N-glycosylation proteome of endoplasmic reticulum in mouse liver by ConA affinity chromatography coupled with LTQ-FT mass spectrometry

Glycosylation is the most versatile and one of the most significant protein post-translational modifications. It is generally classified into three categories according to the amino acid to which the glycan is attached: N-glycosylation, O-glycosylation and C-glycosylation. Synthesis of N-glycoproteins occurs in the rough endoplasmic reticulum (rER), and all N-glycoproteins synthesized in rER have uniform glycan endings with mannose (Man) and glucose (Glc). A systematic strategy was developed to comprehensively profile N-glycoproteins in complex biological samples. The lectin Concanvalin A (ConA), which has a high affinity for glycans ending with Man, was used to extract the N-glycoproteins synthesized or located in the rER, and identified the N-glycoproteins and their glycosylation sites by LTQ-FT MS. The analysis was repeated three times at both the biological sample and mass spectrometry levels. In total, 3.3.glycosylation sites on 212 N-glycoproteins were identified in the mouse liver. Of these, 13. were known N-glycoproteins in the Swissprot database. The identified N-glycoproteins were classified according to their cell location by the Swissprot database and GO software. The identified N-glycoproteins in this study would serve as a good complement to the N-glycoprotein database for the mouse liver.

In the human physiological activities, liver is not only the largest gland in metabolism of carbohydrates, fats, proteins, vitamins and hormones, but also an organ of secreting bile, detoxifcation, devourment, and protection function. The human fetal liver aged 22 weeks of gestation (HFL22w) is the major source of stem/progenitor cells of embryonic hematopoiesis and immune system. It also has a lot of genes relating to cellular recognition, migration, and homing. The study of the human fatal liver will help us understanding liver development and its physiological functions. For this reason, systemic analysis of human fetal liver proteome is performed in five aspects as below.

A NUMERICAL RESEARCH OF NONLINEAR BODY-WAVE INTERACTIONS

This paper presents numerical research results of nonlinear body-wave interactions. The body-wave interac- tion is treated as a transient problem with known initial conditions. The development of the flow can be obtained by a time-stepping procedure, in which the velocity potential of the flow at any instant is obtained by utilizing a source density distribution on all boundary surfaces. The Orlanski's method is used to implement the boundary con- dition at the open boundary. The position of intersection points are determined by a direct method. The contents of this paper are: (1) A study of the nonlinear radiation problem of a floating body; (2) A study of nonlinear wave diffraction problem around large structures; (3. Interaction of nonlinear waves with a free floating body; (4) An attempt in generating a numerical wave tank. Pretty good agreement is met between the numerical results and the analytical solution.

This paper investigates the upconversion luminescence of Yb 3. doped oxyfluoride vitroceramics excited by 960 nm diode laser. A new upconversion cooperative radiation luminescence is found, which is rather strong and positioned at 479 5 nm. It comes from a coupled state of two Yb 3. ion cluster. It is of significance due to the effective method to obtain compact blue light.

A time domain prediction of wave-induced ship motions by a Rankine panel method is investigated. Linear boundary conditions on free surface and mean wetted body surface are adopted, while the numerical damping method is used for the radiation conditions. The motions of two ships in regular head waves are computed by the present method. The related numerical results are compared with the experiment data and those from linear strip theory. The comparison shows satisfactory agreements for pitch and heave transfer functions.

A vertical one-dimensional numerical model for heat transferring within the near-surface snow layer of the Antarctic Ice Sheet was developed based on simplified parameterizations of associated physical processes for the atmosphere, radiation, and snow/ice systems. Using the meteorological data of an automatic weather station (AWS) at Dome A (80°22′S, 70°22′E), we applied the model to simulate the seasonal temperature variation within a depth of 20 m. Comparison of modeled results with observed snow temperatures at 4 measurement depths (0.1, 1, 3. 10 m) shows good agreement and consistent seasonal variations. The model results reveal the vertical temperature structure within the near-surface snow layer and its seasonal variance with more details than those by limited measurements. Analyses on the model outputs of the surface energy fluxes show that: 1) the surface energy balance at Dome A is characterized by the compensation between negative net radiation and the positive sensible fluxes, and 2) the sensible heat is on average transported from the atmosphere to the snow, and has an evident increase in spring. The results are considered well representative for the highest interior Antarctic Plateau.

The full set of hydrodynamic equations governing the oscillation of a bubble is solved numerically by using a modified equation of state with ionization. We show that the hydro-thermal process in the form of compression waves and low-level ionization are dominant inside a sonoluminescing bubble for a wide range of driving pressure. The resulting thermal bremsstrahlung radiation has pulse heights and widths that agree with experimental data for cases considered in this paper.

Correlative ionospheric disturbances in the sunlit hemisphere during the flare on July 14,2000

Using GPS data obtained from GPS sites located on the sunlit hemisphere during the flare on July 14, 2000, the ionospheric response to the flare was analyzed. A kind of tiny correlative ionospheric fluctuations were revealed from the temporal total electron content (TEC) curves derived from GPS measurements, and the values of this kind of disturbances are about 1015 m-2. A comparative analysis of these disturbances and soft X-ray flux detected at GOES satellite during the flare is done. It is found that this kind of disturbances revealed in the temporal TEC curves did occur in a very large area of sunlit ionosphere and its behavior is mainly controlled by the characteristics of the flare’s extra radiation. Because the similar fluctuations are not found in the curve of soft X-ray flux, it is concluded that the flare extra radiation in EUV band should have corresponding fluctuations and it is the fluctuations that cause the TEC disturbances.
