Objective:We have previously reported that central administration of urocortinⅢelicited transient increase in mean arterial blood pressure and heart *** we aimed to examine the effects of urocortinⅢon the hypothalamic paraventricular nucleus(PVN) magnocellular neurons by whole-cell patch-clamp recordings and single-cell reverse transcription-polymerase chain reaction(RT-PCR) techniques,to explore the mechanisms of cardiovascular function modulation by urocortinⅢ.Methods:Hypothalamic slices were prepared from P12-14 day old male Wistar *** potentials and/or currents were monitored with an Axopatch-ID amplifier,acquired through a Digidata 1200 series analog-to-digital interface on a personal computer using Clampex 8.1 *** of cytoplasm and reverse transcription were carried out after the end of whole-cell *** and nested PCR amplification was used for screening oxytocin(OT),glyceraldehyde-3-phosphate dehydrogenase(GAPDH),CRFR-1 mRNA and CRFR-2 ***:Under current-clamp,urocortin HI(10 nmol/L to 600 nmol/L) resulted in a decrease in basal firing rate,and a dose-dependent hyperpolarization which was unaffected by the mixture of tetrodotoxin (0.5μmol/L),CNQX(10μmol/L) and bicuculline(10μmol/L).Under voltage-clamp,SCP(300 nmol/L) increased an inwardly rectifying K *** UrocortinⅢ-induced current was abolished by extracellular application of tertiapin(30 nmol/L),a selective blocker of G protein-activated,inward rectifying potassium(GIRK) channels. Moreover,the UrocortinⅢ-induced current was completely blocked by extracellular application of a selective corti-cotrophin -releasing factor(CRF) receptor 2 antagonist,antisauvagine-30(1μmol/L).Conclusion:These results suggest that UrocortinⅢinhibites the excitability of the hypothalamic paraventricular nucleus magnocellular neurons through enhancement of GIRK channels activity via CRF receptor 2 in vitro in mice.

Objective:Although the sensory stimulus precisely evokes response in Purkinje cells(PC),the cellular mechanisms of sensory information processing in cerebellar cortex are *** the present study,we aimed to study the cerebellar PCs in response to natural stimulus on ipsilateral whisker pad by in vivo whole-cell recording ***:Adult(6-8 weeks old) ICR mice were anesthetized with *** a watertight chamber was created around the hemisphere,a craniotomy was drilled to expose the cerebellar surface in correspondence to CrusⅡ.Tactile stimulus on ipsilateral whisker pad was performed by air-puff through a stainless tube connected with a pressurized injection *** potentials and/or currents were monitored with an Axopatch-1D amplifier,acquired through a Digidata 1200 series analog-to-digital interface on a personal computer using Clampex 8.1 *** between the mean values recorded under control and test conditions were evaluated by Student’s paired t-test using SPSS ***:We found that air-puff stimulus on trigeminal afferents evoked neither complex spike nor simple spike,but indeed elicited a strong GABA receptor-mediated inhibition of PCs in cerebellar cortex folium CrusⅡin urethane anesthetized *** potential recordings from both molecular layer and PC layer showed that air-puff stimulus evoked a sequence of parallel fiber volley followed by a GABA receptor-mediated inhibition,which was completely blocked by AMPA receptor ***-attached recordings from PCs showed that air-puff stimulus evoked a pause of spike firing,GABA receptor antagonist abolished the pause,revealed the evoked firing of these PCs in response to the *** current-clamp,air-puff stimulus did not evoke complex spike or simple spike in PCs,but induced strong inhibitory postsynaptic potential (IPSP) in both PCs somata and *** the presence of GABA receptor antagonist,air-puff stimulus could not evoke complex spik

Flow instability of buoyant-Marangoni convection in the LEC GaAs melt

Flow transitions and instabilities have significant effects on the quality of the crystals. The flow and heat transfer in the LEC GaAs melt are numerically studied by a time-dependent and three-dimensional turbulent flow model. The effects of the change of the buoyancy and Marangoni force on the flow state are analyzed by changing the temperature difference between the crystal and the crucible walls. The results show that the flow will transform from axisymmetric steady flow to non-axisymmetric oscillatory flow when the temperature difference exceeds the critical value, and that the mechanism of the transition is attributed to the Marangoni instability. The critical temperature differences for the flow transitions corresponding to different melt depth H are numerically predicted. Several important characteristics of the non-axisymmetric buoyant-Marangoni convection are numerically observed and compared with that of the non-axisymmetric mixed convection coupled with crystal rotation.

Molecular dynamics simulation study on zwitterionic structure to maintain the normal conformations of Glutathione

Molecular dynamics simulations were applied to normal conformational Glutathione (GSH) and GSH over zwitterionic and hydrophobic surfaces respectively. Conformational analysis of GSH during the simulation time on RMSD, conformational flexibility and dihedral distribution were performed. The re- sults showed that zwitterionic structure maintains the normal conformations of GSH to a better extent, which should be a first good proof of the hypothesis of "maintain of normal structure".

