Under the prerequisite that the incidence of cancer or tumor in negative-control nude mice inoculated subcutaneously with primary feline or canine kidney cell cultures purified in vitro at passage 3.was 0 (0/22) and 0 (0/10), respectively. The incidence of the progressively-growing malignant tumor(MT) in positive-control nude mice inoculated subcutaneously with Hela cell cultures of KB, X, or NM20/X strain was 10/ 10, 25/25 and 5/51, respectively. The results showed that the incidence of tumor in nude mice with di-and hy-perploid YB strain of MDCK cell during 17 - 23.passages, with hyper- and hypoploid KA strain of MDCK cell during 6-8 passages, with hypoploid WB strain of MDCK cell on passage 6, with hyper-and hypoploid H strain of MDCK cell during 8-24 passages was 2/24, 6/10, 5/10 and 10/15, respectively. The chromosomal analysis results showed that the ratio of difference in the rate of modal chromosome number between high(mcs + n) and lowest (mcs)passages was not more than 5 - 15% and the structure aberrations was generally 0 -3.%. These results proved that the genetic characteristics of chromosomal number of cell lines determines their tumorigenicity, but it is species-specific. MDCK line has tumorigenicity no matter what its chromosome kary-otype is, at least it has very low tumorigenicity even when its modal chromosome number is hypoploid. It is thus evident that MDCK cell of WB or H strain can be approved as substrate for the preparation of attenuated viral vaccines, but MDCK cell of YB or KA strain can not be approved as substrate for the preparation of attenuated viral vaccines.

＜正＞Background Amyloid-β peptide（Aβ） is one of the main components of senile plaques,a pathological hallmark of Alzheimer’s disease（AD）.Activated microglia represents a major source of inflammatory factors in Alzheimer’s disease and a possible source of cytotoxic factors.Aβpeptide has been shown to activate microglia and stimulate their production of inflammatory *** evidence has shown that the redox-active biometal Cu（Ⅱ） can modulate the physiochemical properties of Aβ.This article aims to evaluate the effect of oligomeric and fibrillar Aβand Cu（Ⅱ） on microglia activation. The chromosomal number variations & structural aberrations of the MDCK cell line, primary feline or canine kidney cell(FKC or CKC) and Hela cell line were investigated and their karyotypes of conventional chromosome bands were analyzed. The carcinogenesis or tumorigenicity testing of these cell lines in about 23. nude mice and for colony formation in soft agarose and for haemagglutination under different concentration of plant lectins of these cells were carried out. Under the prerequisite that the incidence of cancer or tumor in negative-control nude mice inoculated subcutaneously with primary feline or canine kidney cell cultures purified in vitro at passage 3.was 0 (0/22) and 0 (0/10), respectively. The incidence of the progressively-growing malignant tumor(MT) in positive-control nude mice inoculated subcutaneously with Hela cell cultures of KB, X, or NM20/X strain was 10/10, 25/25 and 5/51, respectively. The results showed that the incidence of tumor in nude mice with tetraploid YA strain of MDCK cell during 20 - 45 passages, with hy-podiploid JB strain of MDCK cell on passage 25, with di-and hypoploid JC strain of MDCK cell during 2-15 passages or with hypoploid M strain of MDCK cell during 9 - 27 passages was 28/58, 1/5, 4/18 and 0/3., respectively. The chromosomal analysis results showed that the ratio of difference in the rate of modal chromosome number between high(mcs+ n) and lowest (mcs) passages was not more than 5% - 15% and the structure aberrations was generally 0 - 3. . These results proved that the genetic characteristics of chromosomal number of cell lines determines their tumorigenicity, but it is species-specific. MDCK line has tumorigenicity no matter what its chromosome karyotype is, at least it has very low tumorigenicity even when its modal chromosome number is hypoploid. Lignum Aquilariae Resinatum(agarwood)is a solid-state condensate with mixed with resin and lignin that forms in Aquilaria and Gvrinops trees of the family *** is very expensive for its rare reserve and diverse functions as material of incense,aroma oil,medicine and ornamental *** to rapidly achieve large quantity of high-grade agarwood and underlying formation mechanism has aroused a lot of interests of researchers and industrialists. Aquilaria sinensis(Lour.)Gilg is the agarwood-producing plant species in china. Stem-partial-breaking method is usually used to artificially induce agarwood formation in Chinese A. sinensis *** we investigated changes on stem structure and volatile composition of stem-partial-breaking treated six-year old trees in continuous layers below wounding sites during agarwood formation. Typical wound inducible occlusions,tyloses and gels,were found in the vessels and fiber-tracheids in the initial stage of agarwood *** distribution extended to with increasing the treatment *** was observed that starch grains were exclusively located in the living parenchyma *** were found transporting from the parenchyma cells into surrounding tissues,neighboring fiber-tracheids and vessels,through the pits and a similar wax(oils) secreting way. Obvious four distinct layers including decomposed layer,agarwood layer,transition layer,and white-timber layer were observed below the wound surface days after *** typical agarwood chemical components were detected on days after *** chemicals assayed, previous notable wound inducible chemicals in other plant species were detected in different zones at different time points after wounded. From structure and chemical component aspects,our results support the hypothesis which we previously proposed that agarwood formation in *** was the result of defense response. Objective To search for the microorganisms which have the high selectivity of hydrolyzing glycyrrhizic acid(GL) into 18β-glycyrrhetinic acid-3.O-β-D-glucuronide(GAMG) without glycyrrhetinic acid(GA) *** GL was biotransformed by Aspergillus sp.,the products were separated by chromatography on reverse phase C18 column and semi-preparative HPLC,and their structures were elucidated on the basis of HR-ESI-MS,1D NMR(1H-NMR,13.-NMR,and NOESY) and 2D NMR(1H-1H COSY,HSQC,and HMBC) spectral *** Aspergillus *** partially hydrolyze GL into GAMG(3.,along with two minor byproducts,3.O-β-D-glucuronopyranosyl-18β-liquiritic acid(1) and 3.O-β-D-glucuronopyranosyl-24-hydroxy-18β-glycyrrhetinic acid(2).Conclusion Aspergillus *** the high selectivity of hydrolyzing GL into GAMG without GA byproduct and the yield of GAMG is about 60%.The complete assignments of 1H-NMR and 13.-NMR data for compounds 1 and 2 are reported for the first time.

