Relation of phospholipase A2-Ⅴ and indoxam to hippocampal neuronal death

BACKGROUND: Ⅴ secretory phospholipase A2 (sPLA2-Ⅴ) is abundant in many mammal tissues. However, it remains unknown whether sPLA2-Ⅴ causes biological or pathological response in central nervous system. OBJECTIVE: To observe the effect of phospholipase A2-Ⅴ (PLA2-Ⅴ) and its inhibitor (indoxam) on hippocampal neuron survival. DEsIGN: A repetitive measurement. sETTING: The Animal Center of south Carolina University. MATERIALs: sprague-Dawley pregnancy day-7, 14, 21 female rats were selected; Reagents: sPLA2- Ⅴ and indoxam were obtained from the Dennis Research Laboratories METHODs: The experiment was finished at the animal center in south Carolina University from January to December, 2004. 0, 12.5, 25, 50 and 100 μg/L sPLA2-Ⅴ were added to neuron with none-MgCl2 Eagle’s medium at 37 ℃, then changed to normal neuron culture medium after 3 hours. 1, 2.5, 5 and 10 μmol/L indoxam was added at 6 hours after 100 μg/L sPLA2-Ⅴwas put to Day-21 sD rat hippocampal embryonic neurons with none-MgCl2 Eagle’s medium at 37 ℃. After 3 hours in the inhibition experiment, it was changed to normal neuron culture medium. The embryonic hippocampal neurons were primarily cultured, and the neuron survival ratio was detected with morphological method. MAIN OUTCOME MEAsUREs: survival ratio of hippocampal neurons. REsULTs: ① Effects of sPLA2-Ⅴon neuron survival: When sPLA2-Ⅴ was 0, 12.5, 25, 50 and 100 μg/L, the neuron survival ratios in embryonic neurons of day-7 sD rats were (95.3±1.1)%, (81.4±3.1)%, (74.2±2.2)%, (62.4±1.7)% and (48.9±1.6)%, those in embryonic neurons of day-14 rats were (93.2±1.4)%, (74.3±1.9)%, (68.1±1.7)%, (56.1±1.4)% and (42.5±1.1)%, and those in embryonic neurons of day-21 rats were (91.2±1.2)%, (69.4±2.1)%, (60.3±2.2)%, (49.1±1.2)% and (35.5±1.9)%. Congenital motor nystagmus(CMN)is a relatively common oculomotor disorder characterized by bilateral uncontrollable ocular ***,the FRMD7 gene mutation has been identified as the genetic cause of *** clinical data and the genomic DNA of three Chinese CMN families were collected afcer the consent *** by two-point linkage analysis combined with haplotype analysis was performed and mutation screening was conducted in the FRMD7 gene by direct sequencing.A maximum two-point Lod score of 4.27 at θ=0.00 was obtained with marker DXs8041 in close proximity to the FRMD7 gene on chromosome *** screening in FRMD7 identified two novel missense mutations(781C>G,886G>C)and one reported nonsense mutation(1003C>T).These nucleotide alterations were neither seen in unaffected members of the families nor in 100 unrelated control *** study widens the mutation spectrum of the FRMD7 gene and it is the first report of the FRMD7 gene mutations in Chinese population.

High-and Low-Frequency Probe Tone Tympanometry in Chinese Infants

Objective To assess the utility of low- and high-frequency tympanometry in the diagnosis of middle ear dysfunction in Chinese infants. Methods Tympanograms were obtained with 226 Hz, 678 Hz and 1000 Hz probe tones from infants aged 5-25 weeks with normal auditory brainstem responses(ABRs)(15 infants, 30 ears) and with prolonged wave I latencies(17 infants, 20 ears), suggesting middle ear dysfunction, using a GsI Tympstar middle ear analyzer Version II. Results The single-peaked tympanogram was the most characteristic type in both groups and seen in 25 ears (83.3%) in the normal ABR group and in 18 ears (90%) in the delayed wave I group, respectively. The peak pressure, peak compensated static acoustic admittance and gradient of 226 Hz tympanometry were of no significant differences between the two groups. The 678 Hz tympanograms of admittance, susceptance and conductance demonstrated non-peak, single-, double- and tri-peaked patterns in both groups. The agreement between ABRs and 678 Hz tympanograms of admittance,susceptance and conductance were 70.0%, 58.0% and 64.0%(kappa=0.324, 0.234 and 0.118) respectively. For 1000 Hz probe tone, admittance, susceptance and conductance tympanograms showed single peaked patterns in 28(93.3%), 25 (83.3%) and 26(86.7%) of the 30 normal ears. Admittance, susceptance and conductance tympanograms using the 1000 Hz probe tone were flat in 15 (75%), 17(85%) and 13 (65%) of the ears in infants with prolonged wave I latencies. For 1000 Hz admittance, susceptance and conductance Tympanograms, the agreement between tympanometry and ABR results were 90.0%, 92.0% and 86.0% with kappa at 0.783, 0.831 and 0.690, respectively. Conclusion 1000 Hz probe tone tympanometry is a promising middle ear function test for infants of 1-6 months age, while 226 Hz and 678 Hz probe tones are less efficient in detecting middle ear dysfunction in infants.

Pentastomid parasite Armillifer moniliformis, (A. moniliformis) is a pathogen causing human pentastomiasis. Pentastomiasis is a parasitic zoonosis, which has seldom been reported. When the number of armillifer parasites infecting a person is few or very few, the overwhelming majority of the cases are of asymptomatic or subclinical presentation and are generally discovered during autopsies or at surgical operation. A case of A. moniliformis, with heavy infection caused by nymphal pentastomids and possessing the clinical symptoms of chronic fever and abdominal pain, was admitted to the children’s hospital in Hangzhou, china. As the first case in china, it is hereby reported.
