Role of arachidonic acid in hyposmotic membrane stretch-induced increase in calcium-activated potassium currents in gastric myocytes

Aim: To study effects of arachidonic acid （AA） and its metabolites on the hyposmotic membrane stretch-induced increase in calcium-activated potassium currents (IKCa) in gastric myocytes. Methods: Membrane currents were recorded by using a conventional whole cell patch-clamp technique in gastric myocytes isolated with collagenase. Results: Hyposmotic membrane stretch and AA increased both IK（Ca）and spontaneous transient outward currents significantly. Exogenous AA could potentiate the hyposmotic membrane stretch-induced increase in IK（Ca）The hyposmotic membrane stretch-induced increase inK（Ca）was significantly suppressed by dimethyleicosadienoic acid （100 μmol/L in pipette solution）, an inhibitor of phospholipase A2. Nordihydroguaiaretic acid, a lipoxygenase inhibitor, significantly suppressed AA and hyposmotic membrane stretch-induced increases in IK（Ca）. External calcium-free or gadolinium chloride, a blocker of stretch-activated channels, blocked the AA-induced increase in IK（Ca）significantly, but it was not blocked by nicardipine, an L-type calcium channel blocker. Ryanodine, a calcium-induced calcium release agonist, completely blocked the AA-induced increase inK（Ca）; however, heparin, a potent inhibitor of inositol triphosphate receptor, did not block the AA-induced increase in IK（Ca）. Conclusion: Hyposmotic membrane stretch may activate phospholipase A2, which hydrolyzes membrane phospholipids to ultimately produce AA; AA as a second messenger mediates Ca2+influx, which triggers Ca2+-induced Ca2+release and elicits activation of IK（Ca）in gastric antral circular myocytes of the guinea pig.

Calculating Models of Mass Action Concentrations for Ni-Mn and Co-Mn Melts and Optimization of Their Thermodynamic Parameters

According to phase diagrams, measured activities as well as the coexistence theory of motallic melts structure involving compound formation, the calculating models of mass action concentrations for Ni-Mn and Co-Mn melts are formulated and their thermodynamic parameters are optimized. As a result, the calculated mass action concentrations agree well with the corresponding measured activities, showing that these models can reflect the structural characteristics of both Ni-Mn and Co-Mn melts.

According to the results of research on the thermodynamic propelles of Fe-Cr, Fe-P and Cr-P melts, the measured achvihes of Fe-Cr-P melts from reference sources as well as the coexistence theory of metallic melts structure involving compound formation, a calculating model of the mass action concentrations for Fe-Cr-P melts has been formulated and some of its thermodynamic parameters have been optimized. The calculated mass action concentrations agree with the measured achvities, which shows that this model can reflect the structural reality of Fe-Cr-P melts.

In single rat pancreatic βcells, using fura-2 microfluorometry to measure [Ca2+] response upon different stimuli, the ways of calcium regulation have been studied. When the extracellular calcium concentration was 2.5 mmol/L, either 60 mmol/L KCI, 20 mmol/L D-glucose or 0.1 mmol/L tolbutamide induced increase in [Ca2+]. Such increase in [Ca2+] was absent when the same stimuli were applied under zero extracellular calcium. These results indicate that the increase of [Ca2+] is induced by the activation of voltage-dependent calcium channels in p celis. The manifold forms of [Ca2+] change induced by glucose imply that the effects of glucose are complex. 5 mmol/L caffeine or 5 mmol/L MCh increase the [Ca2+], which is independent of the external calcium, suggesting that [Ca2+] can be regulated by Ca2+ release from not only the IP3.sensitive but also the ryanodine sensitive calcium stores in p celis. In the paper we are concerned with double S-breaking cubic turning points of two-parameter nonlinear problems in the presence of Z(2)-symmetry. Three extended systems are proposed to determine double S-breaking cubic turning points. We show that there exist two kinds of singular point path passing through double S-breaking cubic turning point, One is the simple quadratic turning point path, the other is the pitchfork bifurcation point path.

Based on the phase diagrams, reliable reference experimental data and the coexistence theory of metallic melts structure involving compound formation, calculating models of mass action concentrations for Fe-P and Cr-P melts have been formulated. At the same time, some of their thermodynamic parameters have been optimized. The calculated results not only agree well with the measuredvalues, but also obey the mass action law rigorously, this in turn shows that these models can reflect the structural characteristics of corresponding melts.

Microstructural Features During Strain Induced Ferrite Transformation in 08 and 20Mn Steels

The microstructure evolution during strain induced ferrite transformation was followed in thermal-simulation tests of clean 08 and 20Mn steels. The influences of carbon equivalence and initial austenite grain size on ferrite grain refinement and the volume fraction of ferrite during straining were inspected. The results revealed that the accelerating effect of ferrite transformation by strain was increased as the carbon equivalence decreased. However, finer ferrite grains were obtained at higher carbon content. At strain of similar to1.5 ferrite grains less than 3.mum and 2 mum can be obtained in 08 and 20Mn steels respectively. Whereas the ferrite grain refinement in 08 steel was due to both effects of strain induced transformation and ferrite dynamic recrystallization, that in 20Mn was mainly due to strain induced transformation. Heavy strain can produce fine ferrite grains in coarse austenite grained 08 steel, but it would lead to band microstructure in coarse austenite grained 20Mn.

The unified solutions are presented for stresses and displacements around a circular tunnel subjected to a hydrostatic stress *** rock mass is assumed to be elastic-brittle-plastic and governed by the Unified Strength *** displacements are obtained accounting for three different definitions for elastic strains and different Young's modulus in the plastic *** unified solutions obtained in this paper are especially versatile in reflecting the intermediate principal stress effect to different extents for different *** conventional solutions,based on the Mohr-Coulomb failure criterion and the Generalized Twin Shear Stress yield criterion,are special cases of the present unified *** new unified solutions can compare with those computed by the latest generalized Hoek-Brown failure *** results obtained demonstrate the importance of the intermediate principal stress influence for the stresses and displacements *** effects of different definitions for elastic strains and different Young's modulus in the plastic zone on the displacements are significant.