The GJB2 gene(connexin 26) has been shown to be responsible for DFNB1 and DFNA3. We screened the GJB2 gene in 488 patients with prelingual deafness(Group 1), 124 with postlingual deafness(Group 2), and 117 normal hearing subjects(Group 3.. We found that, in Group 1, 65 patients(13.3.%) were homozygotes or compound heterozygotes and 51 patients (10.45%) carried a single pathogenic mutation. The 23.delC mutation was the most frequent mutation, accounting for 73.22% of the known pathogenic alleles in Group 1. No homozygotes or compound heterozygotes were detected in Group 2 or Group 3. Some postlingual deaf patients (2.42%) and normal hearing subjects(4.27%) were 23.delC carriers. Our preliminary data indicate that 23.delC, the most frequent mutation identified in this study, is a major cause for prelingual deafness.

Application of YLD Calculation in Assessing Disease Data──An Analysis of 4 Diseases in 2 Regions

The objective of the curtent study is to discuss the problems related to how data is used to calculate Yeare Lost with Disability (YLD) with the method recommended by the World Bank. The study includes collecting useful data, estimating disease duration and average age of disease onset,adjusting incidence and prevalence data by means of a software programme, DISMOD (Harvard university Incidence & Prevalence Model), and assessing the importance of YLD calculation for different *** and fatality rates of 3.diseases were estimated by experts at 2 round *** rates, disease duration and average age of disease onset were calculated and adjusted by DISMOD. YLD due to schizothrenia is the highest among 4 diseases in two regions. YLD is 18. 88 % in disability adjusted life year for 4 diseases in Xiacheng District, and 19 .97 % in Fuyang *** data can be used for the calculation of YLD after being adjusted. DISMOD is a useful instrument to test the internal consistency of incidence, prevalence, rendssion and fatality rate. The adjusted data are acceptable to experts and DISMOD. To get rational remission and fatality rates, we can use a cohort method through expert consultations. To renect overall burden of disease, YLD calculation should be used.

The most essential and crucial step during the pathogenesis of transmissible spongiformencephalopathy is the conformational change of cellular prion protein （PrPC） to pathologic isoform (PrPSc).Alot of data revealed that caveolae-like domains （CLDs） in the cell surface were the probable place where theconversion of PrP proteins *** E （ApoE） is an apolipoprotein which is considered toplay an important role in the development of Alzheimer's disease and other neurodegenerative diseases byforming protein complex through binding to the receptor located in the clathrin-coated pits of the cell *** this study,a 914-bp cDNA sequence encoding human ApoE3.was amplified from neuroblastoma cell *** human ApoE isomers were expressed and purified from Escherichia ***-specificantiserum was prepared by immunizing rabbits with the purified ***/His pull-down assay,immunoprecipitation and ELISA revealed that three full-length ApoE isomers interact with the recombinantfull-length PrP protein in *** regions corresponding to protein binding were mapped in the N-terminalsegment of ApoE （amino acid 1-194） and the N-terminal of PrP （amino acid 23.90）.Moreover,the recombinantPrP showed the ability to form a complex with the native ApoE from liver *** data provided directevidence of molecular interaction between ApoE and *** also supplied scientific clues for assessing thesignificance of CLDs on the surface of cellular membrane in the process of conformational conversion fromPrPCto PrPScand probing into the pathogenesis of transmissible spongiform encephalopathy.
