Delimitations of species are crucial for correct and precise identification of *** "spe-cies" is more a subjective than an objective concept in taxonomic practice due to difficulties in revealing patterns of infra-or inter-specific *** phylogenetic studies at the population level solve this problem and lay a sound foundation for DNA *** this paper we exemplify the necessity of adopting a phylogenetic concept of species in DNA barcoding for tree peonies(Paeonia ***).We used 40 samples representing all known populations of rare and endangered species and several populations of widely distributed tree *** currently recognized species and major variants have been included in this *** chloroplast gene fragments,***,rps16-trnQ,trnL-F and trnS-G(a total of 5040 characters,96 variable and 69 parsimony-informative characters) and one variable and single-copy nuclear GPAT gene fragment(2.93.2.97 bp,2.9 variable and 148 parsi-mony-informative characters) were used to construct phylogenetic relationships among the *** evolutionary lineages revealed by the nuclear gene and the chloroplast genes are inconsistent with the current circumscriptions of ***,***,***,and *** based on *** inconsistencies come from(1) significant chloroplast gene divergence but little nuclear GPAT gene divergence among population systems of *** + ***,and(2. well-diverged nuclear GPAT gene but little chloroplast gene divergence between *** and *** incongruence of the phylogenies based on the chloroplast genes and the nuclear GPAT gene is probably due to the chloro-plast capture event in evolutionary history,as no reproductive barriers exist to prevent inter-specific *** also evaluated the suitability of these genes for use as DNA barcodes for tree *** variability of chloroplast genes among well-defined species or population systems of a specie

To minimize instrumentally the induced systematic errors,cosmic microwave background(CMB)anisotropy experiments measure temperature differences across the sky using pairs of horn antennas, temperature map is recovered from temperature difference obtained in sky survey through a map-making *** inspect and calibrate residual systematic errors in the recovered temperature maps is important as most previous studies of cosmology are based on these *** analyzing pixel-ring coupling and latitude dependence of CMB temperatures,we find notable systematic devia- tion from CMB Gaussianity in released Wilkinson Microwave Anisotropy Probe(WMAP)*** detected deviation cannot be explained by the best-fit LCDM cosmological model at a confidence level above 99%and cannot be ignored for a precision cosmology study.

To meet the needs of large-scale genomic/genetic studies, the next-generation massively parallelized sequencing technologies provide high throughput, low cost and low labor-intensive sequencing service, with subsequent bioinformatic software and laboratory methods developed to expand their applications in various types of research. PCR-based genomic/genetic studies, which have significant usage in association studies like cancer research, haven't benefited much from those next-generation sequencing technologies, because the shortgun re-sequencing strategy used by such sequencing machines as the Illumina/Solexa Genome Analyzer may not be applied to direct re-sequencing of short-length target regions like those in PCR-based genomic/genetic studies. Although several methods have been proposed to solve this problem, including microarray-based genomic selections and selector-based technologies, they require advanced equipment and procedures which limit their applications in many laboratories. By contrast, we overcame such potential drawbacks by utilizing a ligation by amplification (LBA) protocol, a method using a pair of Universal Adapters to randomly ligate target regions in a two-step-PCR procedure, whose Long LBA products were easily fragmented and sequenced on the next-generation sequencing machine. In this concept-proven study, we chose the consensus coding sequences of two human cancer genes: BRCA1 and BRCA2.as target regions, specifically designed LBA primer pairs to amplify and randomly ligate them. 70 target sequences were successfully amplified and ligated into Long LBA products, which were then fragmented to construct DNA libraries for sequencing on both a conventional Sanger sequencer ABI 3.3.xl DNA Analyzer and the next-generation 'synthesis by sequencing technology' Illumina/Solexa Genome Analyzer. A novel myotoxic protein phospholipase A2.PLA2., denoted as Gln49-PLA2. has been isolated from snake venom of Agkistrodon blomhoffii ussurensis, which has weak lethal effect and apparent anticoagulant activity, but lacks the PLA2.and hemorrha-genic activity. Gln49-PLA2.obviously increases of plasma creatine-kinase (CK) upon intramuscular injection in mice, suggesting that it may induce a dose-dependent myonecrosis. Histological studies also reveal morphological changes in mouse skeletal muscles, including extensive myonecrosis, hemorrhage and neutrophil infiltration in the treated animals. The myotoxic ability induced by Gln49-PLA2.can be partially inhibited by heparin.

