Objective To establish and optimize the proteomic analysis of protoscoleces-specific antigens from Echinococcus granulosus. To provide a foundation for identifying specific antigens in the soluble proteins of E. granulosus protoscoleces for further research. Methods Brood capsules were collected aseptically from fertile E. granulosus cysts from the livers of an infected patient. The fertile E. granulosus cysts were fractured, and protoscoleces were collected by centrifugation. The soluble proteins of protoscoleces were acquired using the 2D Quant kit according to the manufacturer＇s instructions. We employed two-dimensional electrophoresis （2-DE） combined with immunoblot assay （Western blot） to analyze the soluble components of E. granulosus protoscoleces antigens. The 2-DE and immunoblot maps obtained were analyzed with PDQuest 8.0 image analysis software. Results About 23. soluble protein spots were identified with Coomassie-stained gels. Most of the proteins had a molecular weight of 16 000 Da to 117 000 Da, and an isoelectric point value of 3.0 to 10.0. 2-DE immunoblot was conducted and 57 specific antigen spots were observed, among which 23.spots were identified. Conclusion 2-DE combined with Western blot is the key to successful proteomic analysis and presents a new possibility for searching the specific E. granulosus protoscoleces antigens.

In order to research the possibility of digital close-range photogrammetric surveying in small scale physical simulation experiment, physical model coinciding with engineering practice was constructed based on similar theory. The datum processing method and surveying precision of digital close-range photogrammetric were analyzed. And the function relationship between overburden subsidence factor qr and the ratio z/H of stratum horizon z and mining depth H was researched. The results show that surveying points position mean error along horizontal direction is ±0.13. mm and vertical direction is ±0.192 mm. Therefore, multi-taking station cross direction digital close-range photogrammetric can completely satisfy the precision need of physical simulation experiment. And the empirical formula can be utilized to represent evolution law of stratum subsidence factor.

MiaoXAM2.5X-50X Ultrahigh Precision Surface 3. Profiler is introduced to research the relations be- tween thickness of UHMWPE’s wear debris and positive pressure of tribological system. Research re- sults show that thickness of wear debris owns evident classification, and the first grade thickness math model and the second grade thickness math model are built. By the second grade thickness math model, the true force condition in the tribological system can be gotten, and the model can be used as a tool to modify simulating parameters of the tribological system, and it is an important inspecting method to the airtight system which cannot be examined in real time. The first grade thickness math model can describe the true working load and surface condition of work piece in the tribological sys- tem, and is an effective description method for the running condition of frictional pairs. The research results at different lubricating conditions testify the relations between thicknesses of wear debris and positive pressure of the tribological system. The fractal relation between maximal thickness of wear debris and positive pressure is discussed. The conceptions of the immutability principle and charac- teristic granularity of wear debris thickness are brought forward and discussed. In conclusion, as a stable tribological parameter owning mechanical theory basis, wear debris thickness will play an im- portant part in fault diagnosis, especially for the airtight system such as artificial joint.

Based on the irregular surface of natural cartilage configurations, the tribological characteristics of coupling motion between natural cartilage pairs were investigated by the variation of rotated offset and velocity. Contact displacement, contact load, friction force and coefficient of friction from natural cartilage pairs were measured by a UMT-2 testing machine. In order to obtain the steady part and wavy part, the contact load and contact displacement were decompounded by the five-point sliding average method. These results showed that the contact load was composed of steady load and wavy load. And the contact displacement was composed of cartilage deformation and surface outline. The steady load was similar to exterior load while natural cartilage deformation increased nonlinearly with the sliding time. Wavy load had the correlative coefficient 0.23. with surface outline due to the irregular surface of friction configurations. The coefficient of friction from cartilage friction configurations was less influenced by smaller magnitude in rotated parameters. A few strange summits were obtained in the area of coef-ficient of friction, and significantly related with local minimal values corresponding to the contact loads. The rotated offsets had clear impact on contact load and a slight impact on contact displacement. Larger offset produced higher wavy magnitude of the contact load. The rotated velocities played an evident role in cartilage deformation, but had a less influence on contact load. Lower velocity brought larger cartilage deformation.

Solid backfilling mining can reduce the buildings' damage caused by mining greatly. The reduction of subsidence value, the slow advancing speed and the subsidence caused by backfilling body compaction are the main reasons that solid backfilling mining velocity decreases significantly. Based on the research of mechanism, some principles on subsidence control of solid backfilling mining under buildings were proposed. The equivalent mining height was designed according to the fortification criteria of buildings and their attachment structures, which enables the ground movement and deformation caused by mining to be less than the corresponding fortification criteria.

Solid backfilling mining technology, which decreases the height of fissure zone and caving zone, and alleviates the subsidence, is a new technology for farmland conservation. Based on the situation analysis of farmland destruction in mining area, three ways for farmland protection were proposed. In order to improve the feasibility of this technology, the limited filling materials should be used to increase resources recovery ratio, and then the surplus materials could be backfilled into goaf. An index, namely farmland conservation ability, was put forward to optimize the ways for farmland conservation. At last, the Wanbei coal mine was taken as a case for farmland conservation. It was shown that 3.40 t dull coal was substituted and 52 hm2 farmland was conserved by solid backfilling mining in this coal mine.

This paper refers to Clarke generalized gradient for a smooth composition of max-type functions of the form: f(x) = g(x, maxj∈J1 f1j(x),''', maxj∈Jm fmj(x)), where x ∈Rn, Ji, i = 1,''',m are finite index sets, g and fij,j ∈ Ji, i = 1,... )m, are continuously differentiable on Rm+n and Rn, respectively. In a previous paper) we proposed an algorithm of finding an element of Clarke generalized gradient for f, at a point. In that paper, finding an element of Clarke generalized gradient for f, at a point, is implemented by determining the compatibilities of systems of linear inequalities many times. So its computational amount is very expensive. In this paper) we will modify the algorithm to reduce the times that the compatibilities of systems of linear inequalities have to be determined.

Based on the prerequisite that the earliest acid rock formed because of the initial global expanding tectonic processes, by combining it with other geological limitation conditions, and starting from the lithosphere elastic mechanics, the following is defined : the earth expansion started at about 43.0 Ma ago, the average increasing rate of the earth radius is 0.40 mm/a, the initial expanding earth radius is 4651 km, and the initial global expansion of tectonic processes happened at 4183.7 Ma ago. From the natural and attenuation evolution condition of the earth and terrestrial planets, through the limit condition got from the combination of the mathematics logic and the geological significance, the limited expanding earth evolution model is established as follows:in which R0= 4.651×106 m, A =1.86×106 m, β = -6.01?10-10 a and ts=3.10 a. From this model the maximum radius Rmax of the limited expanding earth evolution should be 6511 km. The earth’s radius increasing rate is about 0.1 mm/a at present.
