为进一步规范影像学技术在我国大动脉炎(Takayasu arteritis,TAK)患者临床诊治中的应用,由复旦大学附属中山医院、中华医学会风湿病专委会、中国初级卫生保健基金会及复旦大学循证医学中心联合发起并撰写了《大动脉炎影像学诊治指南计划书》。工作组将基于现有的大动脉炎影像学文献证据,参照《世界卫生组织指南制订手册》,规范开展指南制订工作。本计划书主要介绍指南制订的意义、指南制订组组建、临床问题的遴选、证据检索与评价、指南形成和发布等过程。

CD134, a member of the tumor necrosis factor receptor superfamily, plays a crucial role in T cell survival. In this study, peripheral blood mononuclear cells （PBMCs） stimulated with phytohemagglutinin （PHA, 50 μg/ml） were treated by CD134 mAb （1μg/ml, 5 μg/ml, 10 μg/ml） for 6 h, 12 h, 24 h and 48 h. The level of perforin mRNA was measured by reverse transcription-polymerase chain reaction （RT-PCR） technique. Our data showed that the expression of perforin mRNA in PBMCs was down-regulated by CD134 mAb in a dose-dependent manner in range of 1 μg/ml to 5 μg/ml and dropped down to its minimum on 24 h （p 〈 0.05）. The level of perforin mRNA reached a plateau when the concentration of CD134 mAb exceeded 5 μg/ml. In conclusion, CD134 mAb can inhibit perforin expression, which may enhance the ability of T cells for survival. Cellular ＆ Molecular Immunology. 2005;2（6）： 467-471.

The immune system has evolved a variety of regulatory mechanisms to ensure the peripheral self-tolerance as well as the optimal capacity to elicit effective anti-infection immunity. At present, there is no satisfactory conceptual framework to explain how the peripheral immunity is regulated at a biological system level, which enables the immune system to perform its essential functions to mount effective immunity to virtually any foreign antigens but avoid harmful immune responses to self. In this regard, during the past few years, an “affinity/avidity model of peripheral T cell regulation” has been proposed and tested, which opens up a new paradigm to understand how the peripheral immunity, to both self and foreign antigens, is regulated. The paradigm is based on the discovery of a subset CD8^＋ T cells with TCRs which specifically recognize a unique set of self-peptides presented by the MHC class Ib molecule Qa-I differentially expressed on T cells as a function of the affinity/avidity of T cell activation. These Qa-1 restricted CD8^＋ T cells represent an example of how the immune system utilizes a unified mechanism to regulate adaptive immunity to both self and foreign antigens. Thus, by selectively down-regulating T cells of intermediate affinity/avidity, to any antigens, the immune system controls the adaptive immunity without the necessity to distinguish self from non-self in the periphery at the level of T cell regulation. Aim:To determine the effects of acetamide-45 on respiratory function,airwayinflammation,and the activity of phosphodiesterase 4（PDE4）in allergic ***:Rats were sensitized by a single intramuscular injection with ovalbumin（OVA）and were challenged with ovalbumin applied by using an aerosol repeat-edly for 7 d after 2 ***-45 at concentrations of 5,10,or 30 mg/kg wasthen administered by intraperitoneal *** in dynamic lung compli-ance and lung resistance,the accumulation of inflammatory cells in bronchoalveolarlavage,PDE4 activity,and the concentration of interleukin-4 in rat lung tissuewere ***:Seven days of treatment with acetamide-45 preventedeosinophil accumulation in allergic *** doses of 5,10,and 30 mg/kg,aceta-mide-45 decreased lung resistance to 0.20±0.04,0.25±0.07,and 0.22±0.05cmH2O·s-1·mL-1,respectively（P39±0.06,and 0.42±0.09 mL/cmH20（P3±57,and 238±36 nmol·g-1·min-1（P3,5.95±1.20,and 5.68±2.20 μtg/g protein（Pinflammation in thisanimal model,and the PDE4 activity of lung tissue was obviously ***-45 was an effective anti-inflammatory agent in respiratory inflammation,and the mechanism of its action might depend on inhibition of PDE4.

