Transforming growth factor-b 1 (TGF-β1), a multi-function polypeptide, is a double-edged sword in cancer. For some tumor cells, TGF-β1 is a potent growth inhibitor and apoptosis inducer. More commonly, TGF-β1 loses its growth-inhibitory and apoptosis-inducing effects, but stimulates the metastatic capacity of tumor cells. It is currently little known about TGF-β1-promoted cell migration in hepatocellular carcinoma (HCC) cells, let alone its mechanism. In this study, we found that TGF-β1 lost its tumor-suppressive effects, but significantly stimulated cell migration in SMMC-7721 human HCC cells. By FACS and Western blot analysis, we observed that TGF-β1 enhanced the expression of α5β1 integrin obviously, and subsequently stimulated cell adhesion onto fibronectin (Fn). Furthermore, we observed that TGF-β1 could also promote SMMC-7721 cells adhesion onto laminin (Ln). Our data also provided evidences that TGF-β1 induced epithelial-to-mesenchymal transformation (EMT) in SMMC-7721 cells. First, SMM

Preparation and characterization of some surface negatively charged residue mutants of cytochrome b_5

Site-directed mutagenesis was used to obtain seven variants of tryptic fragment of bovine liver cytochrome bs (cyt bs), in which the negatively charged residues around the heme exposed edge of cyt bs were replaced by hydropho-bic amino acid alanine. Double-site mutants, triple-site mutants and even quadruple-site mutants were obtained. DNA sequencing and molecular weight measurements of the mutant proteins both confirmed that these site-directed muta-genesises were successfully performed. Spectroelectrochem-istry of these mutant proteins revealed that the apparent redox potentials of these mutant proteins caused a positive shift of 2-10 mV. The global structure of these mutant proteins did not show much difference from that of the wild type cyt bs, providing a solid base for the further study on the roles of the proteins’ surface charges.

Semantic similarity between Gene Ontology (GO) terms is critical in resolving semantic heterogeneousness when integrating heterogeneous biological databases. Traditionally, distance based and information content based measures are two major methods. In this paper, a new method based on semantic pathway covering is proposed and an algorithm, COMBINE algorithm, is presented, which considers information contents of two given nodes and those of all nodes included in the two nodes' pathways. Experiments show that COMBINE algorithm obtains the highest correlation index compared with those distance based and information content based algorithms.

For the standard Lagrangian in classical mechanics, which is defined as the kinetic energy of the system minus its potential energy, we study the rate of convergence of the corresponding Lax-Oleinik semigroup. Under the assumption that the unique global minimum point of the Lagrangian is a degenerate fixed point, we provide an upper bound estimate of the rate of convergence of the semigroup.

The paper studies the effects of changing margin levels on the price of fixtures options and how to organize a market maker＇s position. Black model （1976） becomes a special case of this paper. The paper prices futures options by duplicating them and adopting the theory of Backward Stochastic Differential Equations （BSDEs for short）, Furthermore, the price of a futures option is the unique solution to a nonlinear BSDE.

Pharmacokinetic difference between S-(+)- and R-(-)-etodolac in rats

AIM: To study whether etodolac enantiomers have pharmacokinetic difference after oral administration. METHODS: Fourteen rats, divided into two groups randomly, were orally given S-(+)- or R-(-)-etodolac at a single dose of 20 mg/kg, respectively. Blood samples were collected before and at 5, 10, 20, 30 min and 1, 3, 6, 12, 24, 48, 72 h after treatment. The plasma samples were analyzed with a high-performance liquid chromatographic method. RESULTS: The calibration curves were linear in the range of 0.5-50.0 mg/L (r=0.9999) to S-(+)-etodolac and 2.0-200.0 mg/L (r=0.9999) to R-(-)-etodolac, respectively. The main pharmacokinetic parameters of S-(+)- and R-(-)-etodolac were as follows: t1/2(λz)18±4 h vs 19.4±2.2 h, tmax 3.3±2.6 h vs 4±4 h; Cmax 29±6 mg/L vs97±14 mg/L, AUC0-t 706±100 h·mg·L-1 vs 2940±400 h·mg·L-1, CL(s) 0.030±0.006 L·kg-1·h-1 vs 0.0065±0.0010 L·kg-1·h-1 and V/F 0.25±0.22 L·kg-1 vs 0.03±0.05 L·kg-1. Lipoteichoic acid (LTA),as a primary immunostimulus,triggers the systematic inflammatory responses. Our hypothesis is that apoA Ⅰ can neutralize LTA toxicity,like its effect on ***/c mice were chal- lenged with LTA,followed by human apoA Ⅰ *** found that apoA Ⅰ could attenuate LTA-in- duced acute lung injury and inflammation and significantly inhibit LTA-induced IL-1β and TNF-α accumu- lation in the serum (P<0.01 and P<0.05 respectively),as well as in bronchoalverolar lavage (BAL) fluid (P<0.01 and P<0.05,respectively).Moreover,apoA Ⅰ could significantly reduce the L-929 cell mortali- ty caused by LTA-activated macrophages in a dose-dependent ***,apoA Ⅰ treatment could diminish LTA-mediated NFκB nuclear translocation in *** in vitro binding assay indicated that apoA Ⅰ can bind *** results clearly indicated that apoA Ⅰ can effectively protect against LTA-induced sepsis and acute lung *** mechanism might be related to the binding and neutralization of LTA.
