To observe the effects of simulated microgravity on beta islet cell culture, we have compared the survival rates and the insulin levels of the isolated rat islet cells cultured at micro- and normal gravity conditions. The survival rates of the cells cultured were determined by acridine orange-propidium iodide double-staining on day 3, 7 and 14. The morphology of the cells was observed by electron microscopy. Insulin levels were measured by radio immuno assays. Our results show that the cell number cultured under the microgravity condition is significantly higher than that under the routine condition (P<0.01). Some tubular structure shown by transmission electron microscopy, possibly for the transport of nutrients, were formed intercellularly in the microgravity cultured group on day 7. There were also abundant secretion particles and mitochondria in the cytoplasm of the cells. AIM:To determine whether mitochondrial dysfunction resulting from high-fat diet is related to impairment of the phosphatidylinositol 3-kinase (PI3K)/protein kinase B (Akt,also known as PKB) pathway. METHODS:Rat models of nonalcoholic fatty liver were established by high-fat diet feeding. The expression of total and phosphorylated P13K and Akt proteins in hepatocytes was determined by Western blotting. Degree of fat accumulation in liver was measured by hepatic triglyceride. Mitochondrial number and size were determined using quantitative morphometric analysis under transmission electron microscope. The permeability of the outer mitochondrial membrane was assessed by determining the potential gradient across this ***:After Wistar rats were fed with high-fat diet for 16 wk,their hepatocytes displayed an accumulation of fat (103.1 ± 12.6 vs 421.5 ± 19.7,P 3% vs 83.0% ± 10.9%,P 389.385% ± 18.612% vs 249.121% ± 13.526%,P 3K and Akt proteins in hepatocytes was reduced,as was the expression of the anti-apoptotic protein Bcl-2,while expression of the pro-apoptotic protein caspase-3 was increased. When animals were treated with pharmacological inhibitors of P13K or Akt,instead of high-fat diet,a similar pattern of hepatocellular fat accumulation,mitochondrial impairment,and change in the levels of PI3K,Akt,Bcl-2 was observed. CONCLUSION:High-fat diet appears to inhibit the PI3K/Akt signaling pathway,which may lead to hepa-tocellular injury through activation of the mitochondrial membrane pathway of apoptosis.

The liver is the commonest site of distant metastasis ofcolorectal cancer and nearly half of the patients withcolorectal cancer ultimately develop liver involved duringthe course of their diseases. Surgery is the only therapythat offers the possibility of cure for patients with hepaticmetastatic diseases. Five-year survival rates after resectionof all detectable liver metastases can be up to 40 %.Unfortunately, only 25 % of patients with colorectal livermetastases are candidates for liver resection, while the othersare not amenable to surgical resection. Regional therapiessuch as radiofrequency ablation and cryotherapy may beoffered to patients with isolated unresectable metastasesbut no extrahepatic diseases. Hepatic artery catheterchemotherapy and chemoembolization and portal veinembolization are often used for the patients with extensiveliver metastases but without extrahepatic diseases, whichare not suitable for regional ablation. For the patients withmetastatic colorectal cancer beyond the liver, systemicchemotherapy is a more appropriate choice. Immunotherapyis also a good option when other therapies are used incombination to enhance the efficacy. Selective internalradiation therapy is a new radiation method which can beused in patients given other routine therapies Without effects.

Primary and secondary malignant liver cancer are some of most common malignant tumors in the world. Chemotherapy and radiotherapy are not very effective against them. Surgical resection has been considered the only potentially curtive option, but the majority of patients are not candidates for resection because of tumor size, location near major intrahepatic blood vessels and bile ducts, precluding a margin-negative resection, cirrhotic, hepatitis virus infection or multifocial. Radiofrequence ablation (RFA), which is a new evolving effective and minimally invasive technique, can produce coagulative necrosis of malignant tumors. RFA should be used percutaneously, laparascopically, or during the open laparotomy under the guidance of ultrasound, CT scan and MRI. RFA has lots of advantages superior to other local therapies including lower complications, reduced costs and hospital stays, and the possibility of repeated treatment. In general, RFA is a safe, effective treatment for unresectable malignant liver tumors less than 7.0 cm in diameter. We review the principle, mechanism, procedures and experience with RFA for treating malignant liver tumors.

Aim:To investigate the robust gene signature in liver cancer,we applied anintegrated approach to perform a joint analysis of a highly diverse collection ofliver cancer genome-wide datasets,including genomic alterations and transcrip-tion ***:1-class Significance Analysis of Microarrays coupled withranking score method were used to identify the robust gene signature in livertumor ***:In total,1 625 051 gene expression measurements from 16public microarrays,2 pairs of serial analyses of gene expression experiments,and252 loss of heterozygosity reports obtained from 568 publications were used inthis integrated *** resulting robust gene signatures included 90 genes,which may be of great importance to liver cancer research.A system assessmentanalysis revealed that our integrative method had an accuracy of 92% and acorrelation coefficient value of ***:The system assessment resultsindicated that our method had the ability of integrating the datasets from varioustypes of sources,and eliciting more accurate results,as can be very useful in thestudy of liver cancer.

BACKGROUND: Interleukin-18 (IL-18), a pro-inflamma- tory cytokine that induces interferon-γ (IFN-γ) production in T cells and natural killer cells, plays a critical role in the T-lymphocyte helper type 1 ( Th1) response. This study was designed to explore the effect of IL-18 on peripheral blood mononuclear cells ( PBMCs) derived from chronic hepatitis B (CHB) and on hepatitis B virus (HBV) DNA released by HepG2.2.15 cell lines, which were transfected with hepatitis B virus gene in vitro. METHODS: PBMCs isolated from 25 healthy people and 25 patients with CHB were stimulated with HBcAg and IL-18 of various concentrations for 72 hours. The levels of IFN-γ in the supernatants of cultured PBMCs were determined by ELISA. After the stimulation of IL-18 of various concentra- tions, PBMCs derived from one patient were co-cultured for 96 hours with HepG2. 2. 15 cells which had been cul- tured for 24 hours, and then the supernatants were collected by centrifugation and used for HBV DNA quantitative as- say. RESULTS: When PBMCs were stimulated by HBcAg and IL-18 at various concentrations, the levels of IFN-γ in the supernatants of CHB groups were much higher than those in normal control groups, at 0.2 ng/ml: t =11.70, P0.05). The content of HBV DNA in the superna- tant of co-cultivation of HepG2. 2. 15 cells and PBMCs without stimulated materials was higher than that stimula-ted by HBcAg and EL-18 at various concentrations of HBc- Ag and IL-18 together with IL-12/IFN-α1lb.