Aim:To explore the modulatory effect of bradykinin （BK） on 5-HT3receptor-mediated current in trigeminal ganglion （TG） neurons in ***:The whole-cell patch-clamp technique was used to record 5-HT-activated currents (I5-HT) inneurons freshly dissociated from rat *** were applied by rapid ***:The majority of the neurons examined responded to 5-HTapplied externally with an inward current （76.3%,74/97） that could be blocked bythe 5-HT3receptor antagonist,ICS-205,930 (1×10-6mol/L).In 66 of the 74 cellssensitive to 5-HT （89.2%）,pretreatment for 30 s with BK (1×10-6-1×10-10mol/L)could potentiate I5-HTwith the maximal modulatory effect occurring at10-7mol/L BK （71.6%+4.9%）.BK shifted the 5-HT concentration-response curveupwards with an increase of 68.9%±7.2% in the maximal current response,but withno significant change in the EC50value （19.1±3.2 μmol/L vs 20.9±3.5 μmol/L;t-test,P>0.05;n=8）.BK potentiated I5-HTin a holding potential-independent manner anddid not alter the reverse potential of *** BK-induced potentiation ofI5-HTwas almost completely blocked by Hoe 140 (5×10-7mol/L),a selective B2BKreceptor antagonist,and was removed after intracellular dialysis of GF-109203X（2 μmol/L）,a selective protein kinase C （PKC） inhibitor,with the re-patch ***:Pre-application of BK exerts an enhancing effect on I5-HTvia a PKC-dependent pathway in rat TG neurons,which may explain the peripheral'mecha-nism of pain and hyperalgesia caused by,for example,tissue damage andinflammation.

Aim:To explore whether local blockade of T-box expressed in T cells (T-bet)expression in the lungs could lead to airway ***:Twenty-fourrats were randomly divided into 4 groups:saline group,ovalbumin (OVA)-sensi-tized group,nonsense group,and the antisense *** OVA-sensitized ratswere sensitized and challenged with OVA,and the rats in the nonsense andantisense groups were subjected to an aerosol delivery of the nonsense andantisense oligonucleotides (AS-ODN) of T-bet (0.1%,w/v).The levels of inter-feron-γ(IFN-γ),interleukin-4(IL-4),and IL-5 in the bronchoalveolar lavage fluid(BALF) were detected by ELISA,and the mRNA and the protein expression ofT-bet and GATA-3 genes were examined by in situ hybridization and Western blotanalysis,***:The expression of T-bet mRNA and protein in thelungs of the rats in the antisense group were inhibited *** lungs ofthe rats in the antisense and OVA-sensitized groups showed eosinophil and lym-phocyte inflammatory infiltration,and eosinophilia located predominantly aroundthe *** number of GATA-3 mRNA-positive cells and the level of GATA-3protein in the lungs of the rats in the antisense and the OVA-sensitized groupssignificantly *** level of IL-4 and IL-5 in the BALF in the antisenseand OVA-sensitized groups were elevated,but the level of IFN-γ ***:Antisense ODN-induced local blockade of T-bet expres-sion leads to airway inflammation with a selective alteration in patterns of cytokineexpression and recruitment of eosinophil cells similar to that in the OVA-sensitizedanimals.

The tissue destruction characteristic of syphilis infection may be caused by inflammation due to Treponema pallidum and the ensuing immune responses to the *** membrane proteins are thought to be potent inducers of inflammation during the early stages of ***,the actual membrane proteins that induce inflammatory cytokine production are not known,nor are the molecular mechanisms responsible for triggering and sustaining the inflammatory *** the present study,Tp0751 recombinant protein from *** was found to induce the production of proinflammatory cytokines,including TNF-α,IL-1βand IL-6,in a THP-1 human monocyte cell *** signal transduction pathways involved in the production of these cytokines were then further *** inhibition of TNF-a,IL-1β,or IL-6 production was observed following treatment with the SAPK/JNK specific inhibitor SP600125 or with an ERK inhibitor *** contrast,anti-TLR2 mAb,anti-CD14 mAb,and the p38 inhibitor SB203580 significantly inhibited the production of all three *** addition,pyrrolidine dithiocarbamate (PDTC),a specific inhibitor of NF-κB,profoundly inhibited the production of these ***0751 treatment strongly activated NF-κB,as revealed by Western ***,NF-κB translocation was significantly inhibited by treatment with *** results indicated that TLR2,CD14,MAPKs/p38,and NF-κB might be implicated in the inflammatory reaction caused by *** infection.
