AIM: To study the epitope distribution of hepatitis G virus (HGV) and to seek for the potential recombinant antigens for the development of HGV diagnostic reagents. METHODS: Fourteen clones encompassing HGV gene fragments from core to NS3.and NS5 were constructed using prokaryotic expression vector pRSET and (or) pGEX, and expressed in ***. Western blotting and ELISA were used to detect the immunoreactivity of these recombinant proteins. RESULTS: One clone with HGV fragment from core to E1 (G1), one from E2 (G3.), three from NS3.(G6, G61, G7), one from NS5B (G821) and one chimeric fragment from NS3.and NS5B (G61-821) could be expressed well and showed obvious immunoreactivity by Western blotting. One clone with HGV framment from NS5B (G82) was also well expressed, but could not show immunoreactivity by Western blotting. No obvious expression was found in the other six clones. All the expressed recombinant proteins were in inclusion body form, except the protein G61 which could be expressed in soluble form. Further purified recombinant proteins G1, G3., G61, G821 and G61-821 were detected in indirected ELISA as coating antigen respectively. Only recombinant G1 could still show immunoreactivity, and the other four recombinant proteins failed to react to the HGV antibody positive sera. Western blotting results indicated that the immunoactivity of these four recombinant proteins were lost during purification. CONCLUSION: Core to E1, E2, NS3.and NS5 fragment of HGV contain antigenic epitopes, which could be produced in prokaryotically expressed recombinant proteins. A high-yield recombinant protein (G1) located in HGV core to E1 could remain its epitope after purification, which showed the potential that G1 could be used as a coating antigen to develop an ELISA kit for HGV specific antibody diagnosis.

The Fourier finite-difference(FFD) method is very popular in seismic depth *** its straightforward 3. extension creates two-way splitting error due to ignoring the cross terms of spatial partial *** correction schemes,either in the spatial domain by the implicit finite -difference method or in the wavenumber domain by phase compensation,lead to substantially increased computational costs or numerical difficulties for strong velocity *** propose compensating the two-way splitting error in dual domains,alternately in the spatial and wave-number domains via Fourier ***,we organize the expanded square-root operator in terms of two-way splitting FFD plus the usually ignored cross ***,we select a group of optimized coefficients to maximize the accuracy of propagation in both inline and crossline directions without yet considering the diagonal ***,we further optimize the constant coefficient of the compensation part to further improve the overall accuracy of the operator. In implementation,the compensation terms are similar to the high-order corrections of the generalized-screen method, but their functions are to compensate the two-way splitting error rather than the expansion *** experiments show that optimized one-term compensation can achieve nearly perfect circular impulse responses and the propagation angle with less than 1%error for all azimuths is improved up to 60°from 3.°.Compared with traditional single-domain methods,our scheme can handle lateral velocity variations(even for strong velocity contrasts) much more easily with only one additional Fourier transform based on the two-way splitting FFD method,which helps retain the computational efficiency.

Three-dimensional velocity images of the crust and upper mantle beneath western china are obtained through seismic tomography by using first P wave arrival times of local/regional and teleseismic events recorded in chinese and Kyrgyzhstan seismic *** shallow depths,most mountain belts and tectonically elevated areas are imaged as higher velocities,relative to adjacent sediment-filled basins and foreland depressions with lower *** mid-lower crust depths,the Kyrgyz Tian Shan is underlain by lower velocity material and a distinct low velocity zone in the south-north direction is observed between the eastern and western parts of the chinese Tian *** Moho of the southern Tarim basin has a greater depth over 50 km in front of the western Kun Lun,compared to the northern and western Tarim *** heterogeneity within the upper mantle is apparent at depths from 71 km to 571 *** uppermost mantle beneath the Kyrgyz Tian Shan,Altay,Qi Lian and eastern Kun Lun is characterized by low velocity material that is probably related to the upwelling of hot *** lithospheric thickness varies from 120-170 km beneath the chinese Tian Shan and northern Tibetan plateau to 200-250 km beneath the Tarim *** coupling pattern of the chinese Tian Shan with adjacent provinces can be classified into the embedding,detaching and sinking,and underthrusting of lithospheric *** addition,the lithosphere of the northern edge of the Tibetan plateau seems to be thickened,in contrast to the central part of the northern plateau. North-dipping low velocities exist in the upper mantle between the Tibetan plateau and provinces to its north,which might indicate mantle flow from the Tibetan plateau towards surrounding areas, but it has been blocked by the thick Tarim lithosphere.

FINITE DIFFERENCE METHOD WITH NONUNIFORM MESHES FOR QUASILINEAR PARABOLIC SYSTEMS

The analysis of the finite difference schemes with nonuniform meshes for the problems of partial differential equations is extremely rare even for very simple problems and even for the method of fully heuristic character. In the present work the boundary value problem for quasilinear parabolic system is solved by the finite difference method with nonuniform meshes. By using of the interpolation formulas for the spaces of discrete functions with unequal meshsteps and the method of a priori estimation for the discrete solutions of finite difference schemes with nonuniform meshes, the absolute and relative convergence of the discrete solutions of the finite defference scheme are proved. The limiting vector function is just the unique generalized solution of the original problem for the parabolic system.

Results of a multiple,isotopic,chemical and mineralogical analysis of loess from the three northwestern inland basins(including the Junggar Basin,the Tarim Basin and the Qaidam Basin) and the Loess Plateau region of china are *** suggest a qualification of the conventional views that the three northwestern basins were the important source areas of the Loess Plateau and that the sand deserts were the primary single source of the chinese *** is argued that the gobi(stony desert) in southern Mongolia and the adjoining gobi and sand deserts(including the Badain Jaran Desert,Tengger Desert,Ulan Buh Desert,Hobq Desert and Mu Us Desert) in china, rather than the three inland basins,are the dominant source areas of the Loess ***, although these gobi and sand deserts are regarded as the main source regions,they serve as dust and silt holding areas rather than dominant *** mountain processes(including glacial grinding,frost weathering,salt weathering,tectonic processes,and some fluvial comminution) in the Gobi Altay Mts.,Hangayn *** the Qilian *** played an important role in producing the vast amounts of loess-sized material for forming the Loess Plateau.
