AIM:Pharmacokinetic and pharmacodynamic profiles of Xuesaitong injection in cerebral ischemic patients are in- vestigated in order to optimize dosage regimen of muhiple constituent TCM in clinic. METHODS:63 patients were studied in our protocol. Enumeration and measurement data of all patients were collected during our *** samples were col- lected for two times at different time after intravenous infu- sion Xuesaitong injection at the dosage of 800 mg/kg for two *** of Xuesaitong injection in plas- ma,prothrombin time(PT),thrombin time(TT),activat- ed partial thromboplastin time(APTT)and fibrinogen(Fib) were *** and SAAM Ⅱ were employed for the simulation of PPK parameters and time-course fit- ting of Xuesaitong injection in cerebral ischemic patients. RESULTS:Population pharmacokinetic(PPK)parame- ters of Xuesaitong injection obtained as follow:CL:1.5 L·h(-1)·kg(-1),CL:13.0 L·h(-1)·kg(-1),k:0.1136/ h,k:0.9848 /h,k:0.2057 /***-individual variability were 36.3 %,17.8 %,1.0% and 60.3 %. Residual variability was 8.67%.Age,stature,gender of patients showed no correlation with the prolong ratio of PT (PT%),TT(TT%),APTT(APTT%)and inhibit ratio of Fib(Fib%)whereas dosage(weight),infusion ratio anti concentration of Xuesaitong injection in plasma were marked correlation to PT%,TT%,APTT% and Fib%. Correlation coefficient between dosage,infusion ratio, plasma concentration to PT%,TT%,APTT% and Fib% were 0.598,0.551,0.590,0.505;0.204,0.401, 0.348,0.403 and 0.560,0.436,0.593,O.351,re- spectively.P value was less than 0.01 or *** pharmacological effect obtained when Xuesaitong injection was administered as follow:Dosage was of 12.0-17.0 mg/kg,infusion ratio was of 10.0-18.0 mg/min and plasma concentration was of 25.0-35.0 μg/*** course of Xuesaitong injection in cerebral ischemic patients was successfully predicted and simulated results suggested that better effects were obtained at the dosage of 800- 1200 mg/d than 200-40

clinical Analysis on IVF-ET Treatment of 9 Cases of Post-cesarean Section Uterine Diverticulum

Objective To evaluate the influence of uterine diverticulum patients who have a history of cesarean section on IVF-ET process and pregnancy outcome. Methods Nine patients with uterine diverticulum after cesarean were retrospectively analyzed, who have received IVF-ET treatment. clinical pregnancy rate and embryo implantation rate were measured. Results There were 9 infertility patients in all, 7 cases with tubal factor, 2 with unexplained factor; 3 cases were associated with prolonged menstruation period, including 1 patient was misdiagnosed as dysfunctioned uterine bleeding. There were a total of 16 transplantation cycles, including14 fresh cycles and 2 thawing cycles. Each cycle had at least one high-quality embryo available for transfer. Five cases were difficult to transfer. Two were clinical pregnancie, the implantation rate was 5.13% （2/39）. Conclusion In this study, 14 fresh cycles all had high-quality embryo transfer, uterine diverticulum had no effect on the development of ovums and the formation of high- quality embryos. But forming uterine diverticulum after cesarean section may lead to secondary infertility or patients with prolonged menstruation period, it also may lead it difficult to transfer during the treatment of IVF-ET and affect embryo implantation. So the patients with a history of cesarean section shall receive ultrasonic examination or hysteroscopy routinely before IVF treatment. If necessary surgical treatment is required.

A Study of Factors Related to the Incidence of Cataract in Patients with Non-Insulin Dependent Diabetes Mellitus

Purpose: To investigate the factors related to the development of cataract in patientswith non-insulin dependent diabetes mellitus(NIDDM).Methods: 792 NIDDM patients received ophthalmologic examinations including visualacuity, external status of the eyes, slit lamp microscopy and ophthalmoscopy. Glucose,urea nitrogen (BUN), creatinine (Cr), urine acid (UA), N-acetyl-β2-D-glucosaminidase(NAG), β2-microglobulin(β2-MG) and serum albumin in blood were quantitativelytested. Glucose, pH value, protein, cells, cast and ketobodies in urine were *** of cataract was based on lens opacities classification system Ⅱ. Any patientmeeting "NⅡ", "CⅡ" or "PⅡ" level was diagnosed as ***: The incidence of cataract in this group of NIDDM was 62.37 % (494/792),which significantly related to the duration of the disease course, but not to the sex of thepatient. The occurrence rate of cataract in patients suffering from NIDDM of less thanfive years duration, from five to ten years, and more than ten years was 49.67 % (228/459), 71.84 % (125/174), and 88.68 % (141/159), respectively. The occurrence ofcataract in patients diagnosed of the disease from five to ten years and more than tenyears was much higher than that of those with the course of the disease less than fiveyears( P ＜ 0.05 and P ＜ 0. 001, respectively) . Rising concentrations of blood ureanitrogen, creatinine, glycosylated hemoglobin HbA1c(G-HbA1c), N-acetyl-β2-D-glucosaminidase(NAG) and β2-microglobulin(β2-MG) indicated malfunction of thekidneys, and the rate of cataract occurrence in these patients was ***: This study indicates that prolongation of the duration of non-insulindependent diabetes mellitus, renal dysfunction, as well as poor blood glucose control,may accelerate the development of cataract.

