AIM: Toassess mitotic stability ofthe fragile X fullmutations andits relationshipwith DNA methylation. METHODS: The length change of the expanded CGG repeats was examined and correlated it with the methylation status in the DNA samplesisolated fromthe fibroblasts derived from a fragile Xfemale fetus anda fragileX maleadult, respectively. RESULTS: A dramatic instability of the expanded CGG repeats in the female fetal fibroblasts was observed. South- ern blot analysis revealed that the 6.9-kb major expanded band detected in passage 2 was completely replaced by a 7.7-kb band after passage 3.. Fibroblast clones derived from the passage 3.displayed an unstable expansion of the CGG repeat during clonal proliferation, while methylation status of the CGG repeat region was maintained. In contrast, in fragile X male fibroblasts the expanded CGG repeats were stable during clonal proliferation. CONCLUSION: The mitotic instability of expanded CGG repeat is not always restricted in early development window as proposed previously and other elements rather than DNA methylation could affect the stability of the expanded CGG repeats in fragile X female fetal fibroblast cells.

本文采用HRD2型双光栅单色仪测定了熊果酸的Raman光谱 ,得到熊果酸的特征Raman光谱结构 ,采用Philips 10 0型双光栅单色仪测定了熊果酸的IR光谱 ,得到熊果酸的特征IR光谱结构 ;解析了熊果酸的Raman光谱和IR光谱与其结构特征的关系 ,确定了熊果酸的Raman光谱和IR光谱的结构特征和官能团的归属 ,给出了熊果酸的两个基本骨架特征区域 (A区 :13.6 ,13.0 ,13. 3.m- 1 ,B区 :13. 4 ,12 6 9,12 3.cm- 1 ) ;对熊果酸的Raman光谱和IR光谱作了比较研究 ,指出Raman光谱具有比IR光谱更为丰富的结构信息的特征 ,并且 ,IR光谱的主要特征峰在Raman光谱中均有对应的谱峰 ;Raman光谱的谱峰形态清晰可变 ,使熊果酸的主要结构和官能团得到准确的分析和指认 ,与IR光谱一样 ,Raman光谱是天然药物解析的有效手段之一。

With the method of large-eddy simulation,the equation of spherule motion and the method of immersed boundary condition,numerical simulations of three-dimen-sional turbulent aeolian motion and the formation of sand ripples under three-di-mensional turbulent wind and the mutual actions of saltation and creeping motion were carried out. The resulting sand ripples have the form that is flat on the upwind side and steep on the leeward,which is identical to the sand ripples in nature. We also realized the self-restoration process of three-dimensional sand ripples,which shows the correctness of the method of numerical simulation and the models of saltation and creeping. Finally,We analyzed the influence of sand ripples on the three-dimensional turbulent wind field,and found that due to the appearance and development of sand ripples,in the normal direction of ground there exists stronger energy exchange,and moreover,there is close correspondence between the forms of sand ripples and the vorticity close to the ground surface.

This article is focused on analyzing the key technologies of new malicious code and corresponding defensive measures in the large-scale communication networks. Based on description of the concepts and development of the malicious code, the article introduces the anti-analysis technology, splitting and inserting technology, hiding technology, polymorph virus technology, and auto production technology of the malicious code trends with intelligence, diversity and integration. Following that, it summarizes the security vulnerabilities of communication networks from four related layers aspects, according to the mechanisms of malicious code in the communication networks. Finally, it proposes rapid response disposition of malicious code attacks from four correlated steps： building up the network node monitoring system, suspicious code feature automation analysis and extraction, rapid active malicious code response technique for unknown malicious code, and malicious code attack immunity technique. As a result, it actively defenses against the unknown malicious code attacks and enhances the security performance of communication networks.

