Aim:To explore the biofunctions of human B7-H4 generated from ***:The gene of human B7-H4 extracellular region（IgV-like andIgC-like domains）was obtained by PCR from human cDNA FLJ22418 and theninserted into the prokaryotic expression vector pGEX-5X-3 expressing glutathiones-transferase（GST）fusion *** being identified by restriction enzymedigestion and sequencing,the recombinant vector was transferred into host strainE coli BL21-RIL（DE3）.A 47 kDa fusion protein（GST/hB7-H4）was induced byisopropyl-beta-D-thiogalactopyranoside（IPTG）and purified by standard meth-ods reported in the prokaryotic *** inhibitory effect of GST/hBT-H4 onproliferation of T cells was observed in vitro by CD3mAb activated T-cell cultur-ing system and [3H]-thymidine incorporation *** concentrations ofinterleukin-2 and iterferon-g in the supernatants of T cells were determined ***:We successfully constructed the method for high-level expres-sion and purification of the hB7-H4 extracellular domain as GST fusion proteinfrom E *** GST/hB7-H4 fusion protein produced in bacteria had obviousbiological activity to inhibit T-lymphocyte proliferation and IL-2 ***:The prokaryote expression system could be used to generate hB7-H4 protein with natural spatial conformations and biological functions,whichprovided an efficient and economical way for the preparation of this target protein.

The aim of the study was to demonstrate the degeneration of the dopaninergic nigrostriatal pathway in Parkinson's disease(PD) and Essential Tremor (ET) by using the cocaine derivative 99mTc-TRODAT-1 SPECT and correlate the findings to the clinical severities (Hoehn and Yahr scale, H/Y). 28 patients with idiopathic Parkinson's disease, 10 patients with Essential Trenor and 19 healthy volunteers were investigated. The acquisition were performed 3 h postinjection of 99mTc-TRODAT-1, ROIs were drawn over the images of striatum and cerebellum, and ratios of striatum to cerebellar(ST/CB) were calculated. Ratios differed significantly between PD and controls, but ratios didn't show significant difference between ET patients and controls. A significant correlation didn't exist between ratios and clinical severities. Hemiparkinson's patients revealed significantly diminished 99mTc-TRODAT-1 binding not only clinically affected but unaffected side. Our findings indicated that 99mTc-TRODAT-1 SPECT is not only a reliable method to discriminate between PD and controls but also a useful tool for differential diagnosis in clinically unclear cases such as ET resembling PD.

In order to understand the allograft rejection in orthotopic liver transplantation (OLT), an allograft re- jection rat model was established and studied by proteomic approach. The protein expression profiles of liver tissues were acquired by fluorescence two-dimensional difference gel electrophoresis (2D DIGE) that incorporated a pooled internal standard and reverse fluorescent labeling method. The expression levels of 27 protein spots showed significant changes in acute rejection rats. Among these spots, 19 were identified with peptide mass fingerprinting using matrix-assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF MS) after tryptic in-gel digestion. The results of the present paper could be helpful for our better understanding of allograft rejection in organ transplantation.

BACKGROUND: Surgeons are always concerned about the localization of pancreatic functioning islet cell tumor. If the tumor is accurately localized before operation, resection of the pancreatic body and tail without intention can be avoided. The purpose of this study was to evaluate spiral CT localization of pancreatic functioning islet cell tumors and CT techniques. METHODS: CT manifestations in 6 patients with clinically and pathologically proved pancreatic functioning islet cell tumors ware analyzed retrospectively. RESULTS: In 4 patients with insulinomas and 2 patients with glucagonomas, 5 were localized accurately by CT be- fore surgery and 1 was detected retrospectively. The en- hancement of tumors was greater than that of normal pan- creas in arterial phase and pancreatic parenchymal phase. Four patients showed mild high-density and 2, iso-density in the portal venous phase. CONCLUSION: Spiral CT multi-phase enhanced scan with 1.5 ml/kg contrast agent and 2-5 mm slice width can loca- lize functioning islet cell tumors accurately.

