Synthesis of GoldMag particles with assembled structure and their applications in immunoassay

Micrometer-sized Fe3.4 particles and nano-sized gold particles were first synthesized by methods of self-aggregation of surface-chemically modified Fe3.4 nanoparticles and citrate reduction of the Au3. to Au0, respectively. Interaction between these two types of particles resulted in the assembly of nano-sized gold particles on the surface of the micrometer-sized Fe3.4 particles, forming an assembled structure with the Fe3.4 core particles around which are attached nano-sized gold parti- cles. The Fe3.4/Au structure is named GoldMag particles with assembled structure. The synthetic process, structure, and magnetic property of the GoldMag particles were analyzed. GoldMag particles with assembled structure have an irregular shape, rough surface with a diameter of 2―3.μm. These particles exhibit the superparamagnetic property with saturated magnetization of 41 A·m2/kg. In a single step, antibodies could be readily immobilized onto the surface of the particles with a high binding capacity. The GoldMag particles can be used as a novel carrier in immunoassays. The maximum quantity of human IgG immobilized onto GoldMag particles was 3.0 μg/mg. In order to validate the quality of the GoldMag particles as immunoassay carriers, an immunoassay system was used. The relative amount of immobilized human IgG was measured by HRP-labeled anti human IgG. The coefficient of variation within parallel samples of each group was below 6% and the coefficient of variation of means between five groups carried out separately was below 7%. Based on the sandwich method, the Hepatitis B surface antigen (HBsAg) and interleukin-8 (IL-8) were also analyzed by qualitative and quantitative detection, respectively. The result indicated that the GoldMag particles with assembled structure were an ideal carrier in immunoassay.

In an attempt to improve immune responses and protective efficacy,we constructed tworecombinant bacille Calmette-Guérin(rBCG)strains expressing an 85B antigen(Ag85B)and early secretedantigenic target-6 kDa antigen(ESAT6)of M vcobacterium tuberculosis(MTB)fusion *** rBCGstrains have the same protein insertion but in a different order(Ag85B-ESAT6 and ESAT6-Ag85B).Thecultured supernatant of rBCG strains and the sera from the mice immunized with the fusion protein Ag85B-ESAT6 or ESAT6-Ag85B formed a band with a fraction size of 3. kDa,equalivalent to the sum of Ag85B *** weeks after BALB/c mice were immunized with BCG or rBCG,spleen lymphocytes showedsignificant proliferation in response to culture filtrate protein of *** with the BCG group,micevaccinated with rBCG elicited a high level increase of immunoglobulin G antibodies to culture filtrate proteinin the *** γ-interferon levels in the lymphocyte culture medium supernatants increased remarkably inthe rBCG1 group,significantly higher than that of the BCG immunized group(P3.Rv and a dramatic reduction in the numbers ofMTB colony forming units in the spleens and lungs was observed in the two rBCG immunization *** these rBCG strains were more immunogenic,their protective effect was comparable to the classicalBCG strain,and there were no significant differences between two rBCG groups(P>0.05).

Expression of the somatostatin receptor family mRNAs inlung cancer

To investigate the characteristics of expression and distribution of 5 subtypes of somatostatin receptors (SSTR1～5) in lung cancer, in situ hybridization was used to examine the expression patterns of SSTR mRNAs in 21 cases of different pathologic types of lung cancer tissues with [a-3.S]dATP labeled oligonucleotides of the 5 SSTR subtypes as probes. Additionally, Leica Q-500 image analysis processing system was employed for the semi-quantitatively analysis of the hybridization signals. Patterns of SSTR1～5 expression in lung cancer tissues were found as follows. SSTR2 was prominent in small cell lung cancer (SCLC), whereas in non-small cell lung cancer (NSCLC) including the adenous cancer (Ad) and the squamous cancer (Sq), the expression of SSTR1 mRNA was stronger than that of the other 4 types. The expression density of SSTR1～5 in the NSCLC was higher than the SCLC (p <0.01). The expression patterns and densities of the SSTR subtypes showed heterogeneity in different pathologic types of lung cancer. The expressions of the SSTR mRNAs in both SCLC and NSCLC indicated the positive prospects for somatostatin analog(SSA)--oriented agents in the treatment of both types of the lung cancer.

Reduced cell death by inosine pretreatment after photochemically induced cerebral ischemia in adult rats

Protective effect of inosine against cell death after photochemically induced focal cerebral ischemia was investigated in adult rats. Intraperitoneal injection of inosine was started 24 h before operation and continued till the animals were *** TUNEL method and Nissl staining were used to identify apoptosis and necrosis,***-positive cells were located in the transitional zone between the necrotic core and normal *** mean maximum area of the necrotic cores and mean number of TUNEL-positive cells 24 h after operation were reduced significantly in the inosine-treated rats when compared with those in the saline-treated control animals. We suggest that inosine can significantly reduce both necrosis and apoptosis following cerebral ischemia induced by photothrombosis in adult rats.

Isoprenaline enhances local Ca2+ release in cardiac myocytes

Aim:Contraction of cardiac myocytes is controlled by the generation and ampli-fication of intracellular Ca2+*** key step of this process is the couplingbetween sarcolemma L-type Ca2+channels（LCCs）and ryanodine receptors（RyRs）in the sarcoplasmic reticulum（SR）.β-Adrenergic stimulation is an important regu-latory mechanism for this coupling *** the details underlied the globallevel,which require local Ca2+release study are still *** present study isto explore the effects of β-adrenergic stimulation on local Ca2+***:Using confocal microscopy combined with loose-seal patch-clamp approaches,effects of isoprenaline(1 μmol·L-1),a β-adrenergic agonist,on local SR Ca2+re-lease triggered by Ca2+influx through LCCs in intact rat cardiac myocytes ***:Isoprenaline increased the intensity of ensemble averagedlocal Ca2+transients,the peak of which displayed a typical bell-shaped voltage-dependence over the membrane voltages ranging from～-40mV to～+3.m ***-ther analysis showed that this enhancement could be explained by the increasedcoupling fidelity（which refers the increased probability of RyRs activation upondepolarization）,and the increased amplitude of evoked Ca2+sparks(due to moreCa2+releases through local RyRs).In addition,isoprenaline decreased the firstlatency,which displayed a typical"U"-shaped voltage-dependence,showing theavailable acceleration and synchronization of β-adrenergic stimulation on intrac-ellular calcium ***:Isoprenaline enhances local Ca2+release incardiac *** results underscore the importance of regulation of β-adrenergic stimulation on local intermolecular signals between LCCs and RyRs inheart cells.