Cloning and roles of goldfish maternal factor β-Catenin cDNA in embryonic development

Interaction between nucleus and cytoplasm has been focused in the field of animal embryonic development, in which study of maternal factors is required positively. b-Catenin, an important maternal factor in early embryo-genesis, has been analyzed in its expression pattern and func-tions in this paper. We have cloned goldfish b-Catenin cDNA gene and compared it with zebrafish b-Catenin cDNA. High homology was found in cDNA and in amino acid sequences between them, 93. (2.2./2.0}4 bp) and 98.5% (768/780 aa) respectively. The expression pattern of b-Catenin by in situ hybridization and the roles of b-Catenin on embryonic de-velopment by co-injection of anti-sense RNA and reporter gene, EGFP have been investigated in the whole process of goldfish embryonic development. The results suggest that b-Catenin presents dynamic distribution, mainly locates at body axis, dorsal tissues, head and tail structures after being fertilized. The loss of b-Catenin activity would cause serious destruction of embryo in dorsal tissues and in anteroposte-rior axes, and leads embryos to die before larva get hatched.

Application of three-dimensional discrete element face-to-face contact model with fissure water pressure to stability analysis of landslide in Panluo iron mine

Three-dimensional discrete element face-to-face contact model with fissure water pressure is established in this paper and the model is used to simulate three-stage process of landslide under fissure water pressure in the opencast mine, according to the actual state of landslide in Panluo iron mine where landslide happened in 1990 and was fathered in 1999. The calculation results show that fissure water pressure on the sliding surface is the main reason causing landslide and the local soft interlayer weakens the stability of slope. If the discrete element method adopts the same assumption as the limit equilibrium method, the results of two methods are in good agreement; while if the assumption is not adopted in the discrete element method, the critical φ numerically calculated is less than the one calculated by use of the limit equilibrium method for the sameC. Thus, from an engineering point of view, the result from the discrete element model simulation is safer and has more widely application since the discrete element model takes into account the effect of rock mass structures.

BIFURCATION OF THE NORTH EQUATORIAL CURRENT DERIVED FROM ALTIMETRY IN THE PACIFIC OCEAN

The gridded （1/3.＊1/3.） altimetry data from October 1992.through December 2.04 were analyzed to study the seasonal and interannual variabilities of the bifurcation of the North Equatorial Current （NEC） at the surface in the western North Pacific Ocean. Calculations show that on annual average the bifurcation occurs at about 13.4°N at the surface. The geostrophic flow derived from Sea Surface Height （SSH） data shows that the southernmost latitude of the NEC bifurcation at the surface is about 12.9°N in June and the northernmost latitude is about 14.1°N in December. Correlation analyses between the bifurcation latitude and the Southern Oscillation Index （SOl） suggest that the bifurcation latitude is highly correlated with the E1 Nino/Southern Oscillation （ENSO） events. During the E1 Nino years the bifurcation of the NEC takes place at higher latitudes and vice versa.

We report results of the H2.O and H110α survey toward 2.1 UCHII regions using the Urumqi 2.m radio *** obtained 3. new H2.O detections,and H110α was simultaneously detected in eight of *** H110α was detected in another UCHII *** calculated kinematic distances of nine UCHII regions with the detected H110α and resolved the kinematic distance ambiguity for six of *** detection rate of H2.O of our observation was 13.2.,which is low compared with one of the other *** possible reason is that the sensitivity of our telescope is relatively low.

An efficient quantum secret sharing protocol with orthogonal product states in the 3.3.Hilbert space is presented. The particles in the orthogonal product states form two particle sequences. One sequence is sent to Bob and the other is sent to Charlie after rearranging the particle orders. With the help of Alice, Bob and Charlie make the corresponding local measurement to obtain the information of the or- thogonal product states prepared. This protocol has many distinct features such as great capacity and high efficiency.

Suprathermal particle events observed by WIND spacecraft in interplanetary space during 1995―1999 and their classification

Fifty-five suprathermal particle events were selected from WIND observations between 1995 and 1999. Based on systematic analysis on the observational characteristics of these events a two-parameter (the rising time and the flux ratio of electrons to protons in each event) classification method was proposed to classify these events. The three clas-sified classes are (1) impulsive electron events with the flux ratio of electrons to protons being bigger than 1 and rising time being shorter than 2.0 min, (2. impulsive proton events with the flux ratio being smaller than 1 and rising time being shorter than 2.0 min, and (3. gradual proton events with the flux ratio being smaller than 1 and the rising time being longer than 2.0 min. In the past, "impulsive solar electron events" were under in-tense research. However, because the selection standards of their velocity dispersions or pitch-angle distributions were inadequate, statistical surveys of selected events were dif-ferent from each other and even some conclusions were not consistent with the theory, for example, the relation of type-III solar radio bursts to the "impulsive solar electron events". The first class of impulsive electron events are associated with type-III radio bursts and with clear velocity dispersions; therefore they ought to originate from the Sun. The second class of the events, which have short continuance time and usually are not associated with type-III radio bursts and without velocity dispersion, are still far away from inter-planetary shocks and most of them do not one-to-one correspond to corrotating interact-ing regions (CIRs); such events are possible results of local interplanetary magnetic field reconnection or electromagnetic disturbances. Finally, about 2.3.gradual proton events of the third class occur with interplanetary shocks, the delay times of which are almost equal to the rising time. Some of these events can be understood as particle accelerations by shocks.
