Polyethylenimine (PEI) has been known as an efficient gene carrier with the highest cationiccharge *** transfection efficiency of PEI,along with its cytotoxicity,strongly depends on itsmolecular *** enhance its gene delivery efficiency and minimize cytotoxicity,we have synthesizedsmall cross-linked PEI with biodegradable linkages and evaluated their transfection efficiencies in *** study,branched PEI with a molecular weight of 800 Da was cross-linked by small diacrylate[1,4-butanediol diacrylate or ethyleneglycol dimethacrylate (EGDMA)] for 2-6 *** efficiencies of thecross-linked PEI in in vitro transfection of plasmid DNA containing enhanced green fluorescent protein(EGFP) reporter gene were assessed in melanoma B 16F10 cell line and other cell *** cytometrywas used to quantify the cellular entry efficiency of plasmid and the transgene expression *** of the cross-linked PEI in these cells were evaluated by MTT ***-PEI 800-4h,atypical cross-linked PEI reported here,mediated a more efficient expression of reporter gene than thecommercially available 25-kDa branched PEI control,and resulted in a 9-fold increase in gene deliveryin B16F10 cells and a 16-fold increase in 293. cells,while no cytotoxicity was found at the optimizedcondition for gene ***,the transfection activity of polyplexes was preserved in thepresence of serum proteins.

The role of Chinese expert consensus in standardization and improvement of the clinical diagnosis and treatment of inflammatory bowel disease (IBD) is self-evident. As clinical and basic research in china becomes standardized under a better understanding of IBD, more data on Chinese patients have become available to develop a consensus and guideline. This guideline, collaboratively developed by the IBD Group, Chinese Society of Gastroenterology, incorporates the latest international consensus statements,[1-7] domestic research findings, and practical considerations, as an update based on the 2018 Chinese consensus on IBD management.[8] The IBD guideline consists of two parts: ulcerative colitis (UC) and Crohn's disease, and this manuscript presented the UC part. The evidence used in this guideline was collected and analyzed by the standard of guideline, and the contents were organized as problem statements, for clarity. The formulation of this guideline aimed to reflect the latest progress in IBD, providing comprehensive and valuable guidance for the clinical management of IBD.

The functional magnetic resonance imaging (fMRI) based on blood oxygen level dependent (BOLD) contrast has emerged as one of the most potent noninvasive tools for mapping brain function and has been widely used to explore physiological, pathological changes and mental activity in the brain. Exploring the nature and property of BOLD signal has recently attracted more attentions. Despite that great progress has been made in investigation of the characteristics and neurophysiological basis, the exact nature of BOLD signal remains unclear. In this paper we discuss the characteristics of BOLD signals, the nonlinear BOLD response to external stimuli and the relation between BOLD signals and neural electrophysiological recordings. Furthermore, we develop our new opinions regarding nonlinear BOLD response and make some perspectives on future study.

Extracellular signal-regulated protein kinase (ERK) has been implicated in the pathogenesis ofseveral nerve system *** more and more kinases have been discovered to be the client proteins ofthe molecular chaperone Hsp90,the use of Hsp90 inhibitors to reduce abnormal kinase activity is a newtreatment strategy for nerve system *** study investigated the regulation of the ERK pathway *** showed that Hsp90 inhibitors reduce ERK phosphorylation without affecting the total ERKprotein *** investigation showed that Raf,the UPstream kinase in the Ras-Raf-MEK-ERK pathway,forms a complex with Hsp90 and *** cells with Hsp90 inhibitors facilitates Raf degradation,thereby down-regulating the activity of ERK.

Aim:To explore the effects and mechanisms of caffeine and coffee on bladderdysfunction in streptozotocin-induced diabetic ***:Sprague-Dawleymale rats were divided randomly into 4 groups:control,diabetes mellitus(DM),DM with coffee treatment,and DM with caffeine *** diabetic rat wasinduced by intraperitoneal injection of streptozotocin(60 mg/kg).After 7 weeks oftreatment with coffee and caffeine,cystometrogram,contractile responses to elec-trical field stimulation(EFS)and acetylcholine(ACh),and cyclic AMP(cAMP)concentration of the bladder body and base were ***:The bladderweight,volume threshold for micturition and post-void residual volume(PVR)inthe diabetic rats were significantly higher compared to those in the control *** or caffeine treatment significantly reduced the bladder weight,bladdercapacity and PVR in the diabetic *** caused significant decreases in cAMPconcentration of the bladder and coffee and caffeine caused upregulation of cAMPcontent in the diabetic *** addition,coffee and caffeine tended to normal-ize the altered detrusor contractile responses to EFS and ACh in the diabetic ***:These results indicate that caffeine and coffee may have beneficialeffects on bladder dysfunction in the early stage of diabetes by increasing cAMPcontent in the lower urinary tract,recovering the micturition reflex and improvingthe detrusor contractility.