This paper is concerned with the theoretical foundation of support vector machines (SVMs). The purpose is to develop further an exact relationship between SVMs and the statistical learning theory (SLT). As a representative, the standard C-support vector classification (C-SVC) is considered here. More precisely, we show that the decision function obtained by C-SVC is just one of the decision functions obtained by solving the optimization problem derived directly from the structural risk minimization principle. In addition, an interesting meaning of the parameter C in C-SVC is given by showing that C corresponds to the size of the decision function candidate set in the structural risk minimization principle.

Sensory stimuli evoke responses in cerebellar Purkinje cells(PCs) via mossy fiber-granule cell pathway. However,the properties and characterization of synaptic responses evoked by the tactile stimulus in cerebella PCs are *** the present study,we investigated the synaptic responses of cerebellar PCs in response to the airpuff stimulus on the ipsilateral whisker pad in urethane-anesthetized mice.A total of 33 PCs from 35 adult(6-8 weeks old) ICR mice were studied by somatic or dendritic patch-clamp recording with biolytic staining and pharmacological *** current-clamp conditions(I = 0),air-puff stimulus evoked strong inhibitory postsynaptic potentials (IPSPs) in PCs *** of SR95531,a specific GABA receptor antagonist,blocked IPSPs,and revealed the stimulus-evoked simple spike *** voltage-clamp conditions,tactile stimulus evoked a sequence of transient inward currents followed by strong inhibitory postsynaptic currents(IPSCs) in both PCs stomata and *** of GABA receptor antagonist,SR95531 blocked IPSCs,revealed the excitatory postsynaptic currents(EPSCs) in somata and a temporal summation of parallel-fiber EPSCs in dendrites of *** also demonstrated that PCs respond to both onset and offset of air-puff *** findings indicated that the tactile stimulus could not evoke the strong EPSCs and fire PCs,but induced the rapid activation of strong GABA rereptormediated IPSCs in both stomata and dendrites of *** propose that the inhibition play a crucial role in controlling the output of sensory information in PCs in cerebellar cortex CrusⅡin mice.

Altered subcellular distribution of nucleolar protein fibrillarin by actinomycin D in HEp-2 cells

AIM: To study the effects of actinomycin D on subcellular distribution of nucleolar protein fibrillarin in HEp-2 (human esophageal epithelial type 2) cells, and molecular mechanisms for maintenance of fibrillarin in nucleolus. METHODS: Indirect immunofluorescence assay was employed to investigate subcellular distribution of nucleolar protein fibrillarin and immunoblotting analysis was used to detect the total cellular amount of fibrillarin. RESULTS: Control cells with no drug treatment showed bright clumpy nucleolar staining, which indicated that fibrillarin decorated the nucleolus only. Treatment with actinomycin D caused dislocation of fibrillarin from nucleoli to nucleoplasm with numerous stained small nucleoplasmic entities. Immunoblotting showed that neither total cellular amount of fibrillarin nor the integrity of fibrillarin was changed upon the treatment. The dislocation of fibrillarin in cells treated at a lower concentration (0.05 mg/L) of actinomycin D, was totally reversible after removal of the drug from the medium. However, this reversion was not observed at a high drug concentration (1 mg/L). CONCLUSION: Actinomycin D induced dislocation of fibrillarin from nucleoli to nucleoplasm in HEp-2 cells. The retention of fibrillarin within the nucleolus was related to active RNA synthesis.

Purposeless repeated acquisition time-lapse seismic data processing

In china, most oil fields are continental sedimentation with strong heterogeneity, which on one side makes the reservoir prospecting and development more difficult, but on the other side provides more space for searching residual oil in matured fields. Time-lapse seismic reservoir monitoring technique is one of most important techniques to define residual oil distribution. According to the demand for and development of time-lapse seismic reservoir monitoring in china, purposeless repeated acquisition time-lapse seismic data processing was studied. The four key steps in purposeless repeated acquisition time-lapse seismic data processing, including amplitude-preserved processing with relative consistency, rebinning, match filtering and difference calculation, were analyzed by combining theory and real seismic data processing. Meanwhile, quality control during real time-lapse seismic processing was emphasized.

We consider the permeability estimation problem in two-phase porous media flow. We try to identify the permeability field by utilizing both the production data from wells as well as inverted seismic data. The permeability field is assumed to be piecewise constant, or can be approximated well by a piecewise constant function. A variant of the level set method, called Piecewise Constant Level Set Method is used to represent the interfaces between the regions with different permeability levels. The inverse problem is solved by minimizing a functional, and TV norm regularization is used to deal with the ill-posedness. We also use the operator-splitting technique to decompose the constraint term from the fidelity term. This gives us more flexibility to deal with the constraint and helps to stabilize the algorithm.
