Aim:To investigate the direct effects of resistin and resistin-binding peptide（RBP）on lipid metabolism and endocrine function in adipose ***:Ratwhite adipose tissue was cultured in vitro and incubated for 24 h with 3. ng/mLrecombinant rat resistin protein（rResistin）or combined with RBP of varyingconcentrations(1×1-12mol/L,1×10-10mol/L,1×10-8mol/L).Free fatty acids（FFA）released into medium was measured by a colorimetric *** levels of proteinsecretion and mRNA expression of TNF-α and adiponectin were detected by ELISAkit and RT-PCR ***:The levels of FFA released into mediumwere significantly increased after 24 h of exposure to rResistin,but significantlydecreased after RBP was applied,although there was no difference between the *** protein level and gene expression of TNF-αin adipose tissuewere significantly increased after 24 h of exposure to rResistin,but only obviouslydecreased after incubated with 1×10-8mol/L *** levels of protein secretionand mRNA expression of adiponectin in adipose tissue were significantly de-creased after 24 h of exposure to rResistin,but increased after incubated with RBPwith the higher ***:RBP can effectively antagonize therole of resistin on the lipid metabolism and endocrine function of adipose tissue.

AIM: To explore the influence of angiostatin up-regulationon the biologic behavior of gastric cancer cells in vitro andin vivo, and the potential of angiostatin gene therapy in thetreatment of human gastric ***: Mouse angiostatin cDNA was subcloned intothe eukaryotic expression vector pcDNA3.1(+) and identifiedby restriction endonucleases digestion and sequencing. Therecombinant vector pcDNA3. 1(+)-angio was transfected intohuman gastric cancer cells SGC7901 with liposome andparalleled with the vector control and the mock *** transcription and protein expression wereexamined by RT-PCR and Western blot in the stable celllines selected by G418. Cell proliferation and growth in vitroof the three groups were observed respectively undermicroscope, cell number counting and FACS. The cellsoverexpressing angiostatin, vector transfected and untreatedwere respectively implanted subcutaneously into nude *** 3.days the size of tumors formed was measured, andmicrovessel density count (MVD) in the tumor tissues wasassessed by immunohistochemistry with the primary anti-vWF ***: The recombinant vector pcDNA3.1(+)-angio wasconfirmed with the correct sequence of mouse angiostatinunder the promoter CMV. After 3. d of transfection andselection with G418, macroscopic resistant cell clones wereformed in the experimental group transfected with pcDNA3.1(+)-angio and the vector control. But no untreated cellssurvived in the mock control. Angiostatin mRNAtranscription and protein expression were detected in theexperimental group. No significant differences wereobserved among the three groups in cell morphology, cellgrowth curves and cell cycle phase distributions in ***, in nude mice model, markedly inhibitedtumorigenesis and slowed tumor expansion were observedin the experimental group as compared with the controls,which was paralleled with decreased microvessel density inand around tumor tissues (P＜0. 05).CONCLUSION: Angiostatin does

Aim:To investigate the blood pressure(BP),baroreflex sensitivity(BRS),andorgan damage in hybrids of spontaneously hypertensive rats and ***:Spontaneously hypertensive rats and Sprague-Dawley rats werecrossbred,and the F1hybrids were inbred randomly to produce an *** the age of 52 weeks,the F1and F2hybrids were tested to determine BP and BRSin a conscious *** examinations were carried out after BPrecording and BRS ***:BP and BRS were not different in F1and *** was inversely related to systolic BP(SBP)in male,female,or wholepopulations of ***,BRS values were one-third determined bySBP level(the determinant coefficient was 0.3.6).The indexes for left ventricularhypertrophy,aortic hypertrophy,and renal damage were all positively related toBP,and negatively related to *** multiple-regression analysis,left ventricularand aortic hypertrophy and glomerulosclerosis score were all most significantlyassociated with lower BRS and higher systolic *** contribution of BRS to leftventricular and aortic hypertrophy and glomerulosclerosis was greater than thatof ***:The present work with hybrid rats demonstrated quantita-tively that the BRS value was one-third determined by SBP *** BP leveland BRS value contributed greatly to the hypertensive organ ***,the contribution of BRS to the hypertensive organ damage was greater than thatof BP level in these rats.