Three-dimensional elasticity solution of simple-supported rectangular plate on point supports, line supports and elastic foundation

This paper studies the bending of simple-supported rectangular plate on point supports, line supports and elastic foundation. On the basis of three-dimensional elasticity theory, the exact expressions of the displacement functions, which satisfy the governing differential equations and the simply supported boundary conditions at four edges of the plate, are analytically derived. The reaction forces of the in- termediate supports are regarded as the unknown external forces acting on the lower surface of the plate. The unknown coefficients are then determined by the boundary conditions on the upper and lower surfaces of the plate. Comparing the numerical results obtained from the proposed method to those obtained from Kirchhoff plate theory, Mindlin plate theory and those obtained from the commer- cial finite element software ANSYS, the high accuracy of the present method has been demonstrated.

Anticancer effect of two diterpenoid compounds isolated from Annona glabra Linn

AIM: To study the inhibitory effect of two diterpenoid compounds isolated from Annona glabra Linn (Cunabic acid and ent-kauran-19-al-17-oic acid) on the proliferation of Human Liver Cancer (HLC) cell line SMMC-7721 and its mechanism. METHODS: Inhibition of cell proliferation was measured by MTT assay. The morphological changes of SMMC-7721 cells were observed under inverted phase-contrast microscope, fluorescent microscope, transmission electron microscope (TEM), and scanning electron microscope (SEM). Flow cytometer (FCM) was used to calculate the cell apoptotic rate, and immunohistochemical staining was used to observe the regulation of gene expression. RESULTS: The proliferation of SMMC-7721 cells was obviously inhibited after being treated with Cunabic acid at the concentration >5 μmol/L and ent-kauran-19-al-17-oic acid >10 μmol/L. The biggest inhibitory effect was 81.05 % when treated with Cunabic acid at the concentration of 25 μmol/L. The effect had a linear relationship with concentration. The result indicated that drug-treated cells exhibit typical morphological changes of apoptosis, including condensed chromatin and a reduction in volume. Sub-G0/G1 peak was found by FCM analysis and the cell cycle was arrested at G0/G1 stage. The apoptotic rates of the cells treated by Cunabic acid and ent-kauran-19-al-17-oic acid were 43.3. % and 24.95 %, respectively. It was visualized by immunohis- tochemical staining that the drugs down-regulated the gene expression of bcl-2 gene and up-regulated that of bax gene. CONCLUSION: The two diterpenoid compounds isolated from Annona glabra Linn, Cunabic acid and ent- kauran-19-al-17-oic acid can obviously inhibit the proliferation of HLC cell line SMMC-7721. The mechanism is correlated with the induction of cell apoptosis by down-regulating the gene expression of bcl-2 gene and up- regulating that of bax gene.

Alkali-silica Reactivity of Different Aggregates from Mineral and Textural Characteristics

To identify the aggregates causing the alkali-silica reaction, and the reactivity of rocks in different parts of china, the mineral and texture characteristics of some typical coarse ag gregates and the alkali reactivity of these aggregates were systematically investigated. On one hand, petrographic examination of aggregates, combined with X-ray diffraction analysis, chemical analysis, scanning electron microscopy, was conducted for analyzing their mineralogy and texture. It was found that not only mineral characteristics, but also their interior structure would affect their potential alkali-silica reactivity. Furthermore, the alkali-silica reactivity of aggregates is due to their mineral compositions, such as containing different alkali-reactive minerals, while, different stuctures of those aggregates with similar minerals affect their reactivity to some extent. There are some amount of micro-aperture and cracks across the quartz in the aggregate, which may become the natural accesses for Na^＋, K^＋, and OH^- ions to intrude in the mortar or concrete during the process of alkali-silica reaction. Alkali-silica reactivity of the aggregates was detected by the accelerated mortar bars test. It was revealed that all the aggregates tested were alkali-silica reactive and the results were also in accordance with their mineral and structure analysis.
