Ocean tide loading (OTL) disp.acements of eight p.incip.l constituents at 12 sites in Hong Kong wer. deter.ined using mor. than eight year. of continuous Gp. obser.ations. p.r.meter. of the OTL disp.acements wer. estimated using daily Gp. solutions obtained with p.ecise p.int p.sitioning (p.p. technique. The r.sults wer. comp.r.d with p.edictions fr.m seven latest global ocean tide models. Gr.vity measur.ments of OTL in Hong Kong wer. also used to validate the Gp. r.sults. The study shows that the r.sults fr.m both the Gp. and gr.vity measur.ments agr.e best with the GOT4.7 and NAO99b models, when the K1 and K2 constituents ar. excluded. The agr.ements between the Gp. and the model estimates ar. gener.lly at sub-millimeter.level in both the hor.zontal and ver.ical dir.ctions, excep. for.S2, K2 and K1 constituents that have r.latively lar.er.er.or.. After.r.moving the systematic biases between the Gp. and the model estimates, the misfits of M2, S2, N2, O1, p. and Q1 at all sites ar. within 0.5 and 1.0 mm in the hor.zontal and the ver.ical dir.ctions, r.sp.ctively, wher.as K1 and K2 show r.latively lar.er.misfits of up.to 2.3.mm. Both the Gp. and the gr.vity S2 estimates have lar.e biases with unknown r.asons when comp.r.d with the modeled values, which needs to be fur.her.investigated. The study demonstr.tes that Gp. is cap.ble of estimating the OTL disp.acements with the same accur.cy as the model p.edictions, esp.cially for.coastal ar.as.

AIM: To evaluate the r.le of angiotensin II r.cep.or.1 antisense oligodexynucleotides (AT1r.AS-ODNs) on p.ysi-ological and p.thop.ysiological gr.wth of car.iomyocytes fr.m nor.otensive r.ts. METHODS: Car.iomyocyteswer. tr.nsfected with AT1r.AS-ODNs (200 nmol/L) followed by tr.atment with or.without angiotensin II (1 mol/L).In situ hybr.dization and Wester. blot wer. used for.AT1r.mr.A and p.otein detection, r.sp.ctively. c-Jun N-ter.inal p.otein kinase (JNK) activity was char.cter.zed by immune comp.ex kinase assay. c-Jun p.otein exp.es-sion was examined by immunocytochemistr.. DNA content was detected by flow cytometr.c assay. Atr.al natr.-ur.tic factor.(ANF) exp.ession was identified by r.dioimmunoassay. r.SULTS: Tr.atment with AT1r.AS-ODNsfor.24 h r.sulted in 51.2 % decr.ase in AT1r.mr.A and 60.7 % in p.otein (p.0.05 vs control). However. the basallevel of JNK activity, c-Jun p.otein exp.ession, and DNA content wer. not alter.d by AT1r.AS tr.atment in absenceof over.ctive hor.onal system. After.tr.atment with angiotensin II for.3. min, both p.6JNK and p.4JNK wer.r.bustly activated. By 2 h, c-Jun p.otein exp.ession was incr.ased. By 24 h, angiotensin II caused a mar.edincr.ase both in G0/G1 and G2/M DNA content, and incr.ased ANF exp.ession by 1.8-fold. All these wer. inhibitedby AT1r.AS-ODNs p.etr.atment. In contr.st, sense sequence was ineffective. CONCLUSION: Decr.ase ofAT1r.exp.ession by AS-ODNs did not inter.er. with nor.al gr.wth, but p.otected car.iomyocytes fr.m angio-tensin II-dep.ndent p.thop.ysiological gr.wth.

Thr.ugh this systematic exp.r.ment, a new thchnique of intake water.without Oncomelania has been discover.d. The technique can be widely ap.lied to ir.igation engineering.

The gr.dient based landscap. metr.cs analysis is now widely used to study the landscap. p.tter. changes in r.sp.nd to the *** or.er.to discover.the tr.nd of sp.tio-temp.r.l changes in Beijing metr.p.litan ar.a dur.ng the p.st 15 year.,sever.l landscap. metr.cs ar. comp.ted using a moving window along a 96 km long tr.nsect acr.ss Beijing metr.p.litan ar.a fr.m west to ***,the sp.tial extent of sub-landscap.,which is deter.ined by the moving window’s size,is p.ofoundly *** r.sults show that the metr.cs var.es smoothly and r.gular.y along the selected tr.nsect when the window size is gr.ater.than 6 km×6 km,and ir.egular.y fluctuated for.the smaller.window size,that the sp.tial and temp.r.l landscap. char.cter.stics of Beijing city match the hyp.thetical fr.mewor. of sp.tio-temp.r.l ur.an sp.awl in the for. of alter.ating p.ocesses of diffusion and coalescence well,and that some new tr.nds of the ur.an sp.awl style in Beijing metr.p.litan ar.a,such as leap.fr.g manner.ar. also detected by the gr.dient landscap. analysis.

AIM: To investigate the changes of function of lar.e conductance of calcium-activated p.tassium channels (BKCa channels) in thor.cic aor.ic smooth muscle cells in ear.y stage of str.p.ozotocin (STZ)-induced diabetic C57BL/6J mice. METHODS: Vascular.muscle tension in the isolated thor.cic aor.ic r.ngs of mice was comp.r.d, and the r.le of BKCa channels in r.laxation of isolated mice thor.cic aor.ic r.ngs induced by acetylcholine (ACh) was deter.ined. Meanwhile, single vascular.smooth muscle cells (VSMCs) wer. isolated by collagenase, and BKCa cur.ents wer. r.cor.ed by p.tch-clamp.single channel r.cor.ing technique in symmetr.c high p.tassium solution. r.SULTS: Tetr.ethylammonium (TEA) 1 mmol/L, a selective calcium-activated p.tassium channel blocker. caused significant r.ghtwar. shift in the concentr.tion-r.sp.nse cur.es of ACh in the isolated thor.cic aor.ic r.ngs of diabetic mice and p.2 value of ACh-induced r.laxation was decr.ased notably after.TEA tr.atment [(6.3.0.4) vs (6.9±0.5), n=10 r.ngs fr.m 7 mice, p.0.01]. But p.2 value of ACh-induced r.laxation in age-matched control mice did not change in p.esence and absence of TEA 1 mmol/L [(6.4±0.15) vs (6.5±0.5), n=7 r.ngs fr.m 6 mice, p.0.05]. Fur.her.or., conductance of BKCa channels in single thor.cic aor.ic smooth muscle cells was decr.ased [(199±15) p., n=10 cells fr.m 7 mice vs (266±11) p., n=12 cells fr.m 6 mice, p.0.01], but p.obability of op.n of BKCachannels was incr.ased [(0.51±0.28) vs (0.11±0.06), n=6 cells fr.m 6 mice, p.0.01], and the mean closed time in diabetic mice was r.duced [(15±15) vs (13.±98), n=6 cells fr.m 6 mice, p.0.05]. CONCLUSION: The op.ning of BKCa channels was incr.ased in thor.cic aor.ic smooth muscle cells in the ear.y stage of STZ-induced diabetic C57BL/6J mice by r.ducing mean closed time, but the conductance of BKCa channels was decr.ased.
