Probability parametric imaging with DCE-MRI data to identify prostate cancer in the peripheral zone

To quantitatively analyze prostate dynamic contrast-enhanced magnetic resonance images (DCE-MRI), a retrospective study was carried out to construct a mathematical model to predict whether a lesion is cancerous or not. With known diagnosis of 52 re-gions-of-interest (10 malignant, 26 benign and 16 normal) in peripheral zone (PZ) of prostate from 18 patients undergoing needle biopsy and 8 normal volunteers, we combined feature parameters (k1, k2 and tp) from the linear-slope model with logistic regression to determine the statistical probability model. Prostate cancerous probability maps were derived using the model on each subject to examine the correlation between the model-based prediction and the known diagnosis. Compared the model prediction results of 52 regions-of-interest with the histopathologic results, the total agreement rate was 90.4 % ; the agreement rate was 60 % for the malignant and 97. 6 % for the nonmalignant. The diagnostic results only referencing the probability maps by two radiologists showed a high agreement in tissue characteristics and lesion location with the combining results from other diagnostic techniques such as PSA (prostate specific antigen) level and MRS (magnetic resonance spectroscopy) .

The diagnosis of prostate diseases by magnetic resonance imaging (MRI) in vivo is sometimes difficult for the lower signal to noise ratio (SNR). To increase the SNR of the prostate image, we designed a RF coil that can be inserted into rectum and was named endorectal coil. The properties of RF coil were evaluated using a network analyzer. Moreover the images and spectroscopy of a special phantom were acquired and the results were compared to those of the commercial TORSO coil (G.E. medical Systems, USA). Our coil gave a significantly higher SNR at the region of interest (ROI). The achieved high local SNR and resulting high spatial resolution would add more anatomic and biochemical information to the diagnosis of prostate diseases.

The functional magnetic resonance imaging (fMRI) based on blood oxygen level dependent (BOLD) contrast has emerged as one of the most potent noninvasive tools for mapping brain function and has been widely used to explore physiological, pathological changes and mental activity in the brain. Exploring the nature and property of BOLD signal has recently attracted more attentions. Despite that great progress has been made in investigation of the characteristics and neurophysiological basis, the exact nature of BOLD signal remains unclear. In this paper we discuss the characteristics of BOLD signals, the nonlinear BOLD response to external stimuli and the relation between BOLD signals and neural electrophysiological recordings. Furthermore, we develop our new opinions regarding nonlinear BOLD response and make some perspectives on future study.

Effect of transforming growth factor-beta on activity of connective tissue growth factor gene promoter in mouse NIH/3.3.fibroblasts

AIM: To investigate the regulatory mechanism of transforming growth factor-betaon activity of connective tissue growth factor promoter in mouse NIH/3.3.fibroblasts. METHODS: The regulation fragment of the 5' flanking region of the human CTGF gene was linked to pGL3.Basic vector, a firefly luciferase reporter construct without promoter. The recombinant plasmid pCTGF-luc was transiently transfected to NIH/3.3.fibroblasts. The activity of CTGF promoter after treatment of TGF-β1 and MAPK pathway inhibitors were assayed with luciferase reporter gene assay system. RESULTS: TGF-β1-induced increase of CTGF promoter activity was concentration-dependent, with a plateau at 5 μg/L by 2.67-fold vs control (P3.80 (10 μmol/L), the p3. MAP kinaseinhibitor, decreased basal and TGF-β1-induced activation of CTGF promoter. However, inhibition of c-Jun-N-terminal kinase/stress-activated protein kinase by SP600125 (20 μmol/L) was without effect. CONCLUSION: TGF-β1stimulated the transcriptional activity of CTGF gene promoter in NIH/3.3.fibroblasts in a dose- and time-dependent manner. MAPK pathway may play a role in the regulation of TGF-β1-induced CTGF expression.

Gastric tissue samples were studied using mid-IR fiber-optic attenuated total reflectance (ATR) spectroscopy. FTIR spectra of 90 tissue samples from 48 patients, including 3. normal and 58 malignant tissue samples, were chosen as examples. Malignancy was usually characterized by the absence of CH and C=O bands, a weak amide II band near 1545 cm-1, a shift of the amide I band to lower wave number, a decrease in the ～1450 cm-1 peak to less than the～1400 cm-1 peak. Subtraction spectra indicate that the amide I and amide II bands of normal and malignant tissues have larger differences in peak positions and relative intensities. The statistical analysis results confirm this conclusion. The results indicate that FTIR fiber optic techniques provide important information about cancerous tissue of the stomach, which can be used to differentiate the malignant tissue from the normal tissue. Based on the above results we successfully realize the detection of the tumor tissues of digestive tract in vivo and in situ. And the results of detection cancer near operating room and in vivo and in situ in the operating room are consistent with the conclusions for the samples stored in liquid N2, which is the basis for the clinical application.

