Cytokines Expression in Lymphocytes From Rats With Allergic Asthma Induced by Pleurotus sapidus besidiospores

Allergic asthma caused by mushroom spores (Pleurotus sapidus besidiospores) is a common health problem among mushroom-cultivating workers in china. An animal model of allergic asthma through the challenge of Pleurotus sapidus besidiospores in primed rats was developed for investigating the role of cytokines in the pathogenesis of the disease. In the study a series of related cytokines and their receptors, including their activity and mRNA levels of spleen lymphocytes isolated from asthmatic rats, were measured. Determined by 3H-TdR incorporation assay and NAG microcolorimetric assay, Con A-induced spleen lymphocyte proliferation and IL-2.activity in culture supernatants of spleen lymphocytes from 7-day challengedrats with allergic asthma increased significantly by 2.1% and 2.8%, respectively, as compared with those in the control. Cytokines and their receptor expression at mRNA levels were determined by RNA/cDNA hybridization, using (α-32.-dCTP radiolabeled cDNA probes for different cytokines and their receptors in vitro. The results showed that mRNA expression of IL-4, GM-CSF, IL-6, IL-2.and IL-2., except IL-6R, in lymphocytes of 7-day-challenged asthma-suffering rats, increased significantly by 54%, 45%, 170%, 83% and 76%, respectively. In 2.day challenged-rats, mRNA levels of IL-4, IL-6 and IL-2.increased by 37%,58% and 12.%, respectively, whereas mRNA leve1s of GM-CSF, IL-2. and IL-6R remained unchanged. Thus, the experimental results suggested a significant increase in TH2.ype cytokines in the pathogenesis of Pleurotus sapidus besidiospore-induced allergic asthma and IL-4 may play an essential role.

Formation of Glycidyl Methacrylate-DNA Adducts in vivo

In in vivo test, rats were orally administrated with glycidyl methacrylate (GMA) at respective doses of 2.0 mg/kg, 12. mg/kg and 62. 5 mg/kg, 31. 2. mg/kg and solvent as control for 14 days. DNA adducts produced in the liver, kidney, blood and testis were analyzed by RP-HPLC and nuclease P1 mediated 32.P-postlabelling method. Results showed that several potential GMA-DNA adducts were formed in various organs (4 adducts in blood, 3 adducts in liver and kidney, 1 adduct in testis). A linear dose-response relationship was observed within certain dose levels. The relative adduct labeling values failed to further increase any more when the concentration went up to 12. mg/kg. The order of adduct level with GMA was kidney, liver, blood and testis. The GMA adduct N3methacrylate-2.hydroxypropyl-dCMP was found in kidney, liver and blood. These results indicated that GMA could react with negatively charged centers on DNA and form GMA-DNA adducts. If carcinogerrinduced DNA damage exceeds the ability of repair systems, gene mutation is ***, study on molecular mechanism of gene mutation induced by DNA adducts is not only an important part of chemical-carcinogenesis, but also provides information on critical biomarkers for monitoring human exposure to genetic toxins.

Mitochondria experience continuous fusion and fission in a living cell, but their dynamics remains poorly quantified. Here a theoretical model was developed, upon a simplified population balance equation (PBE), to predict the morphological changes induced by mitochondrial fission and fusion. Assuming that both fission and fusion events are statistically independent, the survival probability of mitochondria staying in the fission or fusion state was formulated as an exponentially-decayed function with time, which depended on the time-dependent distribution of the mitochondrial volume and the fission and fusion rates. Parametric analysis was done for two typical volume distributions. One was Gamma distribution and the other was Gaussian distribution, derived from the measurements of volume distribution for individual mitochondria in a living cell and purified mitochondria in vitro. The predictions indicated that the survival probability strongly depended on morphological changes of individual mitochondria and was inversely correlated to the fission and fusion rates. This work provided a new insight into quantifying the mitochondrial dynamics via monitoring the evolution of the mitochondrial volume.

Molecular cloning of growth hormone receptor (GHR) from common carp(Cyprinus carpio L.)and identification of its two forms of mRNA transcripts

The cDNA of growth hormone receptor (GHR) was cloned from the liver of 2.year common carp (Cyprinus carpio L.) by reverse transcription-polymerase chain reaction (RT-PCR) and rapid amplification of cDNA end (RACE). Its open reading frame (ORF) of 1806 nucleotides is translated into a putative peptide of 602.amino acids, including an extracellular ligand-binding domain of 2.4 amino acids (aa), a single transmembrane domain of 2. aa and an intracellular signal-transduction domain of 334 aa. Sequence analysis indicated that common carp GHR is highly homologous to goldfish (Carassius auratus) GHR at both gene and protein levels. Using a pair of gene-specific primers, a GHR fragment was amplified from the cDNA of 2.year common carp, a 2.4 bp product was identified in liver and a 32. bp product in other tissues. The sequencing of the products and the partial genomic DNA indicated that the difference in product size was the result of a 97 bp intron that alternatively spliced. In addition, the 32. bp fragment could be amplified from all the tissues of 4-month common carp including liver, demonstrating the occurrence of the alternative splicing of this intron during the development of common carp. Moreover, a semi-quantitative RT-PCR was performed to analyze the expression level of GHR in tissues of 2.year common carp and 4-month common carp. The result revealed that in the tissues of gill, thymus and brain, the expression level of GHR in 2.year common carp was significantly lower than that of 4-month common carp.