AIM: To observe effects of propofol on nociceptive response at superspinal and spinal level in rats. METHODS: Two hundreds and fifty-eight Sprague-Dawley male rats were randomized into thirty-two groups. Propofol and bicuculline were microinjected into lateral ventricle (icv), ventrolateral periaqueductal gray (vlPAG), intrathecal (ith), and intraperitoneal (ip). The noxious responses were evaluated by hot plate and formalin test. RESULTS: In hot-plate test, systemic and superspinal administration of propofol (40 mg·kg-1 ip, 100 μg in 10 μL, icv, and 4 μg in 0.4 μL vlPAG microinjection) produced hyperalgesia (P30 min by microinjection of bicuculline (10 ng in 0.4 μL, vlPAG) (P3 %, 54.8 %, 80.8 %, and 97.4 % at 10, 20, 30 and 40 min by ith bicuculline (P30 min by ith bicuculline (P<0.05) after formalin injection. Hyperalgesia produced by ip propofol in both hot plate and formalin test could not be antagonized by vlPAG administration of bicuculline. CONCLUSION: GABAA receptor partly mediated propofol-induced hyperalgesia at superspinal and analgesia at spinal cord in rats.

Aim:To investigate the protective effect of triptolide （TRI） on ischemia/reperfusion-induced injury of transplanted rabbit lungs and to investigate the mechanismsunderlying the actions of ***:We established the rabbit lung trans-plantation model and studied lung injury induced by ischemia/reperfusion andthe inhibitory effect of TRI on *** severity of lung injury was determinedby a gradual decline in PvO2,the degree of lung edema,the increase in themyeloperoxidase （MPO） activity,and the ultrastructural changes of *** activation of NF-kB was measured by *** in intercellular adhesion molecule-1 （ICAM-1）,which is the target gene ofNF-kB,was evaluated by ***:After reperfusion,there was a gradualdecline in the PvO2level in the control group （group Ⅰ）.The level of PvO2in thegroup treated with lipopolysaccharide (group Ⅱ was significantly decreased,whereas that of the group treated with TRI （group Ⅲ） was markedly improved（P<0.01）.In group Ⅲ,the activity of MPO was downregulated,and the pulmonaryedema did not become severe and the ultrastructure of the donor lung *** activity of NF-kB and the expression of ICAM-1 was significantlyincreased in the donor *** blocked NF-kB activation and ***:The effects of TRI on reducing injury to donor lungsinduced by ischemia/reperfusion may possibly be mediated by inhibiting theactivity of NF-kB and the expression of the NF-kB target gene ***,TRI could be used in lung transplantations for improving the function of donorlungs.

AIM: To analyze the correlation between cytokineinduced killer (CIK) cells adoptive immunotherapy and cancer-related death in gastric cancer patients. METHODS: One hundred and fifty-six gastric cancer patients after operation at the Third affiliated hospital of soochow university were enrolled in this study. Their clinical data including demographic characteristics, operation time, tumor size, pathological type and staging, tumor metastasis, outcome of chemotherapy or CIK cells adoptive immunotherapy, survival time or time of death were collected with a standard structured questionnaire. Kaplan-Meier method was used to estimate the median survival time, and the 2- and 5- year survival rates. Hazard risk (HR) and 95% confidence interval (95% CI) of CIK cells adoptive immunotherapy for gastric cancer were calculated using the two-stage time-dependent covariates Cox model. RESULTS: The survival time of gastric cancer patients was longer after CIK cells adoptive immunotherapy than after chemotherapy (χ 2 = 10.907, P = 0.001). The median survival time of gastric cancer patients was also longer after CIK cells adoptive immunotherapy than after chemotherapy (49 mo vs 27 mo, P first stage Cox model was used to define the subjects who remained alive beyond 36 mo as survivors.