The full length cDNA coding for P15 INK4b, which is a cyclin-dependent kinase inhibitor, was cloned to plasmid PXJ41-neo (Eco R I IXho 1 site) and the new constructed plasmid pXJp15 was obtained. pXJp15 was transferred into the human hepatoma SMMC-7721 cells by lipofectine reagent. After G418 selection, a series of cell lines stably expressing high levels of P15 (named SHT) and the clone containing vector PXJ41-neo only (named SVXJ) were obtained by Northern and Western analysis. The results showed that the proliferation of SHT cells is inhibited compared with that of SVXJ cells. Cell cycle analysis indicated that overexpressing of P15 inhibited the growth of SHT cells by decreasing progrssion of cells from G1 to S and G2 to M phases. The levels of c-Myc and c-Fos were obviously decreased in SHT cells compared with control cells by Western blotting. The decreased expression of oncogene may be one of the molecular mechanisms of the effect of P15 on the proliferation of in SHT cells.

Alterations in circadian rhythms of melaton in and cortisol in patients with bronchial asthma

AIM: To investigate the possible relationships between alterations in circadian rhythm of melatonin, cortisol andbronchial asthma. METHODS: Salivary melatonin and cortisol were measured simultaneously by radioimmunoas-say in 10 mild intermittent or persistent patients, 11 moderate-to-severe persistent asthma patients, and 15 controlsubjects. Twelve salivary samples were collected in a series during a 24-h period in each subject. RESULTS: Theresults showed overall lower levels of salivary melatonin in asthma patients compared with control subject(P<0.01). The amplitude, peak-level, and baseline of salivary melatonin were significantly lower in mild intermit-tent or persistent (P<0.01, P<0.05) and moderate-to-severe persistent asthma patients (P<0.01) compared withcontrol group. The 24-h mean level of salivary cortisol was greatly lower and the acrophase was markedly delayedin patients with mild intermittent or persistent asthma (P<0.01) and moderate-to-severe persistent asthma (P<0.05, P<0.01) compared with control subject. CONCLUSION: Disordered circadian rhythms of salivary melato-nin and cortisol were found in asthma patients, which may be involved in the pathogenesis of bronchial asthma.

Identification of Hercynian shoshonitic intrusive rocks in central Hainan Island and its geotectonic implications

An identification has been made of some shoshonitic intrusive rocks in central Hainan Island recently. These rocks are K-rich (K2O=2.9%―5.1%, K2O/Na2O=0.95―2.12), distinctly enriched in LILE and LREE, strongly depleted in Nb, Ta, and moder- ately depleted in Sr and Ti, with (87Sr/86Sr)i = 0.70859―0.71425 and ε Nd(t) = (?2.77―?7.49). They were derived from an EMⅡ-type mantle source. The enrichment process is related to metasomatism of depleted mantle caused by a great amount of fluid-melt released from oceanic crust and terri- genous sediments at great depth (eclogite facies) during the subduction of the South china plate under the Indochina-South china Sea plate in the Carbon- iferous–Early Permian. A SHRIMP U-Pb zircon dating yields a crystallization age of 272±7 Ma for the sho- shonitic intrusions, which is coeval with the strongly peraluminous granites found in central Hainan Island. These two kinds of rocks generally possess syn-intrusion ductile deformation structures. Thus they are considered to have been generated during the early stage (syn-thrust phase) of a post-collisional event. The primary magma of shoshonitic rocks was produced at a depth > 80 km by decompression- dehydration melting of previously enriched litho- spheric mantle wedge, phlogopite-bearing garnet peridotite, which was in turn caused by the break-off of a descendent slab and upwelling of a hot as- thenosphere. The rising of melts was accompaniedby crustal contamination and crystallization fractiona- tion (AFC). Combining with other related data, it is proposed that the southwards subduction and amal- gamation of the South china plate with the Indo- china-South china Sea plate took place at ca. 287- 278 Ma, which was a part of the convergence proc- ess of the Pangea supercontinent. The suture zone was probably located along the line of Song Ma-Beibu Gulf-north margin of the Yunkai Moun- tains-Wuyi Mountains.
