Cloning and characterization of interferon stimulated genes Viperin and ISG15,and their promoters from snakehead Channa argus

By suppression subtractive hybridization, rapid amplification of cDNA ends and gene walking methods, interferon stimulated genes (ISGs), Viperin and ISG15, and their promoters have been cloned and characterized from snakehead Channa argus. The Viperin cDNA was found to be 1474 nt and contain an open reading frame (ORF) of 1059 nt that translates into a putative peptide of 3.2.amino acid (aa). The putative peptide of Viperin shows high identity to that in teleosts and mammals except for the N-terminal 70 aa. The ISG15 cDNA was found to be 758 nt and contain an ORF of 468 nt that translates into a putative peptide of 155 aa. The putative peptide of ISG15 is composed of two tandem repeats of ubiquitin-like (UBL) domains, and a canonical conjugation motif (LRGG) at C-terminal. Viperin and ISG15 promoter regions were characterized by the presence of interferon stimulating response elements (ISRE) and γ-IFN activation sites (GAS). ISRE is a feature of IFN-induced gene promoter and partially overlaps interferon regulatory factor (IRF) 1 and IRF2.recognition sites. GAS is responsible for the γ-IFN mediated transcription. One conserved site for NF-κB was found in the promoter region of Viperin. This is the first report of conservative binding motif for NF-κB in accordance with the consensus sequence (GGGRNNYYCC) among teleost ISG promoters. Moreover, there were also TATA, CAAT and Sp1 transcription factor sites in Viperin and ISG15 promoters. In 5′ untranslated region (UTR), snakehead ISG15 gene contains a single intron, which differs from Viperin gene. The transcripts of Vipeirn and ISG15 mRNA were mainly expressed in head kidney, posterior kidney, spleen and gill. The expression levels in liver were found to increase obviously in response to induction by IFN-inducer poly I∶C.

A Test on Different Types of the Time Curve of Hardness Ratio of Gamma-Ray Bursts based on the Curvature Effect

We analyzed a sample of 66 gamma-ray bursts （GRBs） and statistically confirmed the prediction on the time curve of the hardness ratio of GRBs made by Qin et al. based on the curvature effect. In their analysis, GRB pulses are divided into three types according to the shape of their raw hardness ratio （RHR） time curves, defined as to include the background counts to the signal counts, so as to make use of counts within small time intervals. Of the three types, very hard sources exhibit a perfect pulse-like profile （type 1）, hard bursts possess a pulse-like profile with a dip in the decay phase （type 2., and soft bursts show no pulse-like profile but have only a dipped profile （type 3.. In terms of the conventional hardness ratio, type 3.sources are indeed generally softer than those of type 1 and type 2. in agreement with the prediction. We found that the minimum value of RHR is sensitive in distinguishing the different types. We propose that GRB pulses can be classified according to the minimum value of RHR and that the different type sources may be connected with different strengths of the shock or/and the magnetic field.

Gasdynamic characteristics of toroidal shock and detonation wave converging

The modified CCW relation is applied to analyzing the shock, detonation wave converging and the role of chemical reactions in the process. Results indicate that the shock wave is strengthened faster than the detonation wave in the converging at the same initial Mach number. Euler equations implemented with a detailed chemical reaction model are solved to simulate toroidal shock and detonation wave converging. Gasdynamic characteristics of the converging are investigated, including wave interaction patterns, observable discrepancies and physical phenomena behind them. By comparing wave diffractions, converging processes and pressure evolutions in the focusing area, the different effects of chemical reactions on diffracting and converging processes are discussed and the analytic conclusion is demonstrated through the observation of numerical simulations.

Research on the method for retrieving soil moisture using thermal inertia model

In order to improve accuracy of soil moisture inversion using remote sensing, a new thermal inertia model is proposed in this paper. The improved model needs only surface maximum temperature as the temperature parameter input instead of input of the surface temperature difference, as well as the surface sensible and latent fluxes are introduced into boundary conditions of thermal conductivity equation. Furthermore, surface soil conductive heat transfer equation of two-layer model is used to solve the soil thermal inertia so that the remote sensing thermal inertia method can be ap- plied to regions with better-covered vegetation, but usually only for the bare areas or worse vegetation covered areas. The model has been tested at several locations in the area of west Inner Mongolia. Comparing the simulation of the new model with the measurements obtained by apparent thermal inertia and by field test, the result shows that the inertia thermal model can be used to estimate soil moisture in more reasonable accuracy.

Effects of cosmological constant on motion of UHECR particles

Recent astronomical observations manifest that about two-thirds of the whole energy in the Universe is contributed by a small positive cosmological constant A (> 0). Then, an asymptotically de Sitter spacetime is premised naturally. However, physics in the de Sitter spacetime is very different from that in the Minkowski spacetime. As the first step, a covariant formalism of the kinematics in the de Sitter spacetime is presented here. By solving exactly the equations of motion for a field, we obtain the dispersion relation of a free particle. It is noticed that the dispersion relation is dependent on the degree of freedom of angular momentum of the particle. We show the threshold anomaly of the ultra high energy cosmic ray disappears naturally in the framework of the de Sitter kinematics.