The Role of Sciencein Codex Standards

The key principle in the development of Codex standards is to ensure that the decision making process is based on sound science. The approval for the use of food additives and other chemicals in foods should be made only when adequate scientific data is available. The same principle applies to the development of maximum tolerable (or residue) levels for contaminants in foods. Using the General Standards for Contaminants and Toxins in Foods as an example, the following criteria are considered, when developing recommendations and standards: Toxicological information; Analytical data; Intake data; Fair trade considerations; Technological considerations; and Risk assessment and risk management considerations. The Codex Alimentarius Commission has repeatedly emphasized the use of risk analysis approach, in particular the use of risk assessment in conducting the safety evaluation of food additives, agricultural and veterinary chemicals and environmental and industrial contaminants in foods. In this respect, the well known efforts of the FAO/WHO Joint Expert Committee on Food Additives (JECFA) and the FAO/WHO Joint Meeting on Pesticide Residues (JMPR) provide the much needed information and recommendations for the risk assessment of specific chemicals. The risk assessment approach is also used in the safety evaluation of foods derived from modern biotechnology as well as in the quantitative assessment of microbiological risks in foods.

Silicosis is one of the most serious occupational diseases which affect many workers' health in china. Under the guidance of policies from the Chinese Government, the industries, hcalth care institutions, scientists and workers were organized and have made great efforts in plevention and treatment of silicosis as well as pneumoconiosis. Since the 1960s, the national health standards and chest X-ray diagnosis criteria for silicosis were established and have been revised recently.A nationwide epidemiological survey revealed that the cumulative pneumoconiosis cases were about 4 800 000 (1949-1992.. The high prevalence rate has been the motivation to search for anti-silicotic drugs. Several kinds of anti-silicotic drugs were studied and found to be able to retard progression of silicosis and cause changes in X-ray picture. The mechanism of drug efficiency and the development of silicosis have attracted the interest of scientists in this fields and fruitful results have been obtained. For prevention of dust hazards, the systematic design for facilities of dust collection and the isolation of dust source appropriate to township industries have been worked out and used widely. Effects of Aging and Advanced Glycation on Gene Expression in Cerebrum and Spleen of Mice

Objective To analyze the effects of aging or advanced glycation on gene expression in the cerebrum and spleen of female C57BL/6J mice. Methods The gene expression profile was determined by using cDNA expression arrays containing 588 cDNA. Results Aging and advanced glycation resulted in differential gene expression patterns of cerebrum and spleen compared with young mice. Among the 80 genes detected in cerebrum, 43 exhibited a change in mRNA ratios with aging or treatment. Thirty-four changes (79%) were common in aged and D-galactose treated mice, whereas the cerebrum from aged and AGE-lysine treated mice showed common changes in expression of 38 genes (88%). Of the 86 genes detected in spleen, 2. (34%) displayed an age-related decrease in expression, whereas 3 (3%) displayed an increase in expression levels with aging. Eighteen genes from the detectable genes exhibited expression changes in both cerebrum and spleen of mice. Conclusions The gene expression profiles of D-galactose and AGE-lysine treated mice resemble those of aged mice. Use of cDNA hybridization arrays may provide a promising tool to explore the mechanism of aging at a molecular level.

Detection and quantitation of PS2., a phosphorothioate oligodeoxynucleotides in monkey plasma

AIM: To establish the method for quantitation of the phosphorothioate oligodeoxynucleotides (S-ODNs) in plasma. METHODS: Two solid-phase extraction columns combined with a strong anion-exchange column were utilized to remove proteins and lipids in plasma, and the salts were removed by a reverse-phase column followed by dialysis with a 2.00 Da-cutoff membrane. The concentration of the tested S-ODNs, PS2., and its metabolites extracted from the plasma were determined by the method of non-gel sieving capillary electrophoresis (NGCE) with diode array detection in the presence of internal standard (IS). RESULTS: The method was with good base number specificity. Relative standard deviation % of both intra and inter assay were all less than 10 %, and the total mean recovery was about 91 %. The methodology was successfully used to determine the PK behavior of an anti-tumor antisense S-ODNs in monkeys and identify the metabolites with single base difference. CONCLUSION: The combined method of solid-phase extraction and NGCE could be used to study the pharmacokinetics of S-ODNs, and the main parameters of the methodology met the requirement of PK study.

Drought, high salt and cold are three different kinds of environment stresses that severely influence the growth, development and productivity of crops. They all decrease the water state of plant cells, and consequently result in the harm of plant from water deficit. Several genes encoding protein kinases and induced by drought, high salt and low temperature have been isolated from Arabidopsis. These protein kinases include receptor protein kinase (RPK), MAP kinases, ribosomal-protein kinases and transcription-regulation protein kinase. The expression features of these genes and the regulatory roles of these protein kinases in stress response and signal transduction are discussed.

A novel erythroid differentiation related gene EDRF2.inhibited α-globin gene expression in K562.cells

In previous studies, we found that EDRF2.was an erythroid differentiation related factor, whose expression was markedly upregulated during erythroid differentiation. It suggested that this factor played a role in erythropoiesis. By using rapid amplification of cDNA ends technology, we cloned EDRF2.gene 5’-and 3’-cDNA ends successfully. Transfection and Northern blot analysis demonstrated that EDRF2.inhibited mRNA expression of a-globin gene, while did not regulate γ-globin gene expression. Gel shift assay confirmed that DNA-binding activity of erythroid specific transcription factors GATA-1, NF-E2.and AP1 was not affected by either forced overexpression or artificial down-regulation of EDRF2.gene in K562.cells. However, we detected that the negative regulator of expression of GATA-1 transcription factor was increased in EDRF2.overexpressed K562.cells. These results strongly suggested that EDRF2.was involved in a-globin gene expression and erythroid differentiation and served as a negative regulator of