The shear strength parameters property of rock-soil aggregates in embankment slope of reservoir,that is,the relationship between cohesion and gravel content,between friction angle and gravel content,and the relationship between cohesion and water content,between friction angle and water content,is studied based on the direct shear test results,the shear strength change law of the rock-soil aggregates is given,and the unsaturated shear strength formulation of rock-soil aggregates that could consider suction and saturation degree influence is put forward in this paper,through which the sliding or failure physical mechanism of this type of slope under the condition of rainfall infiltration is studied. Also the 3D unsteady saturated-unsaturated seepage field and its FEM resolving mode are established based on the analysis of the slope rainfall infiltration process. Case study with this method indicates that the minimum safety factor of the accumulated rock-soil aggregates dose not arrive at the moment of rainfall cessation,but appears several hours after the rainfall cessation,this phenomenon is in accordance with the practical slope engineering's failure process and could explain appropriately the slope failure caused by rainfall infiltration. Research results in this paper have an important reference value for the research on stability of the accumulated rock-soil aggregates in embankment slope of reservoir,and can enrich the stability analysis method and relevant theory of reservoir embankment slope.

ANALYSIS OF WATER QUALITY IN SHALLOW LAKES WITH A TWO-DIMENSIONAL FLOW-SEDIMENT MODEL

The governing equation for sediment pollutions was derived based on the turbulent diffusion of pollutants in shallow lakes. Coupled with shallow water equations, a depth-averaged 2-D flow and water quality model was developed. By means of the conservation law, a proposed differential equation for the change of sediment pollutants was linked to the 2-D equations. Under the framework of the finite volume method, the Osher approximate Riemann solver was employed to solve the equations. An analytical resolution was used to examine the model capabilities. Simulated results matched the exact solutions especially well. As an example, the simulation of CODMn in the Wuli Lake, a part of the Taihu lake, was conducted, which led to reasonable results. This study provides a new approach and a practical tool for the simulation of flow and water quality in shallow lakes.

HBsAg loading on dendritic cells in patients with chronic hepatitis B: expressions of phenotypic molecules

BACKGROUND: The antigen reducing ability of dentritic cells (DCs), a kind of antigen presenting cells (APCs) initiating immune response, is associated with the specific immune tolerance of chronic hepatitis B(CHB) patients. However, the dysfunction of DCs can be possibly reversed by the stimulation of antigen peptides. In this study, DCs were cultured from peripheral blood monocytes (PBMCs) in patients with CHB in vitro, and the expression of phenotypic molecules on DCs loaded by different concentrations of HBsAg was observed. METHODS: Forty patients with CHB were divided randomly into 4 groups(10 patients in each group). PBMCs were isolated, and DCs were cultured after addition of granulocyte/macrophage colony-stimulating factor (GMCSF) and interleukin 4(IL-4). On the 9th day, DCs of the experimental groups were loaded at HBsAg concentrations of 2.5mg/L, 5mg/L and 10mg/L for 24 hours, whereas those of the control group were not loaded. An electron microscope was used to analyze the morphological changes of the DCs. The expression of phenotypic molecules on DCs in different groups was detected with flow cytometry. RESULTS: A combination of GM-CSF and IL-4 produced DCs from PBMCs in patients with CHB after being cultured for 9 days, whose morphological changes were tested by an electron microscope. The expression of phenotypic molecules on DCs in the control group was as low as CD83 (8.02±3.99)%, CD80(8.77±2.06)%, and MHC-DR (14.05±2.66)%. Loaded by different concentrations of HBsAg, the up-regulation of phenotypic molecules on DCs was found, with CD83(18.35±2.93)%, CD80(42.63±7.15)% and MHC-DR(47.49±6.59)% in 2.5mg/L HBsAg loading group, CD83(17.88±3.12)%, CD80(45.24± 10.93)% and MHC-DR(47.07±8.52)% in 5mg/L HBsAg loading group and CD83(16.74±2.86)%, CD80(44.59±6.99)% and MHC-DR(48.59±7.42)% in 10mg/L HBsAg loading group, respectively. The study site is located in Qianyanzhou experimental station and the surrounding area. Based on CBERS-02 satellite data and field measurement, we not only discussed the relationship between NDVI and biomass of two species of coniferous plantations, namely, Pinus massoniana Lamb and Pinus elliottii Engelm, but also introduced the biomass models based on NDVI. The comparison between measured biomass in Qianyanzhou and biomass derived from CBERS-02 CCD data showed that it is feasible to estimate biomass based on NDVI. But its limitations cannot be ignored. This kind of model depends on the dominant vegetation species. There are some effect factors in estimating biomass based on NDVI. This paper analyzes these factors based on fine-resolution CBERS-02 CCD data and some conclusions are drawn: In Qianyanzhou, the area with good vegetation coverage, the nonlinearity of NDVI has little influence on scaling-up of NDVI. As a result of surface heterogeneity, scaling-up can cause NDVI within each pixel to change. Because scaling-up can cause pixel attribute to change, the applicability of biomass model is one of the sources of error in estimating biomass.