Bruton’s tyrosine kinase inhibitors(BTKis)have revolutionized the treatment of B-cell ***,safety issues related to the use of BTKis may hinder treatment continuity and further affect clinical efficacy.A comprehensive and systematic expert consensus from a pharmacological perspective is lacking for safety issues associated with BTKi treatment.A multidisciplinary consensus working group was established,comprising 3. members from the fields of hematology,cardiovascular disease,cardio-oncology,clinical pharmacy,and evidencebased *** evidence-based expert consensus was formulated using an evidence-based approach and the Delphi *** Joanna Briggs Institute Critical Appraisal(JBI)tool and Grading of Recommendations Assessment,Development,and Evaluation(GRADE)approach were used to rate the quality of evidence and grade the strength of recommendations,*** consensus provides practical recommendations for BTKis medication based on nine aspects within three domains,including the management of common adverse drug events such as bleeding,cardiovascular events,and hematological toxicity,as well as the management of drug-drug interactions and guidance for special *** multidisciplinary expert consensus could contribute to promoting a multi-dimensional,comprehensive and standardized management of BTKis.

Study on the coating of nano-scale SiO_2.film on the surface of nanocrystalline Mg-Al layered double hydroxides

The coating process of a nano-scale SiO2.film on the nanocrystalline Mg-Al layered double hydroxides via a sol-gel process was investigated. The uniform and dense SiO2.ilm with a thickness of about 5 nm on the nano-LDHs particles was characterized by the solubility test in the dilute HNO3.or HCl acid, TEM and FT-IR, XRD, TG and DSC. The chemical shifts of binding en- ergies of Al 2., Mg 2., Si 2. and O 1s on the coated particles indicate that the coating of the SiO2.nano-film on the surface of the nano-LDHs proceeds through the formation of Mg-O-Si and Al-O-Si bonds. The thermal analysis shows that both the SiO2.coated nano-LDHs and the nano-LDHs have a similar mass loss process, in which there are three obvious stages of mass loss in the temperature range of 40—700℃. Furthermore, the more the coatedamount of SiO2.n the surface of the nano-LDHs is, the less the mass loss of the samples is at 700℃.The nano- LDHs have two obvious endothermic peaks at 2.4.67℃ and 43..13., whose corresponding heat absorption capacities are 412.2. J/g and 3.6.3. J/g, respectively. In contrast, the coated nano-LDHs have only one endothermic peak at 2.3.60℃ with a heat absorption capacity of 2.1.2. J/g.

With large-scale production and application at large scale, carbon nanotubes (CNTs) may cause ad-verse response to the environment and human health. Thus, study on bio-effects and safety of CNTs has attracted great attention from scientists and governments worldwide. This report briefly summa-rizes the main results from the in vitro toxicity study of CNTs. The emphasis is placed on the descrip-tion of a variety of factors affecting CNTs cytotoxicity, including species of CNTs, impurities contained, lengths of CNTs, aspect ratios, chemical modification, and assaying methods of cytotoxicity. However, experimental information obtained thus far on CNTs' cytotoxicity is lacking in comparability, and some-times there is controversy about it. In order to assess more accurately the potential risks of CNTs to human health, we suggest that care should be taken for issues such as chemical modification and quantitative characterization of CNTs in cytotoxicity assessment. More importantly, studies on physical and chemical mechanisms of CNTs' cytotoxicity should be strengthened; assaying methods and evaluating criteria characterized by nanotoxicology should be gradually established.

Validating global hydrological models by ground and space gravimetry

The long-term continuous gravity observations obtained by the superconducting gravimeters (SG) at seven globally-distributed stations are comprehensively analyzed. After removing the signals related to the Earth's tides and variations in the Earth's rotation, the gravity residuals are used to describe the seasonal fluctuations in gravity field. Meanwhile, the gravity changes due to the air pressure loading are theoretically modeled from the measurements of the local air pressure, and those due to land water and nontidal ocean loading are also calculated according to the corresponding numerical models. The numerical results show that the gravity changes due to both the air pressure and land water loading are as large as 100×10-9 m s-2.in magnitude, and about 10×10-9 m s-2.for those due to the nontidal ocean loading in the coastal area. On the other hand, the monthly-averaged gravity variations over the area surrounding the stations are derived from the spherical harmonic coefficients of the GRACE-recovered gravity fields, by using Gaussian smoothing technique in which the radius is set to be 600 km. Com-pared the land water induced gravity variations, the SG observations after removal of tides, polar mo-tion effects, air pressure and nontidal ocean loading effects and the GRACE-derived gravity variations with each other, it is inferred that both the ground- and space-based gravity observations can effec-tively detect the seasonal gravity variations with a magnitude of 100×10-9 m s-2.induced by the land water loading. This implies that high precision gravimetry is an effective technique to validate the re-liabilities of the hydrological models.