Objective To establish a rapid,sensitive and selective liquid chromatography-tandem mass spectrometry (LC-MS/MS) method for the determination of acyclovir (the metabolite of valacyclovir hydrochloride) in human plasma. Methods After addition of ganciclovir as internal standard (IS),plasma samples were prepared by one-step protein precipitation using acetonitrile as precipitant,followed by an isocratic elution with 0.1% formic acid solution-methanol (95∶5,v/v) on an Agilent ZORBAX SB-C18 (150mm×2.1mm i.d.,3.5μm) column. Detection was performed on a triple-quadrupole mass spectrometer utilizing electrospray ionization (ESI) interface operating in positive ion and selected reaction monitoring (SRM) mode with the precursor to product ion transitions m/z 226.2→152.1 for acyclovir and m/z 256.2→152.1 for the IS. Results The analytical results demonstrated a good linearity over the ranges from 0.005 to 4μg/mL (r=0.9999) for valacyclovir hydrochloride. The relative standard deviations (RSD) of intra-batch and inter-batch were less than 4.06% and 9.23.,respectively. The limit of detection and lower limit of quantification in human plasma were 2ng/mL and 5ng/mL,respectively. Conclusion The method was simple,sensitive,accurate and reproducible and has been successfully applied to a bioequivalence study of valacyclovir hydrochloride capsules in Chinese healthy male volunteers.

Fifty-six samples of nannofossil ooze were collected from Core PC5794 in the northern equatorial Pacific at 5 cm intervals. With the methods of mass spectrometer (VG3.4) and ICP, the Nd isotopic compositions (εNd(t)), Mn contents and Mg/Sr ratios of carbonate phase have been analyzed. CaCO3.contents of bulk sediments were obtained by dissolution of 0.5 mol/L HCl. Based on these data, the high-resolution εNd(t) profile of seawater in early Miocene with core depth(or time) have been established. The values of εNd(t) range from -6.2 to -2.97 and 4 fluctuation cycles existed during 24.06-22.02 Ma. 4 low εNd(t) values (about -6.4) correspond to high CaCO3.contents, which implicates that there were 4 cold epochs or 4 times of Antarctic Bottom Water activity. They occurred at the time of 24.06 Ma, 23.85 Ma,22.88 Ma and 22.26 Ma, respectively. High εNd(t) values correspond to the high Mn contents and high values of Mg/Sr ratio, which indicates the existence of 4 intense hydrothermal activity periods during 24.06-22 Ma, the durations of them are 4.05-23.98 Ma, 23.69-23.15 Ma, 22.74-22.3. Ma and 22.06-22.02 Ma, respectively.

The shear strength parameters property of rock-soil aggregates in embankment slope of reservoir,that is,the relationship between cohesion and gravel content,between friction angle and gravel content,and the relationship between cohesion and water content,between friction angle and water content,is studied based on the direct shear test results,the shear strength change law of the rock-soil aggregates is given,and the unsaturated shear strength formulation of rock-soil aggregates that could consider suction and saturation degree influence is put forward in this paper,through which the sliding or failure physical mechanism of this type of slope under the condition of rainfall infiltration is studied. Also the 3. unsteady saturated-unsaturated seepage field and its FEM resolving mode are established based on the analysis of the slope rainfall infiltration process. Case study with this method indicates that the minimum safety factor of the accumulated rock-soil aggregates dose not arrive at the moment of rainfall cessation,but appears several hours after the rainfall cessation,this phenomenon is in accordance with the practical slope engineering's failure process and could explain appropriately the slope failure caused by rainfall infiltration. Research results in this paper have an important reference value for the research on stability of the accumulated rock-soil aggregates in embankment slope of reservoir,and can enrich the stability analysis method and relevant theory of reservoir embankment slope.

In this paper, to overcome the drawbacks of POT which minimize the reminder, not only the optimal functional with minimal residual is put forward, but also the method of global optimization is used. Under the condition of large reminder, the optimal bases can also be obtained through the method of POTres without the approaching of initial conditions. In addition, compared with local optimization, the advantage of global optimization is remarkable. On the one hand, we can expect that the dynamical system based on the global optimal bases will include more information than the ones based on the local optimal bases, since the global optimal bases are much more precise than the local optimal bases. On the other hand, from the point of view of error, the global optimal bases are independent of the choice of the object functional and the initial bases.
