Identification of Prorocentrum minimum and Takayama pulchella by fluorescence in situ hybridization through epifluorescence microscopy and flow cytometry

Partial rDNA sequences of Prorocentrum minimum and Takayama pulchella were amplified, cloned and sequenced, and these sequence data were deposited in the GenBank. Eight oligonucleotide probes （DNA probes） were designed based on the sequence analysis. The probes were employed to detect and identify P. minimum and T. pulchella in unialgal and mixed algal samples with a fluorescence in situ hybridization method using flow cytometry. Epifluorescence micrographs showed that these specific probes labeled with fluorescein isothiocyanate entered the algal cells and bound to target sequences, and the fluorescence signal resulting from whole-cell hybridization varied from probe to probe. These DNA probes and the hybridization protocol we developed were specific and effective for P. minimum and T. pulchella, without any specific binding to other algal species. The hybridization efficiency of different probes specific to P. minimum was in the order： PM18S02 PM28S02 〉 PM28S01 〉PM18S01, and that of the probes specific to T. pulcheUa was TP18S02 TP28S01 〉 TP28S02 〉TP18S01. The different hybridization efficiency of the DNA probes could also be shown in the fluorescent signals between the labeled and unlabeled cells demonstrated using flow cytometry. The DNA probes PM18S02, PM28S02, TP18S02 and TP28S01, and the protocol, were also useful for the detection of Mgae in natural samples.

By applying nonequilibrium Green's function formalism combined with first-principles density functional theory, we investigate the electronic transport properties of the dihydroazulene optical molecular switch. Three kinds of adsorption sites including the hollow, bridge and top sites are studied. The two forms of this molecule, namely the open form and the closed form, can reversibly switch from each other upon photoexcitation. Their transmission spectra are remarkably distinctive. Theoretical results show that the current of the closed form is always significantly larger than that of the open form for all three adsorption sites, which promises this system as possibly one of the good candidates for optical switches due to its unique advantage, and which may have some potential applications in the future molecular circuit.

Noninvasive estimation of the soft tissue kinematics properties from medical image sequences has many important clinical and physiological implications, such as the diagnosis of heart diseases and the understanding of cardiac mechanics. In this paper, we present a biomechanics based strategy, framed as a priori constraints for the ill-posed motion recovery problems, to realize estimation of the cardiac motion and deformation parameters. By constructing the heart dynamics system equations from biomechanics principles, we use the finite element method to generate smooth estimates of heart kinematics throughout the cardiac cycle. We present the application of the strategy to the estimation of displacements and strains from in vivo left ventricular magnetic resonance image sequence.

This paper demonstrates an atmospheric correction method to process HJ-1A/B CCD images over Chinese coastal waters with the aid of MODIS-Terra aerosol *** on the assumption of zero water-leaving radiance at the two near-infrared (NIR)bands or the shortwave infrared(SWIR)bands,the atmospheric aerosol optical depth(AOD)is firstly retrieved from MODIS-Terra with a simple extension of the NIR-SWIR combined atmospheric correction approach embedded in SeaDAS (SeaWiFS data analysis system).Then the"turbid"and"non-turbid"waters are separated by the turbid water *** probability of AOD at 551 nm band is derived based on the NIR information over"non-turbid"waters and FLAASH model is selected to do the atmospheric correction of the HJ-1A/B CCD imagery using the AOD values of highest probabilities as ***,according to the histogram of the AOD distribution at 551 nm band,the studied turbid water area is divided into several blocks and HJ-1A/B CCD imagery is corrected with the corresponding AOD values displayed with the highest frequency in each *** method has been applied to several HJ-1A/B images over Chinese coastal waters and validated by synchronous in-situ *** results have shown that this method is effective in the atmospheric correction process of HJ-1A/B CCD images for ocean color remote sensing study and application in the coastal waters.

Fourteen vitrinite reflectance profiles from the Tarim Basin,NW China,show that the vitrinite reflectance profiles of individual wells follow a faulted and dislocated dual-stage pattern in the eastern section of the Tazhong Uplift and in the Tabei *** reflectance values in these profiles change sharply at the unconformity beneath the Lower Carboniferous Bachu or Upper Devonian Donghetang Formations,where the overlying Triassic to Carboniferous strata are still in a mature phase within the "liquid oil window".However,the underlying Lower Paleozoic reached the overmature phase beyond the "liquid oil window" towards the end of the Silurian,or in the Early Devonian at the *** the vitrinite reflectance profiles are attributed to a continuous,single-stage pattern in the western section of the Tazhong Uplift,in which the Lower Paleozoic is also in an overmature phase,their overmaturity would have been achieved relatively late in geological *** stratigraphic thermohistory has the following implications to regional geoevolution:(1) The overmature Lower Ordovician to Cambrian strata in the eastern section of the Tazhong Uplift and in the Tabei Uplift,as well as in the Manjiaer Depression,could not have acted as the source kitchen for normal oil(so-called black oil);(2) The dissimilarity in vitrinite reflectance profiles between the eastern and western sections of the Tazhong Uplift reveals Early Paleozoic paleotectonic features,i.e.,lower at the east and higher at the west,whereas recent tectonic features formed since the Late Paleozoic are in reverse,i.e.,higher in the east and lower at the west;(3) Reconstruction of the denuded thickness of sediments overlying the Lower Ordovician strata suggests reconsideration on the paleotectonic features in the Tarim Basin;(4) Based on the sustained duration of the "liquid oil window" for overmature source beds,it is predicted that the Suntuoguole Lower Uplift between the Awati and Manjiaer Depressions is a

Aquatic vegetation plays an important role in the flow structure of open channels and thus changes the fate and the transport of sediment. This article proposes a three-dimensional turbulence model by introducing vegetation density and drag force into the control equations of water flow in the presence of vegetation. The model was used to calculate the impacts of submerged vegetation on the vertical profiles of longitudinal flow velocities, the changes of the depth-averaged flow velocities in a compound channel with emergent vegetation in the floodplain, the removal of suspended sediment from the channels by emergent vegetation, and the bed changes around and in a vegetated island. Numerical investigations show that aquatic vegetation retards flow in the vegetation zone, reduces the sediment transport capacity, and contributes to erosion on both sides of the vegetated island. Calculated results agree well with experimental results.

Peridynamics (PD), a recently developed theory of solid mechanics, which employs a non-local model of force interaction and makes use of integral formulation rather than the spatial partial differential equations used in the classical continuum mechanics theory, has shown effectiveness and promise in solving discontinuous problems at both macro and micro scales. In this paper, the peridynamics theory is used to analyze damage and progressive failure of concrete structures. A non-local peridynamic model for a rectangular concrete plate is developed, and a central pairwise force function is introduced to describe the interior interactions between particles within some definite distance. Damage initiation, evolution and crack propagation in the concrete model subject to in-plane uni-axial tension, in-plane uni-axial compression and out-of-plane impact load are investigated respectively. The numerical results show that discontinuities appear and grow spontaneously as part of the solution to the peridynamic equations of motion, and no special failure criteria or re-meshing techniques are required, which proves the potential of peridynamic modeling as a promising technique for analyzing the progressive failure of concrete materials and structures.

In order to ascertain the invasive mechanism and control strategy of the invasive Crofton weed, Ageratina adenophora, its ecological adaptability and population differentiation,the formation of single dominant population, displacement of native plants and sustainable management strategies were investigated. The present results helped to clarify and explain such issues as the adaptability post invasion,interaction and competition between inter-and intra-species and community resistance, thereby providing important references to researches on other invasive alien species.
