To review the inhibitory effect of preventive approaches on recurrence after operation in patients with hepatocellular carcinoma (HCC), we summarized all available publications. reporting randomized control trial indexed in PubMed. The treatment approaches presented above included preoperative transcatheter arterial chemoembolization (TACE),post-operative TACE, systemic or locoregional chemotherapy,immunotherapy, Interferons and acyclic retinoic acid Although no standard treatment has been established,several approaches presented promising results, which were both effective and tolerable in post-operative patients. Preoperative TACE was not effective on prolonging survivals,while post-operative TACE was shown with both diseasefree survival and overall survival benefits in some papers,however, it was also questioned by others. Systemic chemotherapy was generally not effective on prolonging survival but also poorly tolerated for its significant *** immunotherapy using LAK cells was proved to be beneficial to patients' survival in a recent paper. Interferon and Interferon β can inhibit recurrence in HCC patients with HCV infection background, though the mechanism is not fully understood. Acyclic retinoic acid was shown to decrease multi-centric recurrence after operation, which was reported by only one group. In conclusion, several adjuvant approaches have been studied for their efficacy on recurrence in HCC patients in randomized control trials; however, multi-centric randomized control trial is still needed for further evaluation on their efficacy and systemic or local toxicities;in addition, new adjuvant treatment should be investigated to provide more effective and tolerable methods for the patients with HCC after operation.

To develop target thrombolytic agents with fibrinolytic activity, antiplatelet aggregation activity and reduced immunogenicity, two staphylokinase variants containing Arg-Gly-Asp (RGD) motif were constructed. Gene expression was induced in E. coli JF1125 and the variants, designated DGR and RL1, were purified with gel filtration and ion-exchange chromatography and the purity was over 95%. The fibrinolytic activity and kinetic constants of the two variants were comparable to those of recombinant wild-type staphylokinase. Both the variants can inhibit the platelet aggregation at a final concentration of 2 μM. The titers of antibodies against variants were much lower than those against recombinant staphylokinase in guinea pigs, which indicated that the immunogenicity of the variants was greatly reduced. These results confirm that it is possible to design and produce a bifunctional protein that possesses fibrinolytic and antiplatelet aggrega- tion activities.

This paper is addressed to develop an approximate method to solve a class of infinite dimensional LQ optimal regulator problems over infinite time horizon. Our algorithm is based on a construction of approximate solutions which solve some finite dimensional LQ optimal regulator problems over finite time horizon, and it is shown that these approximate solutions converge strongly to the desired solution in the double limit sense.

The conformation of Kunitz domain 1 of human tissue factor pathway inhibitor-2 (hTFPI-2/KD1) has been studied by Fourier transform infrared spectroscopy, circular dichroism, and Raman spectroscopy. It was found that hTFPI-2/KD1 contained approximately 17% s-helices, 24% β-strands, 46% random coils, 13% β-turns, and two kinds of disulfide bonds (ggg and tgt) at 25℃. The detailed conformational changes of the heated protein observed by Fourier transform infrared spectroscopy, circular dichroism and Raman spectroscopy revealed that hTFPI-2/KD1 was thermally stable. However, KD1 could form an intermediate form at high temperature, then return to its normal conformation when the temperature was lowered. Activity assays also showed that hTFPI-2/KD1 was able to keep its inhibitory activity on plasmin after being heated to 80℃ for 5min.

AIM： TO detect the germline mutations Of hMLH1 and hMSH2 based on mRNA sequencing to identify hereditary non polyposis oolorectal cancer （HNPCC） families. METHODS： Total RNA was extracted from peripberal blood of 14 members from 12 different families fulfilling Amsterdam criteria II. mRNA of hMLH1 and hMSH2 was reversed with special primers and heat-resistant reverse tmnscriptase, cDNA was amplified with expand long template PCR and cDNA sequendng analysis was followed. RESULT： Seven germline mutations were found in 6 families （6/12, 50%）, in 4 hMLH1 and 3 hMSH2 mutations （4/12, 33.3%）; （3/12, 25%）. The mutation types involved 4 missense, 1 silent and 1 frame shift mutations as well as 1 mutation in the non-coding area. Four out of the seven mutations have not been reported previously. The 4 hMLH1 mutations were distributed in exons 8, 12, 16, and 19. The 3 hMSH2 mutations were distributed in exons 1 and 2. Six out of the 7 mutations were pathological, which were dislTibuted in 5 HNPCC families. CONCLUSION： Germline mutations of hMLH1 and hMSH2 can be found based on cDNA sequencing so as to identify HNPCC family, which is highly sensitive and has the advantages of cost and time saving.

The deformation of a compact complex Lagrangian submanifold in a hyper-Kaehler manifold and the moduli space are studied. It is proved that the moduli space Mc1 is a special Kaehler manifold, where special means that there is a real flat torsionfree symplectic connection satisfying dI = 0 (I is a complex structure of Mcl). Thus, following [4], one knows thatT*Mcl is a hyper-Kaehler manifold and then that Mcl is a complex Lagrangian submanifold in T* Mcl.

Aim:To investigate the effects of deferiprone on doxorubicin-inducedcardiotoxicity and determine its protection on cardiac contractility in vivo at tis-sue ***:Spontaneously-beating isolated atria from rats were pre-treated with deferiprone for 10 min at 1.2 mmol/L or 0.3 mmol/L,respectively beforeco-incubation with doxorubicin(DOX)at 0.03 mmol/L for 60 ***(dF/dt)was assessed every 10 min during the *** that,the tissuesaround the sinuatrial nodes were fixed for ultrastructural study;succinate dehy-drogenase(SDH)and Cu,Zn superoxide dismutase(Cu,Zn-SOD)activity,as wellas malondialdehyde(MDA)level of the atria were ***:Treatmentwith DOX alone resulted in a 49.34% reduction of the contractility,mitochondriaswelling,disruption of mitochondrial crista and decreased electron density of ***,with the presence of deferiprone,the negative inotropiceffect and lesions in the cardiac mitochondria structure induced by DOX ***,Zn-SOD activity increased by 12.97%-12.11%,the MDA leveldecreased by 29.12%-39.82% and succinate dehydrogenase(SDH)activity wasameliorated by 25.15%-34.76%.Conclusion:Deferiprone can efficiently pre-serve cardiac ***,the results of this study indicate thatdeferiprone is able to protect mitochondrial function and structure form damageinduced by *** cardiac protective potential of deferiprone could be due toits defense capability against oxidative damage.