Anticancer effect of two diterpenoid compounds isolated from Annona glabra Linn

AIM: To study the inhibitory effect of two diterpenoid compounds isolated from Annona glabra Linn (Cunabic acid and ent-kauran-19-al-17-oic acid) on the proliferation of Human Liver Cancer (HLC) cell line SMMC-7721 and its mechanism. METHODS: Inhibition of cell proliferation was measured by MTT assay. The morphological changes of SMMC-7721 cells were observed under inverted phase-contrast microscope, fluorescent microscope, transmission electron microscope (TEM), and scanning electron microscope (SEM). Flow cytometer (FCM) was used to calculate the cell apoptotic rate, and immunohistochemical staining was used to observe the regulation of gene expression. RESULTS: The proliferation of SMMC-7721 cells was obviously inhibited after being treated with Cunabic acid at the concentration >5 μmol/L and ent-kauran-19-al-17-oic acid >10 μmol/L. The biggest inhibitory effect was 81.05 % when treated with Cunabic acid at the concentration of 25 μmol/L. The effect had a linear relationship with concentration. The result indicated that drug-treated cells exhibit typical morphological changes of apoptosis, including condensed chromatin and a reduction in volume. Sub-G0/G1 peak was found by FCM analysis and the cell cycle was arrested at G0/G1 stage. The apoptotic rates of the cells treated by Cunabic acid and ent-kauran-19-al-17-oic acid were 43.31 % and 24.95 %, respectively. It was visualized by immunohis- tochemical staining that the drugs down-regulated the gene expression of bcl-2 gene and up-regulated that of bax gene. CONCLUSION: The two diterpenoid compounds isolated from Annona glabra Linn, Cunabic acid and ent- kauran-19-al-17-oic acid can obviously inhibit the proliferation of HLC cell line SMMC-7721. The mechanism is correlated with the induction of cell apoptosis by down-regulating the gene expression of bcl-2 gene and up- regulating that of bax gene.

Small-Scale Spatial Variability of Soil Nutrients and Vegetation Properties in Semi-Arid Northern china

A field experiment was conducted at Kezuohouqi County, Inner Mongolia Autonomous Region of china, which was located on the southeastern edge of the Horqin Sandy Land, to study the spatial variability of soil nutrients for a smallscale, nutrient-poor, sandy site in a semi-arid region of northern china; to investigate whether or not there were 'islands of fertility' at the experimental site; and to determine the key nutrient elements that sustained ecosystem stability. Results obtained from geostatistical analysis indicated that the spatial distribution pattern of soil total nitrogen (STN) was far different from those of soil organic matter (SOM), total phosphorus (STP), and total potassium (STK). Compared to SOM, STP, and STK, STN had a lower structural heterogeneity ratio and a longer range, while other elements were all similar. In addition, STN had an isotropic spatial structure, whereas the others had an anisotropic spatial structure. The spatial structure patterns of herbage species, cover,and height also differed, indicating that spatial variability was subjected to different ecological factors. Differences in the spatial variability patterns among soil nutrients and vegetation properties showed that soil nutrients for a small-scale were not the primary limiting factors that influenced herbage spatial distribution patterns. Incorporating spatial distribution patterns of tree species, namely, Pinus sylvestris var. mongolica Litv. and shrub Lespedeza bicolor Turcz. in a research plot and using fractal dimension,SOM, STP, and STK were shown to contribute to the 'islands of fertility' phenomenon, however STN was not, possibly meaning that nitrogen was a key limiting element. Therefore, during restoration of similar ecosystems more attention should be given to soil nitrogen.
