活性。本文将30只雄性Sprague-dawley大鼠完全随机分成雷公藤醋酸乙酯提取物(TWEE)组、强的松组和对照组三组。分别口服给药(TWEE,25 mg/kg,bid;强的松,2 mg/kg,bid;对照组,0.5%羧甲基纤维素钠),并随时根据体重的变化调整用药量。所有大鼠于第31天9:30～11:00断头处死,分离提取垂体和收集血液。对垂体进行HE组织学染色和免疫组织化学染色;对血液则应用放射免疫分析法检测血浆中的促肾上腺皮质激素(ACTH)含量。实验结果表明:TWEE能促进大鼠垂体远侧部ACTH细胞合成和分泌ACTH,从而为临床综合评价雷公藤对下丘脑-垂体-肾上腺轴的影响提供了参考实验数据和理论依据,进一步指导临床安全、合理用药。

methylating agents are involved in bladder carcinogenesis. O6-methylguanine-dNA methyltransferase (mGmT) is a dNA repair protein that removes methyl group from O6-methylguanine and thus plays an important role in the etiology of cancer. We hypothesized that two mGmT polymorphisms in exon 3, C16195T (or mGmT L53L) and C16286T (or mGmT L84F), are associated with risk of bladder cancer. In a hospital-based case-control study of 167 patients with bladder cancer and 204 cancer-free controls frequency-matched by age, sex, smoking status, and alcohol use, we genotyped these two mGmT polymorphisms. We found that these two polymorphisms alone had a nonsignificant main effect on risk of bladder cancer. However, when these two polymorphisms were evaluated together, individuals with the combined genotypes or haplotypes with one or more variant alleles (i.e., the 16195T and 16286T alleles) had statistically significantly increased risk of bladder cancer (adjusted odd ratio [OR] = 1.67, 95% confidence interval [CI], 1.01-2.77) compared with those with no variant allele. In the stratification analysis, the risk of bladder cancer was increased in a dose-response manner as the age increased (Ptrend - 0.010), and the increased risk was more pronounced among old subjects (> 65 years) (adjusted OR = 2.51, 95% CI, 1.05-6.04), men (1.76, 1.00-3.10), and non-drinkers (1.91, 1.08-3.36). In conclusion, these two mGmT polymorphisms may jointly play a role in the etiology of bladder cancer in southern Chinese population. Larger studies are warranted to validate our findings.

Recent studies have revealed that the γ-chain of theIL-2 receptor is shared by the receptors for IL-4, IL7, IL-9, IL-13, and IL-15, and it is therefore also referred toas the common γ-chain (γc). mutations of γc result inX-linked severe combined immunodeficiency syndrome inhumans, indicating that rye is essential for normal development and function of the immune system. We demonstratethat human hematopoietic cells express two γc transcriptsdiffering in their carboxyl terminal coding region. Onetranscript is the previously reported sequence (γc-long),whereas the newly identified sequence exhibits a deletion of72 nucleotides close to the 3'-end of the open reading frame(γc-short). This alteration predicts a loss of 24 amino acidsincluding a conserved tyrosine residue which is shared byseveral members of the cytokine receptor family. Thepresence of these two distinct forms of rye transcripts wasdemonstrated by sequencing of reversely transcribed andpolymerase chain reaction (RT-PCR) amplified mRNA, restriction digestion of the RT-PCR products, RNAse protection, and Northern blotting from human cell lines andhuman peripheral blood lymphocytes. Furthermore, thetwo variants were present in peripheral blood lymphocytesfrom both female and male donors, which rules out allelicvariants since rye is a single copy gene located on the Xchromosome. A truncation mutant at a site near the observed changes in γc-short has been reported by othersto alter biochemical events activated by cytokines. Thiscombined with the loss of a potential SH2 "docking" sitein γc-short suggests that γc-long and γc-short may link todifferent signaling pathways and may play an importantrole in determining the cellular response to IL-2, IL-4, IL-7, IL-9, IL-13, IL-15.