AIM: To investigate the regulatory effects of vasonatrin peptide (VNP) on the expression of C-type natriuretic peptide receptor (NPR-C) in cultured neonatal rat cardiac myocytes and fibroblasts. METHODS: Quantitative RT- PCR was undertaken to evaluate the levels of NPR-C mRNA and radioimmunoassay was used to determine the formation of intracellular cGMP. RESULTS: Twenty-four hours hypoxic exposure increased the level of NPR-C mRNA in cardiomyocytes, while did not alter the expression of NPR-C in cardiac fibroblasts. VNP (1×10-8-1×10-6 mol/L) reduced the levels of NPR-C mRNA in cardiac myocytes induced by hypoxia in a concentration-dependent manner, and with high concentration (1×10-6mol/L) also decreased the expression of NPR-C in cardiac fibroblasts and air-control cardiac myocytes. The inhibitory effects of VNP on the expression of NPR-C was mimicked by 8- bromo-cGMP 1×10-6 mol/L (a membrane permeable analog of cGMP). VNP (1×10-8-1×10-6mol/L) increased the formation of intracellular guanosine-3.,5'-cyclic monophosphate (cGMP) in both cardiac myocytes and fibroblasts. HS-142-1, the particulate guanylyl cyclase-coupled receptor antagonist, partially abrogated the above effects of VNP. CONCLUSION: Hypoxic exposure for 24 h up-regulated the expression of NPR-C in cultured neonatal rat cardiac myocytes. VNP decreased the expression of NPR-C in cardiac myocytes and fibroblasts under both air- control and hypoxic condition, which was at least partially mediated by guanylate cyclase linked natriuretic peptide receptors through increasing the intracellular cGMP.

Human placenta-derived stem cells (hPDSCs) were isolated by trypsinization and further induced into cartilage cells in *** engineered cartilage was constructed by combining hPDSCs with collagen sponge and the cartilage formation was observed by implantation into nude *** showed that hPDSCs featured mesenchymal stem cells and maintained proliferation in vitro for over 3. passages while remaining *** results indicated that hPDSCs have the potential to differentiate into functional cartilage cells in vitro when combined with collagen sponge,which provided experimental evidence for prospective clinical application.

AIM:To clarify the association between CYP2E1 PstⅠ/RsaⅠ polymorphism and susceptibility to colorectal ***:A meta-analysis based on 10 eligible casecontrol studies involving 4979 cases and 6012 controls was carried out to summarize the data on the association between CYP2E1 RsaⅠ/PstⅠ polymorphism and colorectal cancer ***:In comparison of the homozygote c2c2 and c2 carriers(c1c2 + c2c2) and the homozygous wild-type genotype(c1c1),no association was found between CYP2E1 RsaⅠ/PstⅠ polymorphism and colorectal cancer risk [odds ratio(OR)=1.24(95% CI:0.93.1.66) for c2c2;OR = 1.02(95% CI:0.88-1.19) for c2 carriers].In stratif ied analysis,Caucasians with c2c2 homozygote appeared to have an increased risk of colorectal cancer(OR=2.67,95% CI:1.03.6.89,P=0.043.,no signif icant associations were found in other ***:c2c2 homozygote of CYP2E1 PstⅠ/RsaⅠ polymorphism may be associated with the increased risk of colorectal cancer in Caucasians,which needs further investigations.

The present study examined cardiac myocytecontractile and Ca2+ transient responses to insulin duringsimulated ischemia/reperfusion (I/R) and furtherinvestigated the role of protein kinase B (Akt) in the insulin-induced inotropic effect. Ventricular myocytes wereenzymatically isolated from adult Sprague-Dawley rats and perfused with Tyrode solution while electrically field-stimulated. Simulated I/R was induced by perfusing the cells with chemical anoxic solution including sodiumcyanide-sodium lactate for 15 min followed by reperfusionwith normal oxygenated Tyrode solution with or withoutinsulin. It is found that insulin only at concentration as high as 10 IU/L could increase cell shortening (16±5%, P 3.-kinase (PI3.kinase), significantly inhibited both Akt phosphorylation and thepositive inotropic response to insulin in the I/Rcardiomyocytes. These results suggest that insulin exertsdirect positive inotropic effect by increasing Ca2+ transient of cardiomyocytes, which is enhanced in the pathologicalcondition of I/R. Akt activation plays an important role inthe insulin-induced improvement of myocyte contractile function following I/R.

Clinical Observation on Preservation of Lens Anterior Capsule in Reduction of Complications Associated with Silicone Oil

Purpose: To observe the effect of preservation of anterior capsule on the incidence of complications associated with silicone oil.Methods: 82 patients(82 eyes) accepted trans pars plana vitrectomy(TPPV) combined with lensectomy of whom , 3. eyes with preservation of anterior capsule(PAC) and 52eyes with no preservation of anterior capsule (NPAC). Analysis of the incidence of complications associated with use of oil.Results:The incidence rate was 50.0 % in NPAC group, and 23.3.% in PAC group( P ＜ 0. 025 ). There were seconda'y glaucoma (21. 1% ), band keratopathy ( 13.5 % ) and corneal decompensation(9.6 % ) in NPAC group, while there was none of them in PAC group.Conclusions: Preservation of anterior capsule is an eftective method to reduce the complications associated with silicone oil.
